Atomic force microscopy(AFm)has been consistently supporting nanosciences and nanotechnologies for over 30 years and is used in many fields from condensed matter physics to *** enables the measurement of very weak forces at the nanoscale,thus elucidating the interactions at play in fundamental ***,we leverage the combined benefits of micro/nanoelectromechanical systems and cavity optomechanics to fabricate a sensor for dynamic mode AFm at a frequency above 100 *** frequency is two decades above the fastest commercial AFm probes,suggesting an opportunity for measuring forces at timescales unexplored thus *** fabrication is achieved using very-large-scale integration technologies derived from photonic silicon *** probe's optomechanical ring cavity is coupled to a 1.55 pm laser light and features a 130 mHz mechanical resonance mode with a quality factor of 900 in air.A limit of detection in the displacement of 3×10^(-16)m/VHz is obtained,enabling the detection of the Brownian motion of the probe and paving the way for force sensing experiments in the dynamic mode with a working vibration amplitude in the picometer *** inserted in a custom AFm instrument embodiment,this optomechanical sensor demonstrates the capacity to perform force-distance measurements and to maintain a constant interaction strength between the tip and sample,an essential requirement for AFm *** indeed show a stable closed-loop operation with a setpoint of 4 nN/nm for an unprecedented subpicometer vibration amplitude,where the tip-sample interaction is mediated by a stretched water meniscus.

Aim:Angiogenesis is observed in acute myeloid leukemia(AmL).AmL cells abnormally proliferate and are resistant to *** regulators of angiogenesis,VEGF-A and matrix metalloproteinases(mmPs)2 and 9 are markers of disease status in *** natural phloroglucinol hyperforin(HF)displays antitumoral properties of potential pharmacological ***,we investigated the effects of HF on mmP-2/9 and VEGF-A expression and survival of primary AmL ***:Blood and bone marrow samples were collected in 45 patients with distinct subtypes defined by French American British classification,i.e.,m0,m1,m2,m3,m4,and *** of mmPs and VEGF-A in leukemic blood cells and culture supernatants were determined by RT-PCR,ELISA,and gelatin zymography(mmPs).The balance between cell death and survival was assessed by flow cytometry with analysis of phosphatidylserine externalization and caspase-3 ***:The administration of HF promoted a caspase-associated apoptosis in primary AmL blasts(from blood and bone marrow),but not normal blood cells and *** addition,HF inhibited the levels of secreted prommP-2,prommP-9,and VEGF-A without altering *** induction of apoptosis by HF significantly paralleled the inhibition of mmP-2/9 and VEGF-A release by *** differences were seen in response to the deleterious effects of HF between AmL cells of distinct ***:Our results suggest that HF,through its proapoptotic and potential antiangiogenic properties(by inhibiting mmP-2/9 and VEGF-A)on primary AmL cells,might be a useful experimental agent,in combination with existing drugs,for new therapeutic approaches in the treatment of this incurable disease.

目前商用原子力显微镜(AFm)大多使用的微悬臂式探针,力灵敏度可达到pN级别。然而受到工艺水平及检测方法限制,微悬臂谐振频率难以超过3.5 mHz,且Q值较低,制约了AFm的成像速度以及在液体中的成像效果。另外,光杠杆的检测方法无法与探针本身进行片上集成,较小的悬臂也给激光束的聚焦带来困难。基于以上考虑,本文提出两种基于mEmS谐振器的探针,振频率可达11 mHz,Q值为4 000,并集成了执行与传感功能以及批量加工纳米针尖的工艺。目前两种探针都已经实现对树脂图案的成像功能,力灵敏度最高可达5pN/√Hz。

为了测量高覆盖条件下(LAI=5)玉米冠层的方向孔隙率,利用一台基于工业用吊车平台的窄视角热红外成像仪开展测量。在此基础上建立了一个几何光学和辐射传输混合模型模拟半球空间的孔隙率分布。测量结果表明,大多数的孔隙出现在邻近的植株行之间,这导致了在热红外图像中,冠层还保持着垄行结构的特征。基于已经得到的几何参数,本研究发展的模型具有垄行作物孔隙率分布特征,模拟的孔隙率与被观测的孔隙率保持很好的一致。更进一步的改进,需通过计算机模拟以建立更加真实的作物结构描述,这也是以后的一个研究方向。

