A step-by-step protocol for formaldehyde-assisted isolation of reg.latory elements from Arabidopsis thaliana

The control of g.ne expression by transcriptional reg.lators and other types of functional y relevant DNA transactions such as chromatin remodeling.and replication underlie a vast spectrum of biolog.cal processes in al org.nisms. DNA transactions require the control ed interaction of proteins with DNA sequence motifs which are often located in nucleosome-depleted reg.ons （NDRs） of the chromatin. Formaldehyde-assisted isolation of reg.latory elements （FAIRE） has been established as an easy-to-implement method for the isolation of NDRs from a number of eukaryotic org.nisms, and it has been successful y employed for the discovery of new reg.latory seg.ents in g.nomic DNA from, for example, yeast, Drosophila, and humans. Until today, however, FAIRE has only rarely been employed in plant research and currently no detailed FAIRE protocol for plants has been published. Here, we provide a step-by-step FAIRE protocol for NDR discovery in Arabidopsis thaliana. We demonstrate that NDRs isolated from plant chromatin are readily amenable to quantitative polymerase chain reaction and next-g.neration sequencing. Only minor modification of the FAIRE protocol wil be needed to adapt it to other plants, thus facilitating.the g.obal inventory of reg.latory reg.ons across species.

BACKg.OUND Liver transplantation for the most critically ill remains controversial;however,it is currently the only curative treatment *** To assess immediate posttransplant outcomes and compare the short(1 year)and long.term(6 years)posttransplant survival among.cirrhotic patients stratified by disease *** We included cirrhotic patients underg.ing.liver transplantation between 2015 and 2019 and categ.rized them into compensated cirrhosis(CC),decompensated cirrhosis(DC),and acute-on-chronic liver failure(ACLF).ACLF was further divided into severity *** primary outcomes of interest were total days of intensive care unit(ICU)and hospital stay,development of complications and posttransplant survival at 1 and 6 *** 235 patients underwent liver transplantation(CC=11,DC=129 and ACLF=95).Patients with ACLF had a sig.ificantly long.r hospital stay[8.0(6.0-13.0)vs CC,6.0(3.0-7.0),and DC 7.0(4.5-10.0);P=0.01]and developed more infection-related complications[47(49.5%),vs CC,1(9.1%)and DC,38(29.5%);Plant survival at 1-and 6-years was similar among.g.oups(P=0.60 and P=0.90,respectively).ACLF patients stratified according.to ACLF g.ade[ACLF-1 n=40(42.1%),ACLF-2 n=33(34.7%)and ACLF-3 n=22(23.2%)],had similar ICU and hospital stay leng.h(P=0.68,P=0.54),as well as comparable frequencies of overall and infectious posttransplant complications(P=0.58,P=0.80).There was no survival difference between ACLF g.ades at 1 year and 6 years(P=0.40 and P=0.15).CONCLUSION Patients may benefit from liver transplantation reg.rdless of the cirrhosis *** patients have a long.r hospital stay and frequency of infectious complications;however,excellent,and comparable 1 and 6-year survival rates support their enlisting.and transplantation including.those with ACLF-3.

Concentrations and sources of an emerg.ng.pollutant, decabromodiphenylethane (DBDPE), in sewag. sludg. for land application

The presence of an emerg.ng.brominated flame retardant, decabromodiphenylethane （DBDPE）, has been confirmed in Spanish sewag. sludg.. Thirty one samples from different urban wastewater treatment plants （WWTPs） were analyzed for this brominated flame retardant. DBDPE was positively identified and quantified in all samples at lower concentrations （47.0 ± 29.7 ng.g.dry weig.t （dw）; mean ± SD） than those obtained for decabromodiphenyl ether （Bde-209）（290 ± 236 ng.g.dw; mean ± SD） in a previous study. Influence of the WWTP characteristics in the pollutant levels was evaluated. No sig.ificant correlations were obtained between DBDPE concentrations and the population or sewag. sludg. production rate associated with the plants, neither wastewater treatment method. Sources of DBDPE in the sludg. were also evaluated. Data indicate a common orig.n for DBDPE and Bde-209, which may be related to leaching.processes during.the use and disposal of consumer products containing.these chemicals. Nevertheless, DBDPE contents are not influenced by industrial activities, which sug.ests that the infusion of DBDPE commercial mixture is not a source of this chemical into the environment, and indicates that the use of DBDPE in the Spanish industry is still low compared to deca-Bde.

A Note on Tropical Triang.es in the Plane

We define transversal tropical triang.es （affine and projective） and characterize them via six inequalities to be satisfied by the coordinates of the vertices. We prove that the vertices of a transversal tropical triang.e are tropically independent and they tropically span a classical hexag.n whose sides have slopes ∞, 0, 1. Using.this classical hexag.n, we determine a parameter space for transversal tropical triang.es. The coordinates of the vertices of a transversal tropical triang.e determine a tropically reg.lar matrix. Triang.lations of the tropical plane are obtained.
