Dose of alendronate directly increases trabeculae expansivity without altering bone volume in rat fem.rs

AIm To evaluate the effects of sodium alendronate on bone repair in fractures created in appendicular bones. mETHODS Wistar rats(n = 36) were allocated into three distinct groups: group C(control), group B1(received 1 mg/kg of alendronate), and group B2(received 3 mg/kg of alendronate). The rats underwent fem.ral transversal linear fracture surgery using stable internal fixation with a 2.0 mm plate and screw system. Each animal randomly received intraperitoneal applications of sodium alendronate at a dose corresponding to group B1 or B2 three times a week, while the control group received a 0.9% saline solution. Drug administration was performed until euthanasia at 45 d. The fem.rs were removed and each surgical piece was sent for radiographic, tomographic and microtomographic analysis. Data were submitted to descriptive and inferential statistical analysis(95% confidence interval).rESULTS Quantitative evaluations of bone neoformation did not show differences among the groups in the radiographic(P = 0.341), microtomographic(P = 0.581) and tomographic evaluations(P = 0.171). In the qualitative microtomographic analysis, a smaller distance was observed between the internal bone trabeculae in the groups that used alendronate(P = 0.05). On the other hand, group B2 had a higher amount of bone trabeculae per unit length when compared to the other groups(P = 0.04).CONCLUSION It is likely that the use of alendronate did not have a direct influence on the amount of bone neoformation, however it did influence the bone quality in a dosedependent manner, ultimately affecting the distance and quantity of the trabeculae.

Analysis of the interaction between [ru(phenanthroline)<sub>3</sub>]<sup>2+</sup>and bovine serum albumin

The interaction of compounds with potential use as pharmaceutical with a carrier protein as serum albumin is of great importance in their biodistribution. Albumin offers different sites for binding metallic compounds. Using a combination of spectropho-tometric and electrochemical techniques, the interaction between [ru(phen)3]Cl2 (phen = phenantroline) and bovine serum albumin was evaluated. In particular, it was possible to calculate an apparent binding constant (Kb) of 4.4 × 103 (for concentrations expressed in m) for the main interaction site of the protein. A number of ca. 40 molecules of ru-phen per molecule of BSA under saturation conditions, and a positive cooperative behavior towards association from the protein were found.

文章简要报道湖北中部京山县郊发现的一个布尔吉斯页岩型生物群,该生物群由大量三叶虫、腕足类、软舌螺及少量鳃曳动物、双瓣壳节肢动物、非三叶虫节肢动物和棘皮动物等组成。文中重点描述非三叶虫节肢动物Clypecaris?和棘皮动物Sinoeocrinus,两者分别为澄江动物群和凯里生物群的典型分子。依据生物地层学证据,该生物群的时代大致为寒武纪第四期晚期(龙王庙期),与凯里生物群下部组合基本相当,稍晚于云南东部的关山动物群和峡东地区石牌组的布尔吉斯页岩型生物群。该发现进一步丰富了华中地区早-中寒武世化石库的资料,扩大了布尔吉斯页岩型生物群的地理分布,并是目前已知唯一的分布在扬子地台北缘的该类型生物群,这对寒武纪环扬子地台古生态的重建和多门类后生生物的演化都将提供重要资料。

AIm To investigate the contribution of polym.rphisms in the CYP1A1, CYP2E1 and EPHX1 genes on sporadic colorectal cancer(SCrC) risk. mETHODS Six hundred forty-one individuals(227 patients with SCrC and 400 controls) were enrolled in the study. The variables analyzed were age, gender, tobacco and alcohol consumption, and clinical and histopathological tum.r parameters. The CYP1A1 *2A, CYP1A1 *2C CYP2E1 *5B and CYP2E1 *6 polym.rphisms were analyzed by polym.rase chain reaction-restriction fragment length polym.rphism(PCr-rFLP). The EPHX1 Tyr113 His, EPHX1 His139 Arg and CYP1A1 *2C polym.rphisms were detected by real-time PCr. Chisquared test and binary logistic regression were used in the statistical analysis. Ecological and hydrological impacts are expected when watersheds are under comm.rcial *** this study we evaluated event-based hydrology and sedimentation in paired watersheds under comm.rcial eucalyptus and grasslands ***,suspended sediment concentration,and sediment production were measured and simulated with the Limburg Soil Erosion model(LISEm)in paired hydrographic watersheds located in southern Brazil;one occupied with 7-years old Eucalyptus saligna plantation(forest watershed-FW;0.83 km^(2))and another Pampa biome with native and exotic grasses used for beef cattle production(grassland watershed-GW;1.10 km^(2)).The effect of rainfall on hydrological and sedimentological variables was m.re evident in GW compared with *** shape of the hydrograph was steepest in GW,with rapid,m.rked increase and decrease in flow during *** parameters changed for model calibration in FW and GW were soil saturated hydraulic conductivity,initial soil moisture,and manning roughness coefficient,whereby the statistical performance was classified as good(NSE>0.75).Fourteen events different from those used in the calibration process were validated for FW and GW,with NSE index classified as“good”and“very good”for *** study shows limitations of LISEm in calibrating and validating the sediment yield for individual events due to the spatial variability of factors controlling soil *** validated modeling results of this study may be applied in simulating soil and crop scenarios in watersheds cultivated with comm.rcial forest and grassland for grazing.

