Effects of methyl palmitate and lutein on LPS-induced acute lung injury in rats

AIM: To investigate the effects of methyl palmitate and lutein on lipopolysaccharide(LPS)-induced acute lung injury(ALI) in rats and explore the possible mechanisms. METhODS: Male Sprague-Dawley rats were divided into 4 groups:(1) control;(2) LPS;(3) Methyl palmitate; and(4) Lutein groups. Methyl palmitate(300 mg/kg, ip) was administered 3 times per week on alternating days while lutein(100 mg/kg, oral) was given once daily. After 1 wk of vehicle/methyl palmitate/lutein treatment, ALI was induced by a single dose of LPS(7.5 mg/kg, iv). After 24 h of LPS injection, animals were sacrificed then biochemical parameters and histopathology were assessed. RESULTS: Treatment with methyl palmitate attenuated ALI, as it significantly decreased the lung wet/dry weight(W/D) ratio, the accumulation of the inflammatory cells in the bronchoalveolar lavage fluid(BALF) andhistopathological damage. however, methyl palmitate failed to decrease lactate dehydrogenase(LDh) activity in BALF. On the other hand, lutein treatment produced significant anti-inflammatory effects as revealed by significant decrease in accumulation of inflammatory cells in lung, LDh level in BALF and histopathological damage. Methyl palmitate and lutein significantly increased superoxide dismutase(SOD) and reduced glutathione(GSh) activities with significant decrease in the lung malondialdehyde(MDA) content. Importantly, methyl palmitate and lutein decreased the level of the inflammatory cytokine tumor necrosis factor-α(TNF-α) in the lung. Lutein also reduced LPS-mediated overproduction of pulmonary nitrite/nitrate(NO-2/NO-3), which was not affected by methyl palmitate pretreatment. CONCLUSION: These results demonstrate the potent protective effects of both methyl palmitate and lutein against LPS-induced ALI in rats. These effects can be attributed to potent antioxidant activities of these agents, which suppress inflammatory cell infiltration and regulated cytokine effects.

Study of Radionuclides and Radon Exhalation Rate in Soil and Sand Samples from Tiba, Luxor, Governorate

In this study, the natural radionuclides in soil and sand have been measured by using high purity germanium (hPGe) detector. While, radon exhalation rate has been measured by Alpha GUARD. The data analysis is performed to determine 226Ra, 232Th, and 40K activity concentrations in addition to 222Rn exhalation rate. The values of radium equivalent activity (Raeq), external hazard index (hex), internal hazard index (hin), and absorbed dose rate were ranged from 46.46 to 124.16 Bq⋅kg−1, 0.07 to 0.33 Bq⋅kg−1, 0.09 to 0.42 Bq⋅kg−1, and 13.24 to 58.37 nGy⋅h−1 respectively in all samples. The area and mass exhalation rates were increased from 9.16 ±2.83 to 16.18 ±2.83 Bq⋅m−2⋅h−1 and 1.8 ±1.34 to 11.35 ±0.98 Bq⋅kg−1⋅h−1 respectively.

Immunotherapy becomes a promising line of treatment for breast cancer(BC)however,its success rate is still ***:The study was designed to optimize the condition for producing an effective dendritic cell(DCs)based immunotherapy by using DCs and T lymphocytes together with tumor-inﬁltrating lymphocytes(TILs)and tumor-inﬁltrating DCs(TIDCs),treated with anti-PD1 and anti-CTLA4 monoclonal *** mixture of immune cells was co-cultured with autologous breast cancer cells(BCCs)isolated from 26 BC ***:There was a signiﬁcant upregulation of CD86 and CD83 on DCs(p=0.001 and 0.017,respectively),similarly upregulation of CD8,CD4 and CD103 on T cells(p=0.031,0.027,and 0.011,respectively).While there was a signiﬁcant downregulation of FOXP3 and combined ***8 expression on regulatory T cells(p=0.014 for both).Increased CD8/Foxp3 ratio(phere was a signiﬁcant increase in interferon-γ(IFN-γ,phydrogenase(LDh,p=0.02),and a signiﬁcant decrease in vascular endothelial growth factor(VEGF,ph ligand 1(PDL-1)were downregulated in BCCs(ph),similarly cytotoxic T lymphocyte antigen-4(CTLA4,p=0.02),Programmed cell death 1(PD-1,ph immune checkpoint inhibitors could produce a potent and effective BC ***,these data should be validated on an experimental animal model to be transferred to the clinical setting.

