Genomic dissection of widely planted soybean cultivars leads to a new breeding strategy of crops in the post-genomic era

Soybeans specially the widely planted cultivars have been dramatically improved in agronomic performance and is well adapted to local planting environments after long-time domestication and *** the unique genomic features of popular cultivars will help to understand how soybean genomes have been modified through *** re-sequenced 134 soybean cultivars that were released and most widely planted over the last century in *** analyses established that these cultivars comprise two geographically distinct sub-populations:Northeast China (NE) versus the HuangHuai-Hai River Valley and South China (HS).A total of 309 selective regions were identified as being impacted by geographical *** HS sub-population exhibited higher genetic diversity and linkage disequilibrium decayed more rapidly compared to the NE *** study the association between phenotypic differences and geographical origins,we recorded the vegetative period under different growing conditions for two years,and found that clustering based on the phenotypic data was closely correlated with cultivar geographical *** iteratively calculating accumulated genetic diversity,we established a platform panel of cultivars and have proposed a novel breeding strategy named "Potalaization"for selecting and utilizing the platform cultivars that represent the most genetically diversity and the highest available agronomic performance as the "plateau"for accumulating elite loci and traits,breeding novel widely adapted cultivars,and upgrading breeding *** addition to providing new genomic information for the soybean research community,the "Potalaization"strategy that we devised will also be practical for integrating the conventional and molecular breeding programs of crops in the post-genomic era.

metallicity and star formation activities of the interacting system Arp 86 from observations with mOS on the Xinglong 2.16m telescope

We present an analysis of the metallicity and star formation activities of H II regions in the interacting system Arp 86, based on the first scientific observations using mulri-object spectroscopy with the 2.16 m telescope at the Xinglong Observing Station. We find that the oxygen abundance gradient in Arp 86 is flatter than that in normal disk galaxies, which confirms that gas inflows caused by tidal forces during encounters can flatten the metallicity distributions in galaxies. The companion galaxy NGC 7752 is currently experiencing a galaxy-wide starburst with a higher star for- marion rate surface density than the main galaxy NGC 7753, which can be explained in that the companion galaxy is more susceptible to the effects of interaction than the primary. We also find that the galaxy 2mASX J23470758＋2926531 has similar abun- dance and star formation properties to NGC 7753, and may be a part of the Arp 86 system.

A truncated hepatitis E virus ORF2 protein expressed in tobacco plastids is immunogenic in mice

AIm： To cost-effectively express the 23-ku pE2, the most promising subunit vaccine encoded by the E2 fragment comprising of the 3＇-portion of hepatitis E virus （HEV） open reading frame 2 （ORF2） in plastids of tobacco （Nicotiana tabacum cv. SR1）, to investigate the transgene expression and pE2 accumulation in plastids, and to evaluate the antigenic effect of the plastid-derived pE2 in mice. mETHODS： Plastid-targeting vector pRB94-E2 containing the E2 fragment driven by rice psbA promoter was constructed. Upon delivery into tobacco plastids, this construct could initiate homologous recombination in psaB-trnfm and trnG-psbC fragments in plastid genome, and result in transgene inserted between the two fragments. The pRB94-E2 was delivered with a biolistic particle bombardment method, and the plastid-transformed plants were obtained following the regeneration of the bombarded leaf tissues on a spectinomycin-supplemented medium. Transplastomic status of the regenerated plants was confirmed by PCR and Southern blot analysis, transgene expression was investigated by Northern blot analysis, and accumulation of pE2 was measured by ELISA. Furthermore, protein extracts were used to immunize mice, and the presence of the pE2-reactive antibodies in serum samples of the immunized mice was studied by ELISA. RESULTS： Transplastomic lines confirmed by PCR and Southern blot analysis could actively transcribe the E2 mRNA. The pE2 polypeptide was accumulated to a level as high as 13.27 μg/g fresh leaves. The pE2 could stimulate the immunized mice to generate pE2-specific antibodies. CONCLUSION： HEV-E2 fragment can be inserted into the plastid genome and the recombinant pE2 antigen derived is antigenic in mice. Hence, plastids may be a novel source for cost-effective production of HEV vaccines.

Implantable vascular devices are widely used in clinical treatments for various vascular diseases. However, current approved clinical implantable vascular devices generally have high failure rates primarily due to their surface lacking inherent functional endothelium. Here, inspired by the pathological mechanisms of vascular device failure and physiological functions of native endothelium, we developed a new generation of bioactive parylene (poly(p-xylylene))-based conformal coating to address these challenges of the vascular devices. This coating used a polyethylene glycol (PEG) linker to introduce an endothelial progenitor cell (EPC) specific binding ligand LXW7 (cGRGDdvc) onto the vascular devices for preventing platelet adhesion and selectively capturing endogenous EPCs. Also, we confirmed the long-term stability and function of this coating in human serum. Using two vascular disease-related large animal models, a porcine carotid artery interposition model and a porcine carotid artery-jugular vein arteriovenous graft model, we demonstrated that this coating enabled rapid generation of self-renewable “living” endothelium on the blood contacting surface of the expanded polytetrafluoroethylene (ePTFE) grafts after implantation. We expect this easy-to-apply conformal coating will present a promising avenue to engineer surface properties of “off-the-shelf” implantable vascular devices for long-lasting performance in the clinical settings.

＜正＞SUmmARY This study has examined the factors which appear to be related,to the auditors’ decisions of issuing different types of modified opinions（explanatory paragraph,except for qualifications and disclaimers） in the Hong Kong *** results,based on a multivariate logit model,are consistent with the conceptual arguments presented in the earlier literature that auditors’ decisions relating to going concern modifications can be predicted satisfactorily using financial,contrary,return and qualification ***,firms associated with different opinion types differ systematically on their financial condition, nature of their contrary information,internal control problems reported,and stock *** results are consistent with the notion that the different types of modified opinion serve as signals of varying severity of the going concern problems faced by the *** results have implications for audit practices in the United States and around the world.

In pulsar timing, timing residuals are the differences between the observed times of arrival and predictions from the timing model. A comprehensive timing model will produce featureless resid- uals, which are presumably composed of dominating noise and weak physical effects excluded from the timing model （e.g. gravitational waves）. In order to apply optimal statistical methods for detecting weak gravitational wave signals, we need to know the statistical properties of noise components in the residuals. In this paper we utilize a variety of non-parametric statistical tests to analyze the whiteness and Gaussianity of the North American Nanohertz Observatory for Gravitational Waves （NANOGrav） 5- year timing data, which are obtained from Arecibo Observatory and Green Bank Telescope from 2005 to 2010. We find that most of the data are consistent with white noise; many data deviate from Gaussianity at different levels, nevertheless, removing outliers in some pulsars will mitigate the deviations.
