Region-dependent effects of diabetes and insulin-replacement on neuronal nitric oxide synthase-and heme oxygenase-immunoreactive submucous neurons

AIm To investigate the intestinal segment-specific effects of diabetes and insulin replacement on the density of different subpopulations of submucous neurons. mETHODS Ten weeks after the onset of type 1 diabetes samples were taken from the duodenum, ileum and colon of streptozotocin-induce diabetic, insulin-treated diabetic and sex-and age-matched control rats. Whole-mount preparations of submucous plexus were prepared from the different gut segments for quantitative fluorescent immunohistochemistry. The following double-immunostainings were performed: neuronal nitric oxide synthase(n NOS) and Hu C/D, heme oxygenase(HO) 1 and peripherin, as well as HO2 and peripherin. The density of n NOS-, HO1-and HO2-immunoreactive(IR) neurons was determined as a percentage of the total number of submucous neurons. RESULTS The total number of submucous neurons and the proportion of n NOS-, HO1-and HO2-IR subpopulations were not affected in the duodenal ganglia of control, diabetic and insulin-treated rats. While the total neuronal number did not change in either the ileum or the colon, the density of nitrergic neurons exhibited a 2-and 3-fold increase in the diabetic ileum and colon, respectively, which was further enhanced after insulin replacement. The presence of HO1-and HO2-IR submucous neurons was robust in the colon of controls(38.4%-50.8%), whereas it was significantly lower in the small intestinal segments(0.0%-4.2%, P m and a decrease in the colon of the proportion of HO-IR neurons in insulin-treated diabetic animals. CONCLUSION Diabetes and immediate insulin replacement induce the most pronounced region-specific alterations of n NOS-, HO1-and HO2-IR submucous neuronal density in the distal parts of the gut.

Tensile and fracture properties of an mg-RE-Zn alloy at elevated temperatures

magnesium alloy EZ10 （mg-RE-Zn） was deformed in tension at temperatures from 20 up to 520℃. A rapid decrease of the yield and tensile strength with temperature was observed at temperatures higher than 300 ℃. On the other hand, ductility of sam-ples rapidly increased in the same temperature range. Light microscopy and scanning electron microscopy was used to reveal the rea-son for these behaviours. Intermetallic particles in grain boundaries are responsible for excellent mechanical properties at lower tem-peratures. Diffusional processes occurring at temperatures higher than 300 ℃ significantly influenced the deformation mechanism as well as the fracture character.

Characterization of the High-Strength mg–3Nd–0.5Zn Alloy Prepared by Thermomechanical Processing

magnesium alloys based on Nd and Zn are promising materials for both aviation industry and medical *** mechanical properties of these materials can be achieved by thermomechanical processing such as extrusion or rolling and by aging treatment, which can significantly strengthen the alloy. The question remains especially about the connection of texture strength created in the alloys based on the specific conditions of preparation. This work focuses on the mg–3 Nd–0.5 Zn magnesium alloy prepared by hot extrusion of the as-cast state at two different temperatures combined with heat pre-treatment. Extrusion ratio of 16 and rate of 0.2 mm/s at 350 and 400 °C were selected for material preparation. The structures of prepared materials were studied by scanning electron microscopy and transmission electron microscopy. The effect of microstructure on mechanical properties was evaluated. Obtained results revealed the strong effect of thermal pre-treatment on final microstructure and mechanical properties of extruded materials. The Hall–Petch relation between grain size and tensile yield strength has been suggested in this paper based on the literature review and presented data. The observed behavior strongly supports the fact that the Hall–Petch of extruded mg–3Nd–0.5 Zn alloys with different texture intensities cannot be clearly estimated and predicted. In addition, Hall–Petch relations presented in literature can be sufficiently obtained only for fraction of the mg–3Nd–0.5 Zn alloys.

Copper(I)selenide-nanocrystalline semiconductor was synthesized via one-step mechanochemical synthesis after 5 min milling in a planetary ball *** kinetics of synthesis was followed by X-ray powder diffraction analysis and specific surface area measurements of milled 2Cu/Se *** X-ray diffraction confirmed the orthorhombic crystal structure of Cu_(2)Se with the crystallite size∼25 *** surface chemical structure was studied by X-ray photoelectron spectroscopy,whereby the binding energy of the Cu 2p and Se 3d signals corresponded to Cu^(+)and Se^(2−)oxidation *** electron microscopy revealed agglomerated nanocrystals and confirmed their orthorhombic structure,as *** optical properties were studied utilizing ultraviolet-visible spectroscopy and photoluminescence *** direct bandgap energy 3.7 eV indicated a blue-shift phenomenon due to the quantum size *** type of Cu_(2)Se synthesis can be easily adapted to production dimensions using an industrial vibratory *** advantages of mechanochemical synthesis represent the potential for inexpensive,environmentally-friendly,and waste-free manufacturing of Cu_(2)Se.

