Natural Products Modulate the Multifactorial Multidrug Resistance of Cancer

Multidrug resistance (MDR) is a critical problem in cancer chemotherapy. Cancer cells can develop resistance not only to a single cytotoxic drug, but also to entire classes of structurally and functionally unrelated compounds. Several mechanisms can mediate the development of MDR, including increased drug efflux from the cells by ABC-transporters (ABCT), activation of metabolic enzymes, and defective pathways towards apoptosis. Many plant secondary metabolites (SMs) can potentially increase sensitivity of drug-resistant cancer cells to chemotherapeutical agents. The present thesis investigates the modulation of MDR by certain medicinal plants and their active compounds. The inhibition of ABCTs (P-gp/MDR1, MRP1, BCRP) and metabolic enzymes (GST and CYP3A4), and the induction of apoptosis are useful indicators of the efficacy of a potential medicinal drug. The focus of this study was the possible mechanisms of drug resistance including: expression of resistance proteins, activation of metabolic enzymes, and alteration of the apoptosis and how to overcome their resistance effect on cancer cells. The overall goal of this review was to evaluate how commonly used medicinal plants and their main active secondary metabolites modulate multidrug resistance in cancer cells in order to validate their uses as anticancer drugs, introduce new therapeutic options for resistant cancer, and facilitate the development of their anticancer strategies and/or combination therapies. In conclusion, SMs from medicinal plants exhibit multitarget activity against MDR-related proteins, metabolic enzymes, and apoptotic signaling, this may help to overcome resistance towards chemotherapeutic drugs.

Background: Attention-deficit hyperactivity disorder (ADHD) is a widespread and debilitating disorder with relatively high prevalence in Saudi Arabia. Neuropsychological and radiological investigations have revealed that there are some differences in the components of the brain regions in children with and without ADHD. In this study we have performed whole exome sequencing (WES) in four non-familial cases of ADHD from Makkah Region to identify the genetic polymorphisms associated with the disease in our Saudi population. Methods: Exome sequencing was carried out using Ion Proton with AmpliSeq Exome library methods, and the data were analysed by Ion Reporter 5.6 software. Results: A total of 33 variants were identified from 222 genes selected from the GWAS catalogue for ADHD associated genes. However, the SNPs we identified in these genes were not reported to be associated with ADHD in previous studies. We have identified 2 novel missense variants;one in c.3451G > T;p. (Ala1151Ser) in ITGA1 gene and another is c.988G > A;p. (Ala330Thr) in SPATA13 genes. The variants rs928661, rs11150370 and rs386792899 were the only three variants that appeared on all the 4 patients studied. Six missense variants, rs16841277, rs2228209, rs2230283, rs3741883, rs1716 and rs2272606, were found in 3 different patients, respectively. However, the three documented variants are rs13166360 with bipolar disorder, rs920829 with neuropathic pain, and rs6558702 with schizophrenia. Conclusion: We have identified 2 novel variants in ADHD children. SIFT score of all variants indicates that these substitutions have damaging effects on the protein function. Further screening studies are recommended for confirmation.

Study of enteropathogens associated with paediatric gastroenteritis

Objective:To determine the etiology of acute diarrhea in children under 5 years of age and to improve- knowledge of the etiology of gastrointestinal pathogens using traditional and molecular diagnostic ***: Various common enteropathogens(viral,bacterial and parasites) associated with diarrhea were investigated by conventional and molecular techniques(PCR) for verotoxin present in Escherichia coli in 218 children less than 5 years of age admitted to Mansoura University Children hospital- ***:The occurrence of enteropathogens identified was as follows:*** O157:H7 38.8%followed by Salmonella Spp 29.4%,Aeromonas 20%and Shigella Spp 11.8%.Rotavirus was found in of samples 17.1%.Rotavirus was statistically significant in ageella isolates were tested for their susceptibility to common antimicrobial agents and most of the isolates were resistant to ampicillin and trimethoprim /***:This study demonstrated that rotavirus,*** O157:H7,Salmonella Spp,and Aeromonas were significant *** was significantly associated with infantile *** results highlight the value of using a combination of traditional and PCR techniques in the diagnosis of enteropathogens related to acute gastroenteritis in children.

