Plasmid Dna(pDna)isolation from bacterial cells is one of the most common and critical steps in molecular cloning and biomedical *** all pDna purification in-volves disruption of bacteria,removal of membrane lipids,proteins and genomic Dna,purifi-cation of pDna from bulk lysate,and concentration of pDna for downstream *** many liquid-phase and solid-phase pDna purification methods are used,the final pDna preparations are usually contam.nated with varied degrees of host Rna,which cannot be completely digested by Rnase *** develop a simple,cost-effective,and yet effective method for Rna depletion,we investigated whether commercially available size selection magnetic beads(SSmBs),such as mag-Bind®TotalPure NGS Kit(or mag-Bind),can completely deplete bacterial Rna in pDna *** this proof-of-principle study,we demonstrated that,compared with Rnase A digestion and two commercial plasmid affinity purification kits,the SSmB method was highly efficient in depleting contam.nating Rna from pDna *** transfection and bacterial colony formation assays revealed that pDna purified from SSmB method had superior quality and integrity to pDna samples cleaned up by Rnase A digestion and/or commercial plasmid purification *** further demonstrated that the SSmB method completely depleted contam.nating Rna in large-scale pDna ***,the mag-bind-based SSmB method costs only 5-10%of most commercial plasmid purification kits on a per sample ***,the reported SSmB method can be a valuable and inexpensive tool for the removal of bacterial Rna for routine pDna preparations.

Background Tree regeneration is a key component of resilience because it promotes post-disturbance recovery of forests. Northwestern Patagonia from Argentina is occupied by Nothofagus alpina（na）, N. obliqua（No）, and N. dombeyi（Nd） forests that grow along an intense precipitation gradient, m.naged throughout shelterwood silvicultural system by technicians of the Lanin natural Reserve. The objective of this study was to evaluate the influence of seeding cuttings over the dynamics of Nothofagus mixed forests across landscape（precipitation gradient） based mainly on the composition and abundance of tree regeneration, permanent sampling plots and generalized linear mixed models. In particular, we analysed:（i） the structure of sexual and asexual regeneration during 20 years after harvest（the dynamics of m.naged forests）, and（ii） the structure of sexual regeneration in primary and m.naged forests after > 20 years of harvest（the effect of silviculture）.Results Nd was the most abundant species in the regeneration of m.naged forests during all periods in both sites despite its lower contribution to the adult cohort. During the 10–20 years period after harvest, the humid site exhibited higher regeneration density than the mesic site(120,000 and 6000 ind ha-1, respectively), and it decreased afterwards. The number of established regeneration（> 2 m height） was lower for na in the mesic site and for No in the humid site(0 and 57 ind ha-1, respectively). However, in comparison to No, na showed a higher number of sprouted stumps and sprouts per stump, and a higher sprout height in the mesic site. No exhibited higher sprout mortality in the humid site. Finally, the regeneration of primary forests showed lower density and height, and a more balanced composition than that of m.naged *** The silvicultural effects on the mixed forest regeneration dynamics was strongly influenced by the condition of sites. The main objective of this work is to study the effect of sulphur application and irrigation water quantity on some soil properties of the calcareous soils and to limit the suitable concentration to obtain satisfactory yield of faba beab under saline conditions. The study area (Tamyia district, Fayoum Governorate, Egypt) is characterized by a hot and dry climate in general with annual rainfall average of 8 mm/year, whereas the evaporation rates average ranging between 3.5 to10 mm/day. Faba bean (Vicia faba L.) was sown at 18 November 2010 and harvested at 26 April 2011. Total water consumed during faba bean growing season were 623.7 and747.2 m3/ fed for 100% and 120% irrigation treatments with increasing ranged between 3.5% and 3.7% in same sequence. Results showed that values of soil EC, pH and CaCO3 was improved as a result of both irrigation and S treatments. The improvement resulted in a gradual reduction of the studied soil properties by about 10.3%, 3.2%;17.0% and 23.1%, 4.3%;11.3% comparing S treated plot relative to the control plot at 100 and 120% irrigation treatments. Application S improved water content under studied soil constant and these increases were 8.8%, 11.8%, 8.3% and 16.9% for SP, FC, WP and AW, respectively relative to the control plots. Irrigation 100% had a positive effect on the drainable pores% which increased by about 7.0% comparing with 120% irrigation treatments. While hydraulic conductivity increased by about 16.4% and percentage of increase drainable pores was 14.0% and 21.4% for 100% and 120% irrigation treatments comparing 3rd layer with the 1st one, respectively. Increasing