Comparative transcriptomics reveals hidden issues in the plant response to arthropod herbivores

Plants experience different abiotic/biotic stresses,which trigger their molecular machinery to cope with them. Besides general mechanisms prompted by many stresses, specific mechanisms have been introduced to optimize the response to individual threats. However, these key mechanisms are difficult to identify. Here, we introduce an in-depth species-specific transcriptomic analysis and conduct an extensive meta-analysis of the responses to related species to gain more knowledge about plant responses. The spider mite Tetranychus urticae was used as the individual species, several arthropod herbivores as the related species for meta-analysis, and Arabidopsis thaliana plants as the common host. The analysis of the transcriptomic data showed typical common responses to herbivory, such as jasmonate signaling or glucosinolate biosynthesis. Also, a specific set of genes likely involved in the particularities of the Arabidopsis-spider mite interaction was discovered. The new findings have determined a prominent role in this interaction of the jasmonateinduced pathways leading to the biosynthesis of anthocyanins and tocopherols. Therefore, tandem individual/general transcriptomic profiling has been revealed as an effective method to identify novel relevant processes and specificities in the plant response to environmental stresses.

OPTImAL mODE OF SEDImENT TRANSPORT BY SHALLOW FLOWS IN UPLAND AREAS

Soil erosion and sediment movement are serious problems of upland areas of a watershed. In these areas,most of the eroded sediment is transported in overland flows; either in shallow or in concentrated flowsituations. These flows are usually associated with nonlinear waves, generally known as "roll waves" inthe hydraulic engineering literature. Under suitable hydraulic conditions, these waves contain a significantfraction of the total kinetic energy of the flow regime. Hence, the waves act as the prim.ry energy sourcein transporting eroded sediment in shallow flows on steep slopes. Our laborat0ry experiments undercontrolled condition suggest that modulation of the roll waves is effected by the addition of increasingamouflts of sediment in the flow. The optimal condition in the sediment transport rate is reached whenmost of the available wave energy is consumed in carrying the sediment. A related set of experiments ongravitational flows of dry granular material show that the evolving scales of sediment waves are intrinsicto the transport mechanism of the solid phase.

Context: Reduced intake of saturated fat is widely recommended for prevention of cardiovascular disease. The type of macronutrient that should replace saturated fat remains uncertain. Objective: To compare the effects of 3 healthful diets, each with reduced saturated fat intake, on blood pressure and serum lipids. Design, Setting, and Participants: Randomized, 3-period, cross-over feeding study(April 2003 to june 2005)conducted in Baltimore, md, and Boston, mass. Participants were 164 adults with prehypertension or stage 1 hypertension. Each feeding period lasted 6 weeks and body weight was kept constant. Interventions: A diet rich in carbohydrates; a diet rich in protein, about half from plant sources; and a diet rich in unsaturated fat, predominantly monounsaturated fat. main Outcome measures: Systolic blood pressure and low-density lipoprotein cholesterol. Res ults: Blood pressure, low-density lipoprotein cholesterol, and estimated corona ry heart disease risk were lower on each diet compared with baseline. Compared w ith the carbohydrate diet, the protein diet further decreased mean systolic bloo d pressure by 1.4 mm Hg(P=.002)and by 3.5 mm Hg(P=.006)among those with hyperten sion and decreased low-density lipoprotein cholesterol by 3.3 mg/dL(0.09 mmol/L ; P=.01), high-density lipoprotein cholesterol by 1.3 mg/dL(0.03 mmol/L; P=.02) , and triglycerides by 15.7 mg/dL(0.18 mmol/L; Pmpared with the carbo hydrate diet, the unsaturated fat diet decreased systolic blood pressure by 1.3 mm Hg(P=.005)and by 2.9 mm Hg among those with hypertension(P=.02), had no signi ficant effect on low-density lipoprotein cholesterol, increased high-density l ipoprotein cholesterol by 1.1 mg/dL(0.03 mmol/L; P=.03), and lowered triglycerid es by 9.6 mg/dL(0.11 mmol/L; P=.02). Background: Gastroesophageal reflux is a frequent problem after esophageal atr esia (EA) repair. Our aim was to determine the prevalence of esophagitis and Bar rett esophagus more than 10 years after repair of EA. methods: Ninety-two patie nts treated between 1973 and 1985 were included in this prospective study. A que stionnaire was completed by 86 patients; esophagogastroscopy was performed in 49 patients. Results: Only 36 patients had no complaints at all. Thirty-one patie nts complained of difficulties swallowing solid food; 23 complained of heartburn . Esophagogastroscopy revealed grade 3 esophagitis in 2 patients and a macroscop ic image of Barrett esophagus in 2. Histology showed esophagitis in 30 patients, gastric metaplasia in 3, and no intestinal metaplasia (Barrett esophagus). Conc lusions: For epidemiologic reasons, that is, the short interval of follow-up (1 0 years) and the low compliance of the study group, larger numbers are needed to decide if routine long-term endoscopic screening after repair of EA is necessa ry. For now, it cannot yet be recommended. The prevalence of symptoms of gastroe sophageal reflux disease in this study group is higher than that in the general population, but we found no severe complications of gastroesophageal reflux in t he pediatric age group.

