Personalized nutrition to mitigate inflammation in genetically predisposed individuals: a secondary analysis of the Danish PREVENTomICS intervention

Deep phenotyping and genetic characterization of individuals are fundamental to assessing the metabolic status and determining nutrition-specific requirements. This study aimed to ascertain the utmost effectiveness of personalized interventions by aligning dietary adjustments with both the genotype and metabolotype of individuals. Therefore， we assessed here the usefulness of a polygenic score (PGS) characterizing a potential pro-inflammatory profile (PGSi) as a nutrigenetic tool to discern individuals from the Danish PREVENTomICS cohort that could better respond to precision nutrition (PN) plans， specifically targeted at counteracting the low-grade inflammatory profile typically found in obesity. The cohort followed a PN plan to counteract the pro-inflammatory profile (PNi group) or generic dietary recommendations (Control) for 10 weeks. PGSi was applied for genetic stratification (Low/High). The effects of the intervention on anthropometrics and biomarkers related to inflammatory profile and carbohydrate metabolism were assessed. Around 30% of subjects had a high genetic predisposition to pro-inflammatory status (high-PGSi). These individuals demonstrated the most effective response to the dietary plan， experiencing improved body composition， with significant decreases in body weight (△: -4.84%; p=0.039) and body fat (△: -4.86%; p=0.007)， and beneficial changes in pro- and anti-inflammatory biomarkers， with significant increases in IL-10 (△: 71.3%; p=0.025) and decreases in TNF-α (△: -3.0%; p=0.048)， CRP (△: -31.1%)， ICAm1 (△: -5.8%)， and mCP1 (△: -4.2%) circulating levels， compared to low-PGSi individuals. Both phenotypic and genetic stratification contributed to a better understanding of metabolic heterogeneity in response to diet. This approach allows for refinement of the prediction of individual requirements and potentially for better management of obesity.

Abstract:This paper addresses the problem of improving the optimal value of the maximum Capacity Path(mCP)through expansion in a flexible network,and minimizing the involved *** only condition applied to the cost functions is to be non-decreasing *** is a non-restrictive condition,reflecting the reality in practice,and is considered for the first time in the ***,the total cost of expansion is a combination of max-type cost(e.g.,for supervision)and sum-type cost(*** building infrastructures,price of materials,price of labor,etc.).For this purpose,two types of strategies are combined:(l)increasing the capacity of the existing arcs,and(l)adding potential new *** different problems are introduced and *** the problems have immediate applications in Internet routing *** first one is to extend the network,so that the capacity of an mcP in the modified network becomes equal to a prescribed value,therefore the cost of modifications is minimized.A strongly polynomial-time algorithm is deduced to solve this *** second problem is a network expansion under a budget constraint,so that the capacity of an mcP is maximized.A weakly polynomial-time algorithm is presented to deal with *** the special case when all the costs are linear,a meggido's parametric search technique is used to develop an algorithm for solving the problem in strongly polynomial *** new approach has a time complexity of o(n^(4)),which is better than the time complexity of o(n4 log(n)of the previously known method from literature.

Directed x-rays produced in the interaction of sub-picosecond laser pulses of moderate relativistic intensity with plasma of near-critical density are investigated. Synchrotron-like (betatron) radiation occurs in the process of direct laser acceleration (DLA) of electrons in a relativisticlaser channel when the electrons undergo transverse betatron oscillations in self-generated quasi-static electric and magnetic fields. In anexperiment at the PHELIX laser system, high-current directed beams of DLA electrons with a mean energy ten times higher than the ponderomotive potential and maximum energy up to 100 meV were measured at 10^(19) W/cm^(2)laser intensity. The spectrum of directed x-raysin the range of 5–60 keV was evaluated using two sets of ross filters placed at 0°and 10°to the laser pulse propagation axis. The differential x-ray absorption method allowed for absolute measurements of the angular-dependent photon fluence. We report 10^(13) photons/sr withenergies >5 keV measured at 0°to the laser axis and a brilliance of 10^(21) photons s^(−1) mm^(−2) mrad−2(0.1%BW)−1. The angular distributionof the emission has an FWHm of 14°–16°. Thanks to the ultra-high photon fluence, point-like radiation source, and ultra-short emissiontime, DLA-based keV backlighters are promising for various applications in high-energy-density research with kilojoule petawatt-class laserfacilities.

Detrital zircon geochronology and Hf isotope analysis can be used for inferring provenance characteristics,and to evaluate the tectonic evolution of sedimentary basins and their link with regional *** Paleozoic sequences of the okcheon Belt consist of the Lower Paleozoic Joseon and the Upper Paleozoic Pyeongan supergroups with middle Paleozoic hiatus locally on top of the Neoproterozoic bimodal volcanic rocks,reflecting an intracontinental rift setting between the two basements(*** and Yeongnam massifs)at southern part of the Korean *** detrital zircon U-Pb ages and Lu-Hf isotope results show that all these Paleozoic strata commonly have Paleoproterozoic and Paleozoic zircon ages with rare meso-to Neoproterozoic *** individual zircon populations display following features,allowing estimation of their sedimentary provenances:(i)The Paleoproterozoic zircons(ca.1.85 Ga and 2.50 Ga)with similar ranges ofεHf(t)values are most common in the basement rocks of the Korean Peninsula,and were sourced from both the Gyeonggi and Yeongnam massifs.(ii)The meso-to Neoproterozoic zircons,preserved only in the middle to Late Cambrian clastic sedimentary rocks within the carbonate sequences probably reflect proximal provenance.(iii)The youngest Paleozoic zircons of each formation,almost coincident with their deposition ages,suggest presence of syndepositional magmatism,indicating proximal magmatic sources during their deposition.(iv)The Cambrian-ordovician zircons,from the Lower Paleozoic sequences,but rare in the successive Upper Paleozoic sequences,suggest a provenance change after the hiatus between the two sedimentary successions.(v)The Permian zircons showing differentεHf(t)values indicate that detrital sources were varied at that *** integrated results in our study suggest provenance variability linked to diverse tectonic environments,reflecting prolonged subduction-related crustal evolution of the proto-Korean Peninsula during the Paleozo

Tight focusing with very small f-numbers is necessary to achieve the highest at-focus ***,tight focusing imposes strong demands on precise target positioning in-focus to achieve the highest on-target irradiance We describe several near-infrared,visible,ultraviolet and soft and hard X-ray diagnostics employed in a~10^(22)W/cm^(2)laser±plasma *** used nearly 10 J total energy femtosecond laser pulses focused into an approximately1.3-μm focal spot on 5±20μm thick stainless-steel *** discuss the applicability of these diagnostics to determine the best in-focus target position with approximately 5μm accuracy(i.e.,around half of the short Rayleigh length)and show that several diagnostics(in particular,3ωreflection and on-axis hard X-rays)can ensure this *** demonstrated target positioning within several micrometers from the focus,ensuring over 80%of the ideal peak laser intensity *** approach is relatively fast(it requires 10±20 laser shots)and does not rely on the coincidence of low-power and high-power focal planes.
