Chalcone derivatives are of immense importance owing to their superior pharmacological profile. minor changes in their structures create high degree of diversity that results in a broader spectrum of biological activities. The present research paper presents the synthesis of 3-（4-chloro-3-nitrophenyl）-1-phenylprop-2-en-1-one chalcone（I）, single crystal XRD analysis and the evaluation of its possible biological activities like cytotoxic（LD（50） = 5.40 ug/m L）and antileishmanial（LD（50） = 0.81 ug/m L） activities. The results indicate the tested chalcone（I）shows superior inhibition values against leishmanial promastigotes. Further, the possible interaction sites of chalcone（I） with Try R enzyme involved in the redox metabolism of the leishmanial parasite were determined using docking simulation technique. Docking computations demonstrate that the compound establishes prominent binding interactions with the key residues of Try R and possesses the potential to effectively inhibit the catalytic activities of enzyme. Thus the results suggest that the chalcone（I） can serve as a potential scaffold for the treatment of leishmaniasis and deserve further development.

In Vitro Efficacy Assessment of Botanical Extracts against Botrytis gladiolorum Causing Gladiolus Leaf Blight

An experiment was conducted at the mycology laboratory, Department of Plant Pathology, Sher-e-Bangla Agricultural University, Dhaka, Bangladesh to find out the efficacy of selected botanical extracts against Botrytis gladiolorum (Bg) causing gladiolus leaf blight. Infected leaves samples were collected from gladiolus farmers’ field and brought to the laboratory for study. Ten selected botanicals were used against the colony growth of (Bg). The botanicals were mehendi, Chrysanthemum, Basil (Tulsi), Onion, Neem, Bael, Arjuna, Garlic, Aloevera (Ghritkumary) and Turmeric. Botanical extracts were applied at the rate of 5%, 10% and 20%. The radial mycelia growth was found minimum (11.60 mm) in garlic extract treated plate at the dose of 5% at 5 DAI, which was statistically similar with turmeric extract treated (14.00 mm) plate and the inhibition of growth was 60% and 51.72%, respectively. Similar trend was found at 10 DAI and 15 DAI. At 15 DAI, garlic and turmeric extract gave the best result against Bg, which was statistically similar with onion (50.07% inhibition) and mehendi (49.93%). All botanicals showed significantly different results over control and found effective in reducing the mycelial growth at the dose of 10%. At 5 DAI, no radial mycelia growth was found in garlic treated plate, which was statistically similar with onion treated plate, means that the inhibition of growth was 100%. Similar trend was also found at 10 DAI, and 15 DAI but at 15 DAI, onion (30.20 mm) gave the statistically similar results with Garlic (30.10 mm) and the mycelia growth inhibition was (57.70%) and (57.84%), respectively. In case of 20% dose, garlic extract showed the best result at 5 DAI and the radial mycelia growth was found minimum (00.00 mm) which was statistically similar with onion extract treated (00.00 mm) plate and turmeric extract treated plate. DFT-Based Chemical Reactivity Descriptors, Pharmacokinetics and molecular Docking Studies of Thymidine Derivatives

Thymidine-containing derivatives are considered to be among the most significant derivatives in medicinal chemistry. In this study, we employed a combined computational approach involving density-functional theory (DFT) calculations, molecular docking simulations, and absorption, distribution, metabolism, excretion, and toxicity (ADmET) property predictions. Prediction of activity spectra for substances (PASS) revealed promising antiviral, antimicrobial and anti-carcinogenic activities of these thymidine derivatives. Using Gaussian 09, we optimized the molecular structures of the thymidine derivatives to obtain their stable conformations and calculate their electronic properties. Subsequently, molecular docking simulations were performed to explore the binding interactions between the thymidine derivatives and the active site of the Candida albicans (PDB: 1IYL and 2Y7L) proteins. The docking results were evaluated based on docking scores, hydrogen bonding, and hydrophobic interactions and revealed favorable binding interactions between the thymidine derivatives and the proteins, suggesting their potential as antifungal agents. The thermodynamic properties, including binding free energy, enthalpy, and entropy changes were determined to assess the stability and strength of the ligands-protein complexes. The calculated pharmacokinetic parameters, such as ADmET properties, provided insights into the drug-likeness and potential bioavailability of the thymidine derivatives. These results offer a foundation for further experimental investigations and the design of novel antifungal agents targeting Candida albicans infections.