The potential application of red mud and soil mixture as additive to the surface layer of a landfill cover system

Red mud, the by-product of aluminum production, has been regarded as a problematic residue all over the world. Its storage involves risks as evidenced by the Ajka red mud spill,an accident in Hungary where the slurry broke free, flooding the surrounding areas. As an immediate remediation measure more than 5 cm thick red mud layer was removed from the flooded soil surface. The removed red mud and soil mixture（RmSm） was transferred into the reservoirs for storage. In this paper the application of RmSm is evaluated in a field study aiming at re-utilizing waste, decreasing cost of waste disposal and providing a value-added product. The purpose was to investigate the applicability of RmSm as surface layer component of landfill cover systems. The field study was carried out in two steps： in lysimeters and in field plots. The RmSm was mixed at ratios ranging between 0 and 50% w/w with low quality subsoil（LQS） originally used as surface layer of an interim landfill cover. The characteristics of the LQS ＋ RmSm mixtures compared to the subsoil（LQS） and the RmSm were determined by physical–chemical, biological and ecotoxicological methods. The addition of RmSm to the subsoil（LQS） at up to 20% did not result any ecotoxic effect, but it increased the water holding capacity. In addition, the microbial substrate utilization became about triple of subsoil（LQS） after 10 months. According to our results the RmSm mixed into subsoil（LQS） at20% w/w dose may be applied as surface layer of landfill cover systems.

The paper comprises new analytical data on the nature and occurrence of gold in solid pyrobitumen,closely associated with the main gold-bearing sulfide arsenic ores of the Bakyrchik gold deposit(Kazakhstan),related to post-collisional magmatic-hydrothermal *** mineralization of the deposit occurs mainly in the form of an“invisible”type of gold in the structures of arsenian pyrite and arsenopyrite,and the form of gold-organic compounds of pyrobitumen in carbonaceous-terrigenous sequences of Carboniferous *** and electron microscopic analysis,Raman and FT-Infrared analysis,mineralogical and three-step sequential extraction analysis(NH2OHHCl,H2O2,HNO3?HCl)has been carried out using 9 ore samples(from 3 different types of ores)for a comprehensive study of pyrobitumen and sulfide arsenic ores focusing mainly on organic *** sequentially extracted precious metal content of pyrobitumen reaches up to 7 ppm gold and other metals like Ag 4 ppm,Pt 31 ppb,and Pd 26 ppb,forming metal–organic compounds,while arsenic sulfide minerals incorporate 11 ppm gold,39 ppm Ag,0.49 ppm *** enrichment of gold associating with organic matter and sulfide ore minerals was confirmed in this *** matter was active in the migration of gold and the capture of gold by ***,the reductive organic matter agent released gold,most likely for the sulfide arsenic ore *** was a decisive factor in the concentration,transportation,and preservation of gold in the deposit.

We present new large time step methods for the shallow water flows in the lowFroude number *** order to take into accountmultiscale phenomena that typically appear in geophysical flows nonlinear fluxes are split into a linear part governing the gravitational waves and the nonlinear *** propose to approximate fast linear waves implicitly in time and in space bymeans of a genuinely multidimensional evolution *** the other hand,we approximate nonlinear advection part explicitly in time and in space bymeans of themethod of characteristics or some standard numerical flux *** integration is realized by the implicit-explicit(ImEX)*** apply the ImEX Euler scheme,two step Runge Kutta Cranck Nicolson scheme,as well as the semi-implicit BDF scheme and prove their asymptotic preserving property in the low Froude number *** experiments demonstrate stability,accuracy and robustness of these new large time step finite volume schemes with respect to small Froude number.

Insights into the role of jasmonic acid-mediated defenses against necrotrophic and biotrophic fungal pathogens

Jasmonic acid （JA） is a natural hormone regulator involved in development, responses against woundingand pathogen attack. Upon perception of pathogens, JA is synthesized and mediates a signaling cascade initiating various defense responses. Traditionally, necrotrophic fungi have been shown to be the primary activators of JA- dependent defenses through the JA-receptor, COIl. Conversely, plants infected with biotrophic fungi have classically been associated with suppressing JA-mediated responses. However, recent evidence has shown that certain biotrophic fungal species also trigger activation of JA-mediated responses and mutants deficient in JA signaling show an increase in susceptibility to certain biotrophic fungal pathogens. These findings suggest a new role for JA in defense against fungal biotrophs. This review will focus on recent research advancing our knowledge of JA-dependant responses involved in defense against both biotrophic and necrotrophic fungi.

An immunoglobulin Y that specifically binds to an in silico-predicted unique epitope of Zika virus non-structural 1 antigen

Objective:To identify unique immunogenic epitopes of Zika virus non-structural 1(NS1)antigen and produce immunoglobulin Y(IgY)for potential use in he diagnosis of of Zika virus ***:Immunogenic epitopes were identified using in silico B-cell epitope prediction.A synthetic peptide analog of the predicted epitope was used to induce antipeptide IgY production in hens which was purified using affinity *** of purified IgY and its binding specificity were performed by gel electrophoresis and ELISA,***:Out of the nine continuous epitopes identified,the sequence at position 193-208(LKVREDYSLECDPAVI)was selected and used to produce anti-peptide *** produced IgY was found to bind to the synthetic analog of the Zika virus NS1 immunogenic epitope but not to other flaviviruses and random peptides from other ***:In this study,we identified an immunogenic epitope unique to Zika virus that can be used to develop a serodiagnostic tool that specifically detect Zika virus infection.
