Flow cytometric detection of hepatitis C virus antigens in infected peripheral blood leukocytes: Binding and entry

AIM： We designed two synthetic-core-specific peptides core 1 （C1） and core 2 （C2）, and an E1-specific peptide （El）. We produced specific polyclonal antibodies against these peptides and used the antibodies for detection of hCV antigens on surface and within infected peripheral blood leukocytes. METhODS： Peripheral blood from a healthy individual who tested negative for hCV RNA was incubated with hCV type 4 infected serum for i h and 24 h at 37 ℃. Cells were stained by direct and indirect immunofluorescence and measured by flow cytometry. RESULTS： After 1 h of incubation, antibodies against C1, C2, and El detected hCV antigens on the surface of 27%, 26% and 73% of monocytes respectively, while 10%, 5% and 9% of lymphocytes were positive with anti-C1, anti-C2 and anti-E1 respectively. Only 1-3% of granulocytes showed positive staining with anti-C1, anti-C2 and anti E1 antibodies. After 24 h of incubation, we found no surface staining with anti-C1, anti-C2 or anti-E1. Direct immunostaining using anti-C2 could not detect intracellular hCV antigens, after 1 h of incubation with the virus, while after 24 h of incubation, 28% of infected cells showed positive staining. Only plus strand RNA was detectable intracellularly as early as 1 h after incubation, and remained detectable throughout 48 h post-infection. Interestingly, minus RNA strand could not be detected after 1 h, but became strongly detectable intracellularly after 24 h post-infection. CONCLUSION： Monocytes and lymphocytes are the preferred target cells for hCV infection in peripheral blood leukocytes. Our specific anti-core and anti-E1 antibodies are valuable reagents for demonstration of hCV cell cycle. Also, hCV is capable of infecting and replicating in peripheral blood mononuclear cells as confirmed by detection of minus strand hCV RNA as well as intracellular staining of core hCV antigen.

Exploitation of Fenton and Fenton-like reagents as alternative conditioners for alum sludge conditioning

The use of Fenton＇s reagent （Fe^2＋/h2O2） and Fenton-like reagents containing transition metals of Cu（Ⅱ）, Zn（Ⅱ）, Co（Ⅱ）, and Mn（Ⅱ） for an alum sludge conditioning to improve its dewaterability was investigated. The results obtained were compared with those obtained from conditioning the same alum sludge using cationic and anionic polymers. Experimental results show that Fenton＇s reagent was the best among the Fenton and Fenton-like reagents for the alum sludge conditioning. A considerable effectiveness of capillary suction time （CST） reduction efficiency of 47% can be achieved under test conditions of Fe^2＋/h2O2 = 20/125 mg/g DS （dry solid） and ph 6.0. The observation of floc-like particles after Fenton＇s reagent conditioning of alum sludge suggested that the mechanism of Fenton＇s reagent conditioning was different from that of polymer conditioning. In spite of the lower efficiency in the CST reduction of Fenton＇s reagent in alum sludge conditioning compared to that of polymer conditioning, Fenton＇s reagent offers a more environmentally safe option. Tiffs study provided an example of proactive treatment engineering, which is aimed at seeking a safe alternative to the use of polymers in sludge conditioning towards achieving a more sustainable sludge management strategy.

The effect of addition of Zn, Cu, Pb and Cr chlorides as admixtures on the hydration reaction of the system 3CaO·Al2O3-gypsum with molar ratio 1：3 was studied. Different ratios of each salt were used, namely 0.5%, 2% and 4% by weight of the solid mixture. hydration reaction was carried out at 35℃ for various time intervals from 0.5 h to up to 7 d. hydration rate of the system 3CaO·Al2O3-CaSO4·2h2O in absence and presence of different salts was studied via the determination of the combined water contents. X-ray diffraction analysis showed that the ettringite was the only hydration product formed in the different mixes. The hydration products were investigated by scanning electron microscopy （SEM） and thermal gravimetric analysis. The results indicated that the rate of formation of ettringite and its microstructure depend on the admixture and its dosage.

