Detection of Antinuclear Antibodies in Canine Serum by Immunoperoxidase in mDCK and Indirect Immunofluorescence in HEp2 Cells

Antinuclear antibodies are found in animals suffering from Systemic Lupus Erythematosus (SLE) and some other diseases, their presence in the blood is determined by antinuclear antibody (ANA) test using indirect immunofluorescence (IF) with HEp2 cells as a substrate. In this work, an immunoperoxidase (IP) assay was developed to evaluate the ANAs in canine sera, using canine kidney cell lines (mDCK) and compared with a comm.rcial immunofluorescence test on Hep2 cells for this system, a fluoresceinated anti-canine Ig antibody was standardized. The study was performed on 50 sera from dogs submitted to the laboratory with different clinical diagnoses of autoimmune-associated diseases. The procedures on both cells were unified to perform comparisons of the reactions, direct sera or at different dilutions were added to a monolayer of permeabilized mDCK cells, followed by a peroxidized anti-canine IgG conjugate, and a substrate for the IP reaction. The same sera were tested on the comm.rcial IF assay on Hep2 cell system. In 22/50 cases, the presence of LE cells in peripheral blood was determined. A high correlation was found in the detection of antinuclear antibodies between both cell lines and techniques, however there were differences in the reaction patterns in the nucleus and cytoplasm between cell lines. The diffuse nuclear pattern observed in mDCK cells was m.re related to the presence of high percentages of LE cells in peripheral blood. The differences found in the results were possibly associated with the presence of homologous antigens between the mDCK cells and the dog. In addition, the methodology and standardization for the use and interpretation of a reference serum was developed to unify the interpretation criteria in the laboratory.

Hepatitis delta virus(HDV) seems to strongly suppress hepatitis B virus(HBV)replication, although little is known about the mechanism of this interaction. Both these viruses show a dynamic distribution of mutants, resulting in viral quasispecies. Next-generation sequencing is a viable approach for analyzing the composition of these mutant spectra. As the regulatory hepatitis B X protein(HBx) is essential for HBV replication, determination of HBV X gene(HBX)quasispecies complexity in HBV/HDV infection compared to HBV monoinfection may provide information on the interactions between these two *** To compare HBV quasispecies complexity in the HBX 5' region between chronic hepatitis delta(CHD) and chronic HBV mono-infected *** Twenty-four untreated patients were included: 7/24(29.2%) with HBeAgnegative chronic HBV infection(CI, previously termed inactive carriers), 8/24(33.3%) with HBeAg-negative chronic hepatitis B(CHB) and 9/24(37.5%) with CHD. A serum sample from each patient was first tested for HBV DNA *** HBX 5' region [nucleotides(nt) 1255-1611] was then PCr-amplified for subsequent next-generation sequencing(miSeq, Illumina, United States). HBV quasispecies complexity in the region analyzed was evaluated using incidencebased indices(number of haplotypes and number of mutations), abundancebased indices(Hill numbers of order 1 and 2), and functional indices(mutation frequency and nucleotide diversity). Background: Incorporation of HPV tests into cervical cancer screening programs may be advantageous over conventional cytology, especially in developing nations, where the largest burden of cervical cancer is observed. Objectives: To conduct an evaluation of comm.rcially available molecular HPV tests in Brazilian women. Study design: Two groups were recruited: group A was composed of 511 women referred to the clinics because of a previous abnormal Pap test while group B consisted of 2464 subjects under routine screening. Cervical samples were collected using SurePath liquid cytology (LBC) device, and split into aliquots which were submitted to molecular testing by Hybrid Capture and cobas HPV. Colposcopy and biopsies were made according to the standard guidelines, directed by cytological diagnosis. results: Prevalence of HSIL was 5.97% and 0.7% in Group A and B respectively. High-risk HPV DNA was found in about 9% of group B women, while in group A this frequency was 24%. Having CIN3+ as the study end-point, the negative predictive values for molecular methods were above 99.8%. All “in-situ” and invasive cervical carcinomas were detected by both HPV nucleic acid assays. Conclusion: Use of HPV DNA testing was feasible and highly sensitive in cancer screening settings of Brazil.