Flow cytometric detection of hepatitis C virus antigens in infected peripheral blood leukocytes: Binding and entry

AIM： We designed two synthetic-core-specific peptides core 1 （C1） and core 2 （C2）, and an E1-specific peptide （El）. We produced specific polyclonal antibodies against these peptides and used the antibodies for detection of hCV antigens on surface and within infected peripheral blood leukocytes. METhODS： Peripheral blood from a healthy individual who tested negative for hCV RNA was incubated with hCV type 4 infected serum for i h and 24 h at 37 ℃. Cells were stained by direct and indirect immunofluorescence and measured by flow cytometry. RESULTS： After 1 h of incubation, antibodies against C1, C2, and El detected hCV antigens on the surface of 27%, 26% and 73% of monocytes respectively, while 10%, 5% and 9% of lymphocytes were positive with anti-C1, anti-C2 and anti-E1 respectively. Only 1-3% of granulocytes showed positive staining with anti-C1, anti-C2 and anti E1 antibodies. After 24 h of incubation, we found no surface staining with anti-C1, anti-C2 or anti-E1. Direct immunostaining using anti-C2 could not detect intracellular hCV antigens, after 1 h of incubation with the virus, while after 24 h of incubation, 28% of infected cells showed positive staining. Only plus strand RNA was detectable intracellularly as early as 1 h after incubation, and remained detectable throughout 48 h post-infection. Interestingly, minus RNA strand could not be detected after 1 h, but became strongly detectable intracellularly after 24 h post-infection. CONCLUSION： Monocytes and lymphocytes are the preferred target cells for hCV infection in peripheral blood leukocytes. Our specific anti-core and anti-E1 antibodies are valuable reagents for demonstration of hCV cell cycle. Also, hCV is capable of infecting and replicating in peripheral blood mononuclear cells as confirmed by detection of minus strand hCV RNA as well as intracellular staining of core hCV antigen.

Phytochemical and anti-neuropathic investigations of Crocus sativus via alleviating inflammation, oxidative stress and pancreatic beta-cells regeneration

Objective:The aim of this study is to investigate the phytochemical and the long-term anti-neuropathic potentials of Crocus sativus cultivated in the University botanical garden,and explore its most bioactive compounds and their underlying mechanisms of ***:Phytochemical analysis and bio-guided isolation-procedures including RP-hPLC and1 h and13 C NMR utilizing biological models of diabetes,inflammation,and diabetic-neuropathy were *** saffron(S-RCED)and Spanish-saffron stigma(S-SP)alone or in combination with Camellia sinus(CS)were ***:The RP-hPLC analyses showed the presence of picrocrocin,crocin I,crocin II,crocin I’,crocin II’,and safranal(SAF)in both S-SP and S-RCED extracts with *** had been shown that SAF was the most bioactive-compound in Crocus *** S-SP and S-RCED possessed significant(Ph),subchronic(8 d)and chronic(8 weeks)models.S-RCED had been proven with more hypoglycemic potentials when compared to S-SP and SAF.S-SP,S-RCED,and SAF produced significant anti-inflammatory and anti-nociceptive activities against carrageenan-induced inflammatory,hyperalgesic and tactile diabetic-neuropathy models,respectively.S-SP,S-RCED,and SAF elevated serum catalase,reduced glutathione,and insulin serum levels,ameliorated lipid peroxidation and hb A1 c levels,and histopathologically regenerated the pancreatic *** with CS showed more significant efficacy than the single ***:The oxidative stress reduction,insulin secretagogue,and pancreatic beta-cells regeneration potentials might be responsible for the mechanism underlying the anti-diabetic,anti-inflammatory and anti-diabetic neuropathy ***,the cultivated Crocus sativus might be clinically useful for protecting against many serious-disorders.