Objective:To green synthesize and characterize copper nanoparticles(Cu NPs)using Astragalus sinicus,as well as evaluate the acaricidal,larvacidal,and repellent activities of Cu NPs against hyalomma anatolicum(***),one of the most prevalent ticks infesting cattle in Saudi ***:Cu NPs were green synthesized by adding the Astragalus sinicus extract to a copper sulfate *** acaricidal,larvicidal,and repellent activities of Cu NPs against *** were assessed via the adult immersion test,the larval packet test,and the vertical movement behavior of tick larvae,*** effects of Cu NPs on acetylcholinesterase as well as oxidative enzyme activities were ***:The green synthesized Cu NPs displayed a spherical form with a size range of 15-75 *** exposure of adult *** to different concentrations of Cu NPs,the viability rate of adult *** and the mean number,weight,and hatchability of eggs were noticeably reduced,in comparison to the control group(Phe viability rate of larvae considerably declined(Ph the LC_(50)and LC_(90)values of 11.30 and 20.34μg/m L,*** maximum repellent activity of Cu NPs was observed at 50,100,and 200μg/m L with complete repellent activity after 60,120,and 180 min of exposure,*** NPs,mainly at 1/2 LC_(50)and LC_(50)concentrations,markedly suppressed the acetylcholinesterase activity of the larval stage of ***(Phyde level while declining glutathione-S-transferase level in *** larvae(Phow potent acaricidal,larvicidal,and repellent activities against adults and larvae of ***,further studies must be performed to clarify the precise mechanisms and the efficacy of Cu NPs in practical use.

BACKGROUND One of the most popular bariatric procedures is sleeve gastrectomy,and it has become significantly more common in recent *** To evaluate the research activity in sleeve gastrectomy over the last two decades,and to visualize the hot spots and emerging trends in this type of bariatric surgeryusing bibliometric *** The Scopus database was used to search for publications related to sleeve *** retrieved publications were reviewed in terms of year of publication,type of study,country of origin,institutions,journals,and citation patterns by using descriptive *** network and term cooccurrence analysis were visualized by using VOSviewer *** The search strategy yielded a total of 6508 publications on sleeve gastrectomy from 2001 to *** regards the document type,the majority were articles(n=5230;80.36%),followed by reviews(n=544;8.36%).The top three countries are the United States,with 1983 publications(30.47%),followed by France(600;9.22%)and Italy(417;6.71%).The most cited publication was published in 2012 by Schauer et al in the New England Journal of Medicine(n=1435 citations).This publication found that weight loss was greater in the sleeve gastrectomy group than in the medical therapy ***,this study demonstrated that 12 mo of medical therapy plus bariatric surgery greatly improved glycemic regulation in obese patients with uncontrolled type 2 diabetes compared with medical therapy *** focus of the current literature on sleeve gastrectomy was directed toward several themes such as morbidity and potential complications,the complexity of the procedure and different surgical approaches,and diabetes and body mass index in correlation to sleeve *** The number of sleeve gastrectomy publications has gradually grown over the last 20 *** bibliometric analysis could help researchers better understand the knowledge base and research frontiers surrounding sleeve