Evaluation of molecular and immunological methods for diagnosis of tuberculosis pleurisy

Objective:The inefficiency of conventional laboratory methods for diagnosis of Pleural tuberculosis(TBP) and the reliance on pleural biopsy have motivated the evaluation of alternative diagnostic *** goal was to evaluate different laboratory techniques Ziehl Neelsen,Mantoux skin test,determination of interferon gamma in serum and pleural fluid,polymerase chain reaction and serological study of specific IgG,IgM and IgA beside bacteriological culture by BACTEC 460 TB for rapid and accurate diagnosis of tuberculosis pleurisy. Methods:Patients presented with pleural effusions were subjected to study by ZN,PCR,serological study By specific IgG,IgM and IgA for A60 of tuberculosis compared to culture by BACTEC460 *** gamma was determined both in serum and pleural ***:Mantoux skin test was positive in 19 TBP patients and four patients with exudative pleural effusion.Z.N staining results of pleural biopsy specimens were positive in only 1 of 23 patients(4.3%) in the tuberculous pleural effusion *** was positive in 20 cases of group 1(87%).Serum and pleural fluid interferon had significantly elevated levels(Pelation in TBP(PelISA test was positive in 7/23(30.4%),IgM was positive in 17/23(73.9%) patients and IgG was positive in 16/23(69%) *** of the non TBP had either ZN,PCR or positive serum IgA,IgG,or *** the positive results for IgG and IgM were combined together the serological tests correcdy identified 20/23(87%) of patients. Conclusion:We suggest that in TBP serological diagnosis by combined use of IgG and IgM for A60 antigen with serum determination of interferon gamma can provide rapid and non invasive diagnostic tool that can justify the starting of chemotherapy while awaiting the results of culture.

Molecular study of astrovirus, adenovirus and norovirus in community acquired diarrhea in children: One Egyptian center study

Objective: To determine the prevalence of astrovirus, norovirus, adenovirus in children below five years old with diarrhea by multiplex reverse transcriptase polymerase chain reaction(RT-PCR) along with rotavirus antigen detection by enzyme linked immunosorbant ***: The study was conducted on children below five years old complaining of acute diarrhea. The study included stool examination by molecular method for detection of norovirus, adenovirus and astrovirus by multiplex RT-PCR. Rotavirus antigen was detected in the stool by enzyme linked immunosorbant ***: The study included 100 children below 5 years old with acute *** RT-PCR was positive in 34% of the children. The most frequently detected virus was rotavirus(44%), followed by norovirus(30%), adenovirus(20%) and astrovirus(14%). The clinical symptoms were more significantly associated with viral diarrhea such as fever(P = 0.03), bloody diarrhea(P = 0.025), vomiting(P = 0.000 1) and watery diarrheas(P = 0.05). The frequency of diarrhea with viral pathogen was significantly presented in winter season(39.7%). There were significant frequencies of norovirus and adenovirus in age ranging 1–2 years old(P = 0.04, P = 0.01 respectively).Conclusions: The present study spotlights on the prevalence of viral pathogens as an important etiology in diarrhea in children below five years old. Astrovirus, norovirus and adenovirus are common along with rotavirus in this group of patients. Multiplex PCR leads to improve the laboratory diagnosis of these viruses along with antigen detection method. Further longitudinal studies are required to evaluate the epidemiological data associated with these viruses and for proper management of such drastic infection.