Conjugate Effect of Radiation and Thermal Conductivity Variation on mHD Free Convection Flow for a Vertical Plate

A numerical investigation is performed to study the effect of thermal radiation on magnetohydrodynamic (mHD) free convection flow along a vertical flat plate in presence of variable thermal conductivity in this paper. The governing equations of the flow and the boundary conditions are transformed into dimensionless form using appropriate similarity transformations and then solved employing the implicit finite difference method with Keller-box scheme. Results for the details of the velocity profiles, temperature distributions as well as the skin friction, the rate of heat transfer and surface temperature distributions are shown graphically. Results reveal that the thermal radiation is more significant in mHD natural convection flow during thermal conductivity effect is considered. To illustrate the accuracy of the present results, the results for the local skin fraction and surface temperature distribution excluding the extension effects are compared with results of merkin and Pop designed for the fixed value of Prandtl number and a good agreement were found.

Hepatocytes isolated from neoplastic liver-immunomagnetic purging as a new source for transplantation

AIm: To investigate whether hepatocytes isolated from macroscopically normal liver during hepatic resection for neoplasia could provide a novel source of healthy hepatocytes, including the development of reliable protocols for malignant cells removal from the hepatocyte preparation. mETHODS: Hepatocytes were procured from resected liver of 18 patients with liver tumors using optimised digestion and cell-enrichment protocols. Suspensions of various known quantities of the HT-29 tumor cell line and patient hepatocytes were treated or not with Ep-CAm-antibody-coated immunomagnetic beads in order to investigate the effi cacy of tumor-purging by immunomagnetic depletion, using a semi-quantitative RT-PCR method developed to detect tumor cells. Immunomagnetic bead-treated or bead-untreated tumor cell-hepatocyte suspensions were transplanted intra-peritoneally in Balb/C nude mice to assess the rates of tumor development. RESULTS: mean viable hepatocyte yield was 9.3 x 106 cells per gram of digested liver with mean viability of 70.5%. Immunomagnetic depletion removed tumor cells to below the RT-PCR detection-threshold of 1 tumor cell in 106 hepatocytes, representing a maximum tumor purging efficacy of greater than 400 000-fold. Transplanted, immunomagnetic bead-purged tumor cell-hepatocyte suspensions did not form peritonealtumors in Balb/C nude mice. Co-transplantation of hepatocytes with tumor cells did not increase tumorigenesis of the tumor cells. CONCLUSION: Immunomagnetic depletion appears to be an effective method of purging contaminating tumor cells to below threshold for likely tumorigenesis. Along with improved techniques for isolation of large numbers of viable hepatocytes, normal liver resected for neoplasia has potential as another clinically useful source of hepatocytes

This study presents the intercomparison of the outdoor environmental gamma dose rates measured using a Nal （TI） based survey meter along with thermoluminescent dosimeters （TLDs） and estimation of excess lifetime cancer risk （ELCR）, for the inhabitants of Poonch division of the Azad Kashmir, Pakistan. Genetic Architecture of Yield in Bottle Gourd (<i>Lagenaria siceraria</i>(mol.) Standl.)

The genetic architecture of yield was studied in a five parent half diallel cross of bottle gourd at the experimental field of Olericulture Division, Horticulture Research Centre (HRC), Bangladesh Agricultural Research Institute (BARI), Gazipur during the winter season of 2016-17. The values of mean square for GCA (general combining ability) and SCA (specific combining ability) were highly significant which suggested the presence of both additive and non-additive genetic variance in the population. But the higher magnitude of GCA compared to SCA indicated predominance of additive genetic variance. In most of the cases, the cross between poor and poor parents showed positive SCA effect for fruit yield, which indicated the higher yield. The estimates of mid parent heterosis ranged from −19.0 to 31.8 percent and the better parent heterosis ranged from −28.4 to 20.5 percent. Analysis for genetic components of variation suggested that additive components were more important in the inheritance of fruit yield. This character was observed being controlled by two to three pairs of genes or groups of genes. Narrow sense heritability was 55 percent indicating probability of selection in generations. The graphical analysis also indicated wide genetic diversity among the parents.