Evaluation of antidiabetic activity of plants used in Western Sudan

Objective: To investigate the traditional antidiabetic uses of some indigenous Sudanese plants on streptozotocin-induced diabetes rats. Methods: Diabetic rats were treated with a 400 mg/kg dose of aqueous extracts of five plant species orally for 2 h(acute) or 14 days(chronic). In acute model blood glucose levels were monitored at specific intervals. In the chronic model blood samples were collected from overnight fasted diabetic rats on day 15 to estimate blood glucose level. And the body weight, serum lipid profile and activities of liver and kidney enzymes were measured. histopathological observations of liver sections were also ***: In the case of acute treatment, aqueous extracts of Tinospora bakis(T. bakis), Nauclea latifolia(N. latifolia) and Randia nilotica(R. nilotica) at 400 mg/kg significantly lowered(P hereas, chronic treatment of diabetic rats with 400 mg/kg of T. bakis, N. latifolia, R. nilotica and Mitragyna inremis proved to have significant(P hyperglycemic effect and have the capacity to correct the metabolic disturbances associated with diabetes. histopathological studies showed that the aqueous extracts of these four plants reinforced the healing of liver. however, Striga hermonthica aqueous extract did not exert any antihyperglycemic effect to diabetic rats. Conclusions: This study demonstrated that T. bakis, N. latifolia, R. nilotica and Mitragyna inremis have therapeutic value in diabetes and related complications and thus supporting the traditional uses of these plants in Sudanese traditional medicine.

Electrochemical hydrogen compression and purification versus competing technologies: Part Ⅰ. Pros and cons

It is undisputed that hydrogen will play a great role in our future energetic mix, because it enables the storage of renewable electricity(power-to-h2) and the reversible conversion into electricity in fuel cell, not to speak of its wide use in the(petro)chemical industry. Whereas in these applications, pure hydrogen is required, today’s hydrogen production is still largely based on fossil fuels and can therefore not be considered pure. Therefore, purification of hydrogen is mandatory, at a large scale. In addition, hydrogen being the lightest gas, its volumetric energy content is well-below its competing fuels, unless it is compressed at high pressures(typically 70 MPa), making compression unavoidable as well. This contribution will detail the means available today for both purification and for compression of hydrogen. It will show that among the available technologies, the electrochemical hydrogen compressor(EhC), which also enables hydrogen purification, has numerous advantages compared to the classical technologies currently used at the industrial scale. EhC has their thermodynamic and operational advantages, but also their ease of use. however, the deployment of EhCs will be viable only if they reach sufficient performances, which implies some specifications that their base materials should stick to. The present contribution will detail these specifications.

Venous thromboembolism prophylaxis-prescribing patterns among elderly medical patients in a Saudi tertiary care center:success or failure?

Background hospitalized elderly patients are at high risk of venous thromboembolism(VTE),and the appropriate use of thromboprophylaxis can significantly reduce the incidence of VTE in high-risk *** investigated the pattern of VTE prophylaxis administration among elderly medical patients and assessed its appropriateness based on the American College of Chest Physicians(ACCP)*** A cross-sectional single-center study was conducted between October 2019 and March 2020,including hospitalized(>48 h),elderly(≥60 years),medical patients,and excluding patients receiving anticoagulant for other reason,having contraindication to thromboprophylaxis,or had VTE diagnosed within 48 *** Padua prediction score was used to determine the patients’risk for VTE,and thromboprophylaxis use was assessed against the ACCP *** The study included 396 patients with an average age of 75.0±9.01 years,and most patients(71.7%)were classified as high risk for VTE development(Padua score≥4 points).Thromboprophylaxis use was inappropriate in 27.3%of patients,of whom 85.2%were ineligible but still received *** who were classified as low risk of VTE were more likely to receive inappropriate thromboprophylaxis(AOR=76.5,95%CI:16.1-363.2),whereas patients with acute infection or rheumatologic disorder were less likely to receive inappropriate thromboprophylaxis(AOR=0.46,95%CI:0.22-0.96).Conclusions Although the use of thromboprophylaxis among high-risk elderly patients was reasonably adequate,a large proportion of low-risk patients were exposed to unnecessary risk through inappropriate overutilization of ***,healthcare providers should accurately assess patients’risk before prescribing thromboprophylaxis to ensure patient safety.

The Solitary Wave Solutions of Important Model in Particle Physics and Engineering According to Two Different Techniques

In this paper, we utilized the Jaulent-Miodek equation which is one of important models in particle physics and engineering. The exact traveling wave solutions for this equation “involving parameters” according to two different techniques are constructed. When these parameters are taken as special values, the solitary wave solutions are derived from it. A comparison between the obtained results using these two different methods with that obtained by previous authors is demonstrated.