Correlation of biofilm formation of uropathogenic Escherichia coli(UPEC) and fimbriae genotypes

Value of avidity index of IgG and polymerase chain reaction in diagnosis of herpes simplex 2 virus in pregnancy

Objective:The goal of the present study is to highlight the significance of the determination of IgG avidity to herpes simplex2virus(HSV2) compared to detection of viremia by polymerase chain reaction(PCR) in relation to recurrent ***:Serum samples from pregnant patients with bad obstetric histories and control subjects were analyzed for quantitative determination of IgG specific for HSV2 by using elISA with and without sample treatment with 8 M urea as protein-denaturing agents for measurement of avidity index(AI).PCR was performed for samples to determine HSV2 ***:Regarding avidity index 11.5%had avidity index = 100.0,11.5%has avidity index = 47%and 76.9%had avidity index>60%.Control group had AI>70%.There was statistically insignificant difference between OD of IgG in patients with viremia before urea treatment and after urea treatment compared to healthy *** there was statistically insignificant difference in OD before urea and after urea in patients with ***:From this study we can conclude that viremia for HSV2 is common finding in patients with bad obstetric *** avidity immunoglobulin G was not common finding in those *** may be explain that even reactivation of latent HSV 2 infection is associated with fetal loss.

Sentiment analysis attracts the attention of Egyptian Decisionmakers in the education *** offers a viable method to assess education quality services based on the students’feedback as well as that provides an understanding of their *** machine learning techniques offer automated strategies to process big data derived from social media and other digital channels,this research uses a dataset for tweets’sentiments to assess a few machine learning *** dataset preprocessing to remove symbols,necessary stemming and lemmatization is performed for features *** is followed by several machine learning techniques and a proposed Long Short-Term Memory(LSTM)classifier optimized by the Salp Swarm Algorithm(SSA)and measured the corresponding ***,the validity and accuracy of commonly used classifiers,such as Support Vector Machine,Logistic Regression Classifier,and Naive Bayes classifier,were ***,LSTM based on the SSA classification model was compared with Support Vector Machine(SVM),Logistic Regression(LR),and Naive Bayes(NB).Finally,as LSTM based SSA achieved the highest accuracy,it was applied to predict the sentiments of students’feedback and evaluate their association with the course outcome evaluations for education quality purposes.

Dietary effects of A zolla pinnata combined with exogenous digestive enzyme(Digestin^(TM)) on growth and nutrients utilization of freshwater prawn, Macrobrachium rosenbergii(de Man 1879)

The present study was conducted to evaluate the effect of either individual or combined wheat bran(WB) replacement with Azolla pinnata supplemented with Digestin^(TM) in the diet of freshwater prawn, Macrobrachium rosenbergii Postlarvae(PL) on growth performance, nutrient utilization, chemical body composition and survival(%). Experimental diets were a wheat bran-soybean based diet with no Azolla and Digestin^(TM)(control, T_1), and diets containing 17% Azolla supplemented with Digestin^(TM) 0%(T_2), 1%(T_3), 2%(T_4) and 3%(T 5). Each experimental diet was allocated into three tanks(6 m^3/tank) fed for 12 wks. Each tank was subdivided into three equal pens by nets(2 m^3) and stoked with 84 PL/m^2. The experimental diets were readily consumed by prawns PLs where both high growth and good feed efficiency were achieved for all diets. The results showed that the diets containing A. pinnata supplemented with Digestin? at the level up to 3% have the higher growth and better nutrient utilization than the control diet. No differences were observed for moisture and protein content among the experimental diets. However, the highest protein content was observed on prawns fed on diets T_1 and T 5 respectively, while the lowest value was recorded for T_4 diet. The results also show that prawn PLs fed the diets contain A. pinnata and supplemented with Digestin^(TM) recorded the highest values of body lipid content compared to the control diet. Feed efficiency and economic conversion rate(ECR) values show that economic performance and the cost-effectiveness of the A. pinnata supplemented with up to 3% Digestin^(TM) recorded the highest net return, and therefore it is recommended for prawn, M. rosenbergii PL's. These results are clearly indicating that A. pinnata have a good potential for use in prawn diets at reasonable levels than other conventional diets.