Big data and information and communication technologies can be important to the effectiveness of smart *** on the maximal attention on smart city sustainability,developing data-driven smart cities is newly obtained attention as a vital technology for addressing sustainability ***-time monitoring of pollution allows local authorities to analyze the present traffic condition of cities and make *** to air pollution occurs a main environmental problem in smart city *** effect of the deep learning(DL)approach quickly increased and penetrated almost every domain,comprising air pollution ***,this article develops a new Coot Optimization Algorithm with an Ensemble Deep Learning based Air Pollution Prediction(COAEDL-APP)system for Sustainable Smart *** projected COAEDL-APP algorithm accurately forecasts the presence of air quality in the sustainable smart city *** achieve this,the COAEDL-APP technique initially performs a linear scaling normalization(LSN)approach to pre-process the input *** air quality prediction,an ensemble of three DL models has been involved,namely autoencoder(AE),long short-term memory(LSTM),and deep belief network(DBN).Furthermore,the COA-based hyperparameter tuning procedure can be designed to adjust the hyperparameter values of the DL *** simulation outcome of the COAEDL-APP algorithm was tested on the air quality database,and the outcomes stated the improved performance of the COAEDL-APP algorithm over other existing systems with maximum accuracy of 98.34%.

First Report of the Bee Fly Eurycarenus dichopticus Bezzi (Diptera: Bombyliidae) in North Kordofan State, Sudan

Sudan’s semi-arid land constitutes a diversity of habitats, species, and genetic resources of high ecological and economic value. This diversity is gradually degrading due to climate change and human misuse. Biodiversity monitoring began in Sudan before independence when a considerable number of flora and fauna species were identified. Insects were among the identified fauna where many orders, families, and species were classified especially those of economic, medicinal, and veterinary importance. Bombyliidae or bee flies are considered native to African countries and confined to the semi-arid ecosystem as pollinators of higher plants. The bee fly, Eurycarenus dichopticus recorded from these ecosystems, has gained little attention, and meager data on its biology and reproduction have been published. Adults of this insect are nectar feeders, and females also feed on pollen grains while larvae are parasitoids on certain insects. This study was conducted in three Sudanese states that lie within the semi-arid ecosystem;Khartoum, Gezira, and North Kordofan. Field surveys were conducted in these states and insect fauna was obtained and identified. Bee fly specimens were collected from North Kordofan using the butterfly net method. The specimens were then preserved and sent to the Agricultural Research Corporation ARC in (2020) where they were identified as Eurycarenus dichopticus Bezzi (Diptera: Bombyliidae) which is the first report in North Kordofan. This identification may contribute to the understanding of its ecological significance and role.

Improvement of hematopoietic and Immunologic Findings in Sublethal Gamma Irradiated Rats Treated with Bone Marrow Transplantation and Wheat Germ Oil

The objective of this study was to evaluate the efficacy of the wheat germ oil (WGO) and bone marrow transplantation (BMT) in boosting the immuno response and protecting from oxidative stress in irradiated rats. BM was given by intravenous injection to male rats, one hour post gamma irradiation at the dose level of 5 Gy. Rats were orally administrated with 54 mg/Kg body wt of wheat germ oil daily for 2 weeks before irradiation. After 14 days, results revealed that total body irradiation induced significant decreases in RBCs, WBCs and lymphocytes, as well as Glutathione (GSh) and zinc superoxide dismutase (Zn/SOD), splenocyte count, bone marrow lymphocyte count and viability. Tumor necrosis factor alpha (TNF-α) and interleukin 2 (IL-2) also recorded significant decrease while interleukin 6 (IL-6) and lipid peroxidation marker malondialdehde (MDA) in serum and spleen were conversely elevated. In irradiated animals receiving BMT and WGO, values of MDA in serum and tissue were significantly depressed as compared with the irradiated group, while lymphocytes, bone marrow viability percentage, splenocytes percentage, IL-2, IL-6 and GSh were significantly elevated. The curative action of WGO enforcing significant innate response could trigger and augment adaptive immune response by BMT, thus protecting immune system from radiation induced damage as well as oxidative stress.
