Elevated pro-inflammatory and lipotoxic mucosal lipids characterise irritable bowel syndrome

AiM： To investigate the pathophysiology of irritable bowel syndrome （iBS） by compa.ing the global mucosal metabolic profiles of iBS pa.ients with those of healthy controls. METHODS： Fifteen iBS pa.ients fulfilling the Rome ii criteria, and nine healthy volunteers were included in the study. A combined lipidomics （UPLC/MS） and metabolomics （GC × GC-TOF） approach was used to achieve global metabolic profiles of mucosal biopsies from the ascending colon. RESULTS： Overall, lipid levels were elevated in pa.ients with iBS. The most significant upregula.ion was seen for pro-inflammatory lysophospha.idylcholines. Other lipid groups that were significantly upregulated in iBS pa.ients were lipotoxic cera.ides, glycosphingolipids, and di-and triacylglycerols. Among the meo tabolites, the cyclic ester 2（3H）-furanone was almost 14-fold upregulated in iBS pa.ients compared to healthy subjects （P = 0.03）. CONCLUSiON： iBS mucosa.is characterised by a distinct pro-inflammatory and lipotoxic metabolic profile. Especially, there was an increase in several lipid species such as lysophospholipids and cera.ides.

Unusual carbonaceous matter, termed here chiemite, composed of more than 90% C from the Alpine Foreland at Lake Chiemsee in Bava.ia, southeastern Germany has been investigated using optical and atomic force microscopy, X-ray fluorescence spectroscopy, scanning and transmission electron microscopy, high-resolution Raman spectroscopy, X-ray diffraction and differential thermal analysis, as well as by δ13 C and 14 C ra.iocarbon isotopic data analysis. in the pumice-like fragments, poorly ordered carbon matter co-exists with high-ordering monocrystalline α-carbyne, and contains submicrometersized inclusions of complex composition. Diamond and carbyne add to the peculiar mix of matter. The required very high temperatures and pressures for carbyne forma.ion point to a shock event probably from the recently proposed Holocene Chiemgau meteorite impact. The carbon material is suggested to have largely formed from hea.ily shocked coal, vegeta.ion like wood, and peat from the impact target area. The carboniza.ion/coa.ifica.ion high PT process may be attributed to a strong shock that instantaneously caused the complete evapora.ion and loss of vola.ile matter and water, which nevertheless preserved the original cellular structure seen fossilized in many fragments. Rela.ively fresh wood encapsulated in the purported strongly shocked matter point to quenched carbon melt components possibly important for the discussion of survival of orga.ic matter in meteorite impacts, implying an astrobiological rela.ionship.

Due to high demand and limited availa.ility of rare earth elements (REEs), Europe is unable to meet its industrial needs, especially High Tech needs, today for the manufacturing sector. Therefore, the EU has included them in the group of 14 critical minerals. China currently controls completely the mining activity, the enrichment technologies and metallurgy, and end-metal products of rare earths, resulting both Europe and the U.S.A. in full industrial dependency. The exploita.ion and wide use of REEs in fertilizers have led to accumula.ion of these elements in soils, resulting in an agriculture field pollution, which affects directly the soil microfauna with a toxic potential effect.

There is no pa.ina of doubt that the central philosophical theories of Karl Popper and Thomas Kuhn concerning the nature, substance and method for acquiring scientific knowledge constitute milestones in 20th century philosophy of science. Just as Popper＇s fundamental work on the subject, The Logic of Scientific Discovery, marked a decisive break with inductivist epistemologies, Kuhn＇s magnum opus, The Structure of Scientific Revolutions （1962, enlarged ed. 1970）, inaugurated the coming of age of the historical turn in the philosophy of science. Some scholars seem to consider the main doctrines of both philosophers as irreconcilables or contra.ictories. This explains why, for example Popper and Popperians such as imre Lakatos and John Watkins describe themselves as ＂critical ra.iona.ists＂, whereas they refer to Kuhn as an ＂irra.iona.ist＂ or ＂rela.ivist＂-appella.ions that the latter has consistently rejected. The debate between Popper and Kuhn, especially as contained in an important work, Criticism and the Growth of Knowledge （1970）, highlights some of the knotty problems connected with philosophical appraisals of science. it also demonstrates the strengths and weaknesses of logistic approaches in the philosophy of science, on the one hand, and of historically informed socio-psychological analysis of science, on the other. in this paper, we reexa.ine the Popper-Kuhn controversy from an experimenta.ist perspective. in other words, we argue that the ideas of testing and normal science can be systema.ically accommodated by fine-structure dissection of empirical research through which scientists learn about the world, based on the assumption that the progress of science is the growth of experimental knowledge-a fact often neglected in theory-dominated philosophies of science. This study combines the analytical model to build a landside monitoring decision support system of the Web GiS. The landslide area of Lishan is a case study for the research. The analysis of the risk degree for the landslide area.in Lishan is based on the three-layer architecture of Fuzzy Analytic Hierarchical Process （FAHP）. There are four fuzzy model structures used in monitoring devices： rainfall, groundwater level, Time Domain Reflectometry （TDR） monitored the subsurface deforma.ion, and Global Positioning System （GPS） monitored ground displacement. These structures are rela.ive to four membership functions that are used to classify four states, including safety, attention, warning, and danger. The risk degree of the landslide area can be obtained through the fuzzy rules by determining management criteria. Calcula.ing the total scores of historical monitoring record of the rainfall, groundwater level, TDR, and GPS through the fuzzy theory can determine the analytical results of risk degrees in Lishan landslide area. in this whole area, management criterion is in the state of attention when the total score is larger than 72, in the state of warning when total score is larger than 95, and in the state of danger when total score is larger than 113. The system provides real-time monitoring data, and prewarning decision support in order to announce and prevent the disaster at the earliest time.

Alcohol-associated liver disease(ALD)is increa.ingly prevalent throughout the world(1).it remains the lea.ing cause of liver-related morbidity and morta.ity worldwide and contributes to approximately 6%of global deaths(2).ALD encompasses a wide range of hepa.ic pathologies,including steatosis,steatohepa.itis,fibrosis,and cirrhosis(3).Given the severe lack of therapeutic interventions available to reverse the progression of ALD,it is currently the lea.ing indica.ion for liver transplanta.ion(4).Advancements within the field are needed to identify diagnostic biomarkers for early detection and to develop therapeutics to ameliorate hepa.ic dysfunction and pathologies associated with ALD.
