High-throughput sequencing analysis of differential microRNA expression in the process of blocking the progression of chronic atrophic gastritis to gastric cancer by Xianglian Huazhuo formula(香连化浊方)

OBJECTiVE: To explore the mechanism of Xianglian Huazhuo formula( 香连化浊方, XLHZ) blocking the development of chronic atrophic gastritis(CAG) to gastric cancer(GC) through bioinformatics analysis and in vitro. METHODS: Pathological morphology of gastric mucosa of rats were observed. High-throughput sequencing was used to analyze the miRNA expression profile of gastric mucosa. The miRanda, miRDB and miRWalk databases were used to predict the differential target genes. Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis were performed for differential target genes. Real-time quantitative reverse transcription polymerase chain reaction(qRTPCR) was used to verify the differentially expressed miRNAs and target genes. Western blot, EdU, wound healing and flow cytometry were used to observe the effect of XLHZ on epithelial-mesenchymal transition(EMT) markers, proliferation, migration, apoptosis and cell cycle of CAG cells in vitro. RESULTS: A total of five differentially expressed miRNAs and four differential target genes were screened in this study. GO analysis showed that the target genes were enriched in regulation of neuron development, regulation of transcription factor activity and regulation of RNA polymerase. KEGG pathways database differences in gene enrichment of target genes in the Wnt signaling pathway, Phospholipase D signaling pathway and mitogen-activated protein kinase signaling pathway. qRTPCR confirmed that miRNAs and its target genes were consistent with the screening results. in vitro, our study revealed that XLHZ could increase the expression of Ecadherin, decrease the expression of transforming growth factor β1, vimentin and β-catenin, inhibite the proliferation and migration of CAG cells, cause cell cycle arrest at G0/G1 and G2/M phase, induce the apoptosis of CAG cells, and prevent the progression of CAG to GC. Brain mechanism of acupuncture for children with anisometropic amblyopia: a resting functional magnetic resonance imaging study based on voxel-mirrored homotopic connectivity

AiM:To explore the brain mechanism of acupuncture for children with anisometropic amblyopia using the voxelmirror homotopic connectivity(VMHC)analysis method of resting functional magnetic resonance imaging(rs-fMRi)technology based on clinical ***:Eighty children with anisometropic monocular amblyopia were randomly divided into two groups:control(40 cases,1 case of shedding)and acupuncture(40 cases,1 case of shedding)*** control group was treated with glasses,red flash,grating,and visual stimulations,with each procedure conducted for 5min per *** on routine treatment,the acupuncture group underwent acupuncture of“regulating qi and unblocking meridians to bright eyes”,Jingming(BL1),Cuanzhu(BL2),Guangming(GB37),Fengchi(GB20)acupoints were taken on both sides,with the needle kept for 30min each *** groups were treated once every other day,three times per week,for a total of *** the treatment,the overall curative effect of the two groups and the latency and amplitude changes of P100 wave of pattern visual-evoked potential were *** the same time,nine children with left eye amblyopia were randomly selected from the two groups and were scanned with rsfMRi before and after *** differences in the brain regions between the two groups were compared and analyzed with ***:Chi-square test showed a notable difference in the total efficiency rate between the acupuncture(94.87%)and control groups(79.49%).Regarding the P100 wave latency and amplitude,the acupuncture group had significantly shorter latency and higher amplitude of P100 wave than the control ***,the VMHC values of the bilateral temporal lobe,superior temporal gyrus,and middle temporal gyrus were notably increased in the acupuncture group after ***:Acupuncture combined with conventional treatment can significantly improve the corrected visual acuity and optic nerve conduction in children with anisometropic ***

Triboelectric nanogenerator(TENG)converts mechanical energy into valuable electrical energy,offering a solution for future energy *** an indispensable part of TENG,textile TENG(T-TENG)has incredible advantages in harvesting biomechanical energy and physiological signal ***,the application of T-TENG is restricted,partly because the fabric structure parameter and structure on T-TENG performance have not been fully *** study comprehensively investigates the effect of weaving structure on fabric TENGs(F-TENGs)for direct-weaving yarn TENGs and post-coating fabric *** direct-weaving F-TENGs,a single-yarn TENG(Y-TENG)with a core-sheath structure is fabricated using conductive yarn as the core layer yarn and polytetrafluoroethylene(PTFE)filaments as the sheath *** fabrics with five different sets of parameters were designed and *** post-coating F-TENGs,fabrics with weaving structures of plain,twill,satin,and reinforced twill were fabricated and coated with conductive silver ***,the twill F-TENGs have the best electrical outputs,followed by the satin F-TENGs and plain weave ***,the increase of the Y-TENG gap spacing was demonstrated to improve the electrical output ***,T-TENGs are demonstrated for human-computer interaction and self-powered real-time *** systematic work provides guidance for the future T-TENG’s design.