基于热红外成像仪获取的玉米冠层图像,对垄行结构玉米的方向亮温(DBT)进行模型化描述并开展了初步验证工作。模型中假设某一方向上的冠层DBT是组分亮温及各组分在视场中所占面积权重的函数,它们在视场中的比例依赖太阳与传感器的几何位置关系,以及在作物行内,作物行与行之间孔隙的分布。对于玉米冠层的几何特征,简化为横截面是矩形的、其中有空隙透光的一组无限长的平行立方体;立方体内双向孔隙率的方向变化由Kuusk函数来描述。模型模拟表明,玉米亮温组分在视场中的权重具有垄行特性。中午前后,通过对中等密度的冠层DBT模拟,在DBT极坐标图形上发现了一个明显位于垄行方向的热条带的出现,热点出现在太阳位置的周围。最后,利用实地观测的结果与模型模拟结果作对比,对该模型的不足和以后的改进作了初步分析。

c-Fos overexpression increases the proliferation of human hepatocytes by stabilizing nuclear Cyclin D1

AIm: To investigate the effect of stable c-Fos overexpression on immortalized human hepatocyte (IHH) proliferation. mETHODS: IHHs stably transfected with c-Fos (IHH-Fos) or an empty vector (IHH-C) were grown in medium supplemented with 1% serum or stimulated with 10% serum. Cell proliferation was assessed by cell counts, 3H-thymidine uptake and fl ow cytometry analyses. The levels of cell cycle regulatory proteins (Cyclin D1, E, A) cyclin dependent kinases (cdk) cdk2, cdk4, cdk6, and their inhibitors p15, p16, p21, p27, to- tal and phosphorylated GSK-3β and epidermal growth factor receptor (EGF-R) were assayed by Western blot- ting. Analysis of Cyclin D1 mRNA levels was performed by reverse transcription-polymerase chain reaction and real-time polymerase chain reaction (PCR) analysis. Stability of Cyclin D1 was studied by cycloheximide blockade experiments. RESULTS: Stable c-Fos overexpression increased cell proliferation under low serum conditions and resulted in a two-fold increase in [3H]-thymidine incorpora- tion following serum addition. Cell cycle analysis by fl ow cytometry showed that c-Fos accelerated the cell cycle kinetics. Following serum stimulation, Cyclin D1 was more abundantly expressed in c-Fos overexpressing cells. Cyclin D1 accumulation did not result from increased transcriptional activation, but from nuclear stabilization. Overexpression of c-Fos correlated with higher nuclear levels of inactive phosphorylated GSK- 3β, a kinase involved in Cyclin D1 degradation and higher levels of EGF-R mRNA, and EGF-R protein compared to IHH-C both in serum starved, and in serum stimulated cells. Abrogation of EGF-R signalling in IHH- Fos by treatment with AG1478, a specif ic EGF-R tyrosine kinase inhibitor, prevented the phosphorylation of GSK-3β induced by serum stimulation and decreased Cyclin D1 stability in the nucleus. 为了提高玉米冠层半球方向亮温(DBT)测量的准确性,对一套由工业用起重机平台和热红外成像仪组成的地面观测系统的四种测量方法进行了对比和评价。这些方法基于不同的测量原理:(1)通过移动起重机和平台,从不同的角度去测量一个样区的DBT;(2)固定起重机和平台的位置,通过旋转热像仪,从不同的区域收集不同角度的DBT;(3)对方法2的改进,转动热像仪朝向的同时,吊车沿作物垄行方向运动,得到大区域内平均的DBT;(4)当热像仪沿着垂直于垄行方向的起重机臂运动时,用高成像频率对目标热辐射进行采样。在对这些方法进行比较时,着重考虑了玉米冠层亮温分布的时间和空间变化所产生的影响,对本观测系统的优点和局限性进行了初步探讨。最后,本文从实际应用的角度,提出了一个基于这个系统且优化了的DBT测量方法。

Physico-Chemical and Thermal Characterization of a Lignocellulosic Fiber, Extracted from the Bast of <i>Cola lepidota</i>Stem

In this research work, fiber extracted from the bark of Cola lepidota (CL) plant, grown in the flora of Southern part of Cameroon, was investigated for composites reinforcement. The investigation was carried via evaluation of water absorption capacity, moisture content, real density, porosity, chemical composition, chemical structure and thermal behaviour. It was discovered that the new fiber has relatively low moisture content and water absorption capacity similar to those of other investigated natural fibers such as flax, sisal, coconut, hemp and jute. Its porosity was found appropriate for composite production and the fiber was found to be thermally stable up to 230°C, with maximum degradation temperature of 325°C. The main constituents of the fibre include cellulose, hemicellulose and lignin. In conclusion, based on the properties investigated, this fiber is considered suitable for composite manufacture.