PURPOSE. To determine the central cornea thickness (CCT) in Singaporean children and to examine the possible relationship between intraocular pressure (IOP) and other biometric factors and CCT. mETHODS. This was a cross- sectional study. The subjects (n=652) were obtained from the Singapore Cohort Study of the Risk Factors for myopia (SCORm). The subjects’ ages ranged from 9 to 11 years. There were 485 Chinese, 92 malayan, and 75 Asian Indian children. measurement procedures included air- puff tonometry, noncontact slit lamp optical pachymet- ry, cycloplegic autorefraction, and autokeratometry. RESULTS. The mean CCT was 543.6± 32.0 μ m. Chinese children had thicker corneas than malayan or Indian children (P=0.002). The boys had thicker corneas than girls (P=0.011), but the mean difference was only 6.4 μ m. There was high correlation of CCT (r=0.98) and IOP (r=0.88) between right and left eyes. IOP was correlated with CCT (r=0.45, P multiple linear regression model, each millimeter of mercury of IOP was associated with a CCT difference of 5.90 μ m (95% confidence interval [CIAbstract, 4.98- 6.82). The radius of corneal curvature correlated with CCT (r=0.19, P meters were not significantly (P > 0.05) associated with CCT: age, family income, father s education, axial length, and spherical equivalent. CONCLUSIONS. The mean CCT in Singaporean children aged 9 to 11 years was 543.6 μ m and showed ethnic and gender variation. CCT affected measured IOP and correlated weakly with corneal curvature. Compared with data in adults, a change in CCT was associated with a greater difference in measured IOP.

Asymmetric (ADmA) and symmetric dimethylarginine (SDmA) inhibit production of nitric oxide. The concentration of both dimethylarginines is regulated by urina ry excretion, although ADmA, but not SDmA, is also subject to degradation by dim ethylarginine dimethylaminohydrolase, which is highly expressed in the liver but also present in the kidney. The exact roles of the human liver and kidney in th e metabolism of dimethylarginines are currently unknown. Therefore, we aimed to investigate renal and hepatic handling of ADmA and SDmA in detail in 24 patients undergoing hepatic surgery. To calculate net organ fluxes and fractional extrac tion (FE) rates, blood was collected from an arterial line, the portal vein, hep atic vein, and renal vein, and blood flow of the hepatic artery, portal vein, an d renal vein was determined using Doppler ultrasound techniques. Results showed a significant net uptake (median [IQR]) of ADmA in both the liver (9.6 nmol/min [5.6- 13.2] ) and the kidney (12.1 nmol/min [1.3- 17.1]). SDmA uptake was pres ent not only in the kidney (12.7 nmol/min [3.5- 25.4]), but also in the liver ( 7.7 nmol/min [2.8- 16.4]). FE rates of ADmA for the liver and kidney were 5.0% (3.5% - 7.4% ) and 8.4% (1.3% - 13.9% ), respectively. For SDmA, hepat ic and renal FE rates were 3.4% (2.1% - 7.5% ) and 12.5% (3.6% - 16.2% ), respectively. In conclusion, this study gives a detailed description of the hepatic and renal elimination of dimethylarginines and shows that the clearing o f SDmA is not only confined to the kidney, but the human liver also takes up sub stantial amounts of SDmA from the portal and systemic circulation.

A well-established population of Burmese pythons resides in the Everglades of southern *** in part by a report that identified much of southern USA as suitable habitat for expansion or establishment of the Burmese python,we examined the plausibility of this snake to survive winters at sites north of the *** integrated daily low and high temperatures recorded from October to February from 2005-2011 at Homestead,Orlando and Gainesville,Florida;and Aiken,South Carolina,with minimum temperatures projected for python digestion(16℃),activity(5℃)and survival(0℃).mean low and high temperatures decreased northward from Homestead to Aiken and the number of days of freezing temperatures increased *** was impaired or inhibited for 2 months in the Everglades and up to at least 5 months in Aiken,and activity was increasingly limited northward during these *** of overwinter survivorship document that a single bout of low and freezing temperatures results in python *** capacity for Burmese pythons to successfully overwinter in more temperate regions of the USA is seemingly prohibited because they lack the behaviors to seek refuge from,and the physiology to tolerate,cold *** tropical Southeast Asia is the source of the Everglades Burmese pythons,we predict it is unlikely that they will be able to successfully expand to or colonize more temperate areas of Florida and adjoining states due to their lack of behavioral and physiological traits to seek refuge from cold temperatures.

The Gynecologic Oncology Group (GOG) has studied a number of drugs to determin e their activity in patients with previously treated squamous and nonsquamous ca ncer arising in the uterine cervix. A Phase II study with intravenous vinorelbin e was initiated for this purpose in patients with Stage IVB, recurrent, or persi stent nonsquamous carcinomas who had received one prior chemotherapy or were not eligible for other studies. Eligible patients had to have measurable disease, G OG performance status of 0-2 and adequate bone marrow, liver, and renal functio n. The treatment consisted of vinorelbine 30 mg/m2 on days 1 and 8, repeated eve ry 21 days. Tumor measurements and toxicities were recorded every treatment cycl e. Thirty patients were enrolled with 28 patients deemed eligible and evaluable. Only two confirmed partial responses (7.1%) were noted. Neutropenia was the mo st common toxicity with 9 of 28 (32%) experiencing either grade 3 or 4 changes. Anemia was severe in seven. Neuropathy was more than mild in three patients. Se vere events, such as fatigue, hypertension, or pulmonary changes attributed to d rug administration, occurred only in one or two instances. With the dose schedul e and assessment criteria employed, vinorelbine had only minimal activity in non squamous cancer of the cervix.