Valence-tailored copper-based nanoparticles for enhanced chemodynamic therapy through prolonged ROS generation and potentiated GSH depletion

Chemodynamic therapy(CDT),an inventive approach to cancer treatment,exploits innate chemical processes to trigger cell death through the generation of reactive oxygen species(ROS).While offering advantages over conventional treatments,the optimization of CDT efficacy presents challenges stemming from suboptimal catalytic efficiency and the counteractive ROS scavenging effect of intracellular glutathione(GSH).in this study,we aim to address this dual challenge by delving into the role of copper valence states in *** the unique attributes of copper-based nanoparticles,especially zero-valent copper nanoparticles(CuPd NPs),we aim to enhance the therapeutic potential of *** experiments reveal that zero-valent CuPd NPs outperform divalent copper nanoparticles(Ox-CuPd NPs)by displaying superior catalytic performance and sustaining ROS generation through a dual approach integrating peroxidase-like(POD-like)activity and Cu+***,zero-valent NPs exhibit enhanced GSH depletion compared to their divalent counterparts,thereby intensifying CDT and inducing ferroptosis,ultimately resulting in high-efficiency tumor growth *** findings reveal the impact of valences on CDT,providing novel insights for the optimization and design of CDT agents.

Si-Wu-Tang attenuates liver fibrosis via regulating lncRNA H19-dependent pathways involving cytoskeleton remodeling and ECM deposition

Liver fibrosis is a dynamic wound-healing response characterized by the agglutination of the extracellular matrix(ECM).Si-Wu-Tang(SWT),a traditional Chinese medicine(TCM)formula,is known for treating gynecological diseases and liver *** previous studies demonstrated that long non-coding RNA H19(H19)was markedly upregulated in fibrotic livers while its deficiency markedly reversed ***,the mechanisms by which SWT influences H19 remain ***,we established a bile duct ligation(BDL)-induced liver fibrosis model to evaluate the hepatoprotective effects of SWT on various cells in the *** results showed that SWT markedly improved ECM deposition and bile duct reactions in the ***,SWT relieved liver fibrosis by regulating the transcription of genes involved in the cytoskeleton remodeling,primarily in hepatic stellate cells(HSCs),and influencing cytoskeleton-related angiogenesis and hepatocellular *** modulation collectively led to reduced ECM *** extensive bioinformatics analyses,we determined that H19 acted as a miRNA sponge and mainly inhibited miR-200,miR-211,and let7b,thereby regulating the above cellular regulatory ***,SWT reversed H19-related miRNAs and signaling pathways,diminishing ECM deposition and liver ***,these protective effects of SWT were diminished with the overexpression of H19 in *** conclusion,our study elucidates the underlying mechanisms of SWT from the perspective of H19-related signal networks and proposes a potential SWT-based therapeutic strategy for the treatment of liver fibrosis.

Comparison of inebilizumab or rituximab in addition to glucocorticoid therapy for neuromyelitis optica spectrum disorders

AiM:To investigate the short-term efficacy and safety of inebilizumab for neuromyelitis optica spectrum disorders(NMOSD).METHODS:A total of 33 patients with NMOSD treated with inebilizumab(Group iNB,n=15)or rituximab(Group RTX,n=18)in addition to high-dose glucocorticoids were *** groups underwent hormone shock therapy during the acute ***,Group iNB received inebilizumab injections during the remission phase,while Group RTX received rituximab injections.A comparison of aquaporins 4(AQP4)titer values,peripheral blood B lymphocyte counts,and visual function recovery was conducted before and 8wk after ***,adverse reactions and patient tolerability were analyzed after using inebilizumab treatment ***:Following inebilizumab treatment,there was a significantly improvement in the visual acuity of NMOSD patients(Pied by a notable decrease in AQP4 titer values and B lymphocyte ratio(Pinebilizumab treatment showed a partial effect in preventing optic nerve atrophy(Pignificant differences in other therapeutic effects compared to rituximab,which has previously demonstrated substantial therapeutic efficacy(P>0.05).Furthermore,inebilizumab exhibited higher safety levels than that of rituximab ***:The combination of inebilizumab and high-dose glucocorticoids proves to be *** comparison to rituximab injections,inebilizumab displays better tolerance and ***,it demonstrates a partial effect in preventing optic nerve ***,it stands as an effective method to reduce the disability rates and improve the daily living ability of patients with NMOSD.