The characterization and prevalence of virulence factors associated with enterococcal invasiveness and severity of disease are important areas to be investigated. recently, we described the production of a heat-stable hemolysin by clinical isolates of Enterococcus faecalis cultived in BHI-GA (BHI with glucose and L-arginine). Now, we purified the hemolysin from the culture supernatant by ultra-filtration (Pm-10 mem.rane) and ethanol extraction followed by chromatography in a mBondapak C18 and Superdex Peptide columns. The hemolytic activity was not affected by the proteolytic enzymes. Cholesterol, phospholipids, EDTA and also bivalent ions did not inhibit the hemolytic activity. Among the various carbohydrates, only dextran 4 protected the erythrocytes against lyse. Scanning electron microscopy showed that lyse of erythrocytes occured at once after the exposure to the hemolysin. The mito-chondrial activity and the cell mem.rane integrity were significantly affected by the hemolysis, within 20 min of exposure and caused apoptosis after 12 h incubation, 51.92% in HeLa and 68% in HEp-2 cells, analyzed by flow cytometry. These results suggest that the heat-stable pore forming hemolysin might be a putative virulence factor in enterococci infections.

The term Ecology is derived from οἶκος (Greek “house” or environment), but our habitations usually overlooked in ecology or environmental studies. The expression “at home” usually means safety and comfort, but at home we are under risk of innum.rous parasitic/microbial infections and contaminations/ envenomation. During the COVID-19 pandemic we were forced to stay at home, but the virus and other pathogens were also home-delivered. Education for health is highly effective in health promotion, particularly in poor areas. Thus, prophylactic interventions approaching household environment are required. The present activity aims community empowerment and engagement in controlling parasitic diseases and other infections such as Chagas disease, leishmaniasis, malaria, arboviruses etc. Inspired on the use of a house maket by Dr. Virgínia Schall to demonstrate Aedes mosquitoes breeding sites. We also employ house makets displaying pathogen vectors breeding/hiding sites. Although some makets can be opened, revealing intradomiciliary milieu, we intended to offer the public a literally “insider” view of this largely overlooked scenery. The “Interactive House”, also known as “House with no viruses and other bugsmily:Verdana;">” is an educative, interactive, ludic device elaborated on an inflatable igloo, with furniture and utensils crafted using reused/recycled or low-cost materials. Growth and Photosynthetic Pigments of Cotton Cultivars Irrigated with Saline Water

The cultivation of cotton is a relevant socioeconomic activity in the Brazilian agricultural scenario. In the Brazilian Northeast, however, production is limited by low rainfall, requiring water supplementation, a problem for the cotton cultivation practiced in the semi-arid region, due to the presence of saline ions in several water sources, from where the water is used for irrigation. It is necessary to identify cultivars that better tolerate saline stress in order to provide subsidies for their cultivation under such conditions. In this sense, the objective was to evaluate the tolerance of cotton cultivars to salinity. The experiment was conducted in a greenhouse, belonging to the Natural resource Technology Center of the Federal University of Campina Grande, Campus I, Campina Grande, Brazil, in a completely randomized design in a 2 × 5 factorial scheme, with four replications, totaling 40 experimental units (BrS 368 rF and BrS SAFIrA) submitted to five saline levels (1.5, 3.0, 4.5, 6.0 and 7.5 dS·mˉ1). Irrigation with water saline of EC from 1.5 dS·mˉ1 affects adversely the growth of cotton cultivars, causing reductions in plant height, stem diameter, leaf area and number of leaves. Saline stress reduced the levels of the photosynthetic pigments of the cotton plants studied.

目的　明确中国幽门螺杆菌 (Helicobacterpylori,Hp)全长vacA基因的等位变异及其与西方主要菌株基因序列的差异。方法　从 5例胃病患者胃黏膜活检组织中分离培养 2 2个单个Hp克隆 ,以Westernblot确定vacA表型特点 ,以体外细胞毒性试验确定其活性。然后选择 5株有代表性的克隆进行基因组DNA抽提、PCr扩增及vacA基因全序列测定、分析。结果　 5株中 4株vacA表达阳性。只有 1株 (5 0 6 0d)可在体外诱导HeLa细胞产生显著空泡变性。 5株vacA基因有相同的信号序列 (sla)及相似的编码相对分子质量为 37× 10 3 和跨膜输出段 (outermem.raneexporter ,OmE)的基因序列。 4株vacA基因的中间区 (编码相对分子质量为 5 8× 10 3 蛋白 )表现为m2型等位基因 ,1株为m1型。同型中国HpvacA基因相似率为 97.6 %～ 99.2 % ,高于西方同型相似率 (92 .1% )。结论　中国人HpvacA基因与西方菌株相比存在显著的等位变异 ,形成相对独立的一个亚簇。m2型菌株比m1型菌株常见。细胞毒活性与vacA基因中间区类型有关。