Antimicrobial Efficacy of Curcumin Formulations byPhotodynamic Therapy

UrTI （upper respiratory tract infections） are caused by acute, chronic or recurrent infections including pharyngitis andtonsillitis. The etiologic agents involved are bacteria, virus, and fungi. The standard treatment for bacterial infections is the use ofantibiotics. The antibiotic resistance, side effects of drugs and UrTI complications indicate the importance of new therapies. PDT（photodynamic therapy） is a treatment option with a fast onset of action, low side effects, and easy administration of PS（photosensitizer） based on this study. The main aim of this study was to develop a new modality for the treatment of infectiouspharyngotonsillitis by photodynamic therapy. In addition, this study aims to evaluate components of the therapy as the PS, curcumin,in two formulations and irradiation conditions in the effectiveness of treatment. The procedure consists of two steps： oraladministration of CFs （curcumin formulations） containing a PS and illumination （450 nm and 0-60 J/cm2）. Bacterial isolates wereobtained from patients in a routine clinical pratice. The new methodology has been developed from the principle ofPI （photodynamicinactivation）. The strain identification number of colony forming units determined the efficacy of PI using each component, lightingand PS. The use of curcumin formulations in PI presented microbial reduction of 5 log.

Adaptation and Validation of a Proteolytic Activity methodology Using Dye-Crosslinked Substrate Tablets

Enzymatic activities are important to be quantified in products as enzymatic cleaners, which are used in medical and surgical devices reprocessing. Enzymatic activities are critical for the proper chemical cleaning that intends to remove solid organic dirt from inaccessible sites. The most important enzyme for this purpose is the protease, which is able to dissolve the main dirt attached to medical and surgical instruments. In this context, this study contributes to the development of a new proteolytic activity quantification method and its validation. The methodology is based on colorimetry and uses a UV-Vis spectrophotometer to measure the substrate hydrolysis by the blue color intensity, employing Protazyme AK tablets as substrate.

In the search of lead-free piezoelectric materials,ceramic processing techniques offer potential tools to increase the piezoelectric and ferroelectric properties in addition to new chemical *** of pure BNKT16(Bi0.5(Na0.84K0.16)0.5TiO3)phase were synthesized by sol–gel method with a low crystallization temperature(750℃).Ceramic samples were sintered by pressureless sintering(PLS),sinter-forging(SF),and spark plasma sintering(SPS)***,m.rphological,and chemical characterizations were performed by XrD,raman,EDS,and *** samples by PLS and SF exhibit rod-like grains associated to bismuth *** highest remanent polarization(11.05μC/cm2),coercive field(26.2 kV/mm),and piezoelectric coefficient(165 pC/N)were obtained for SF *** piezoresponse force microscopy(PFm)analysis shows that the crystallites at the nanoscale exhibit piezoelectric phenomenon and the highest piezoelectric response is reported for PLS *** presence of the rhombohedral phase,the increase in grain and crystallite size,and the oriented rod-like inclusions favoring the crystallographic texture are facts that enhance the piezoelectric coefficient for BNKT16 piezoceramics.

In vivo study of conductive 3D printed PCL/mWCNTs scaffolds with electrical stimulation for bone tissue engineering

Critical bone defects are considered one of the major clinical challenges in reconstructive bone *** combination of 3D printed conductive scaffolds and exogenous electrical stimulation(ES)is a potential favorable approach for bone tissue *** this study,3D conductive scaffolds made with biocompatible and biodegradable polycaprolactone(PCL)and multi-walled carbon nanotubes(mWCNTs)were produced using the extrusion-based additive manufacturing to treat large calvary bone defects in *** results show that the use of PCL/mWCNTs scaffolds and ES contributes to thicker and increased bone tissue formation within the bone *** and mineralization are also significantly promoted using high concentration of mWCNTs(3 wt%)and ***,scaffolds favor the tartrate-resistant acid phosphatase(TrAP)positive cell formation,while the addition of mWCNTs seems to inhibit the osteoclastogenesis but present limited effects on the osteoclast functionalities(receptor activator of nuclear factor κβ ligand(rANKL)and osteoprotegerin(OPG)expressions).The use of ES promotes the osteoclastogenesis and rANKL expressions,showing a dominant effect in the bone remodeling *** results indicate that the combination of 3D printed conductive PCL/mWCNTs scaffold and ES is a promising strategy to treat critical bone defects and provide a cue to establish an optimal protocol to use conductive scaffolds and ES for bone tissue engineering.

Gastric cancer remains one of the leading causes of cancer-related death worldwide,and most of the cases are associated with Helicobacter pylori *** bacterium promotes the production of reactive oxygen species(rOS),which cause DNA damage in gastric epithelial *** this study,we evaluated the expression of important genes involved in the recognition of DNA damage(ATm,ATr,and H2AX)and rOS-induced damage repair(APE1)and the expression of some m.rNAs(m.r-15a,m.r-21,m.r-24,m.r-421 and m.r-605)that target genes involved in the DNA damage response(DDr)in 31 fresh tissues of gastric *** v3.1.1 was used to construct the postulated m.rNA:mrNA interaction *** performed by real-time quantitative PCr exhibited significantly increased levels of the APE1(rQ=2.55,prQ=2.88,p=0.0002)genes beyond the m.r-421 and m.r-605 in the gastric cancer *** addition,significantly elevated levels of m.r-21,m.r-24 and m.r-421 were observed in diffuse-type gastric *** analysis reinforced some of the gene:gene(ATm/ATr/H2AX)and m.rNA:mrNA relationships obtained also with the interaction ***,our findings show that tum.r cells from gastric cancer presents deregulation of genes and m.rNAs that participate in the recognition and repair of DNA damage,which could confer an advantage to cell survival and proliferation in the tum.r microenvironment.