irrigation water from 100% to 120% resulted increasing in the studied macronutrients, N, P and K, by about 80.8%, 144.9% and 72.9%, respectively;while they increased in plant by about 118.8%, 132.8% and 62.2% as compared with the control one, respectively as a result of S application. The Egyptian older and younger granitic rocks emplaced during pre-and post-collision stages of Neoproterozoic Pan-African orogeny,respectively,are widely distributed in the southern Sinai Peninsula,constituting 70% of the basement *** Wadi el-Akhder,southwestern Sinai,is a mountainous terrain exposing two granitoid suites,namely the Wadi el-Akhder Older Granites(AOG)and the Homra Younger Granites(HYG).The AOG(granodiorites with subordinate tonalite compositions)have geochemical characteristics of medium-K calc-alkaline,metaluminous to mildly peraluminous granitoids formed in an island-arc environment,which are conformable with well-known Egyptian older granitoids rocks,whereas the HYG display calc-alkaline to slightly alkaline nature,peraluminous syeno-,monzogranites and alkali feldspar granites matching well those of the Egyptian younger *** respect to the AOG granitoids,the HYG granites contain lower Al_(2)O_(3),FeO^(*),mgO,mnO,CaO,TiO_(2),Sr,Ba,and V,but higher na_(2)O,K_(2)O,Nb,Zr,Th,and *** AOG are generally characterized by enrichment in LILE and LREE and depletion in HFSE relative to N-mORB values(e.g.,negative Nb and Ta anomalies).The geochemical features of the AOG follow assimilation-fractional crystallization(AFC)trends indicative of extensive crustal contam.nation of magma derived from a mantle *** chemical characteristics of the AOG are remarkably similar to those of subduction-related granitoids from the ArabianNubian Shield(ANS).The compositional variations from monzogranites through syenogranites to alkali feldspar granite within HYG could not be explained by fractional crystallization *** the whole-rock composition of the HYG to melts generated by experimental dehydration melting of meta-sedimentary and magmatic rocks reveals that they appear to be derived by extended melting of psammitic and pelitic metasediments,which is similar to the most of younger granitic suites in the ANS.

The aim of this research is to map the salt-affected soil in an arid environment using an advanced semi-empirical predictive model, Operational Land Imager (OLI) data, a digital elevation model (DEm), field soil sampling, and laboratory and statistical analyses. To achieve our objectives, the OLI data were atmospherically corrected, radiometric sensor drift was calibrated, and distortions of topography and geometry were corrected using a DEm. Then, the soil salinity map was derived using a semi-empirical predictive model based on the Soil Salinity and Sodicity Index-2 (SSSI-2). The vegetation cover map was extracted from the Transformed Difference Vegetation Index (TDVI). In addition, accurate DEm of 5-m pixels was used to derive topographic attributes (elevation and slope). Visual comparisons and statistical validation of the semi-empirical model using ground truth were undertaken in order to test its capability in an arid environment for moderate and strong salinity mapping. To accomplish this step, fieldwork was organized and 120 soil samples were collected with various degrees of salinity, including non-saline soil samples. Each one was automatically labeled using a digital camera and an accurate global positioning system (GPS) survey (σ ≤ ± 30 cm) connected in real time to the geographic information system (GIS) database. Subsequently, in the laboratory, the major exchangeable cations (Ca2+, mg2+, na+, K+, Cl- and SO42-), pH and the electrical conductivity (EC-Lab) were extracted from a saturated soil paste, as well as the sodium adsorption ratio (SAR) being calculated. The EC-Lab, which is generally accepted as the most effective method for soil salinity quantification was used for statistical analysis and validation purposes. The obtained results demonstrated a very good conformity between the derived soil salinity map from OLI data and the ground truth, highlighting six major salinity classes: Extreme, very high, high, moderate, low and non-saline. This works aims at preparing at room stable na2FeO4 and tracking its degradation over time. The synthetic, during this step, was carried out by electrochemical method. The latter was given maximum focus because of its simplicity and the high degree of purity of the resulting product with respect to wet and dry method. This paper reviews the development of the electrochemical method applied to the synthesis of stable at room.na2FeO4, optimizing the parameters impacting the performance of the oxidation of iron(II) in to iron(VI) in alkaline naOH, saturated at a temperature of 61°C and a current density of 1.4 A/dm2, in order to simplify the synthesis process, to minimize the cost and to improve the production of iron(VI) to meet the growing demand of ferrate(VI) useful for water treatment. The supervision of the degradation of synthesized na2FeO4 shows its stability over a period of 10 months, which makes storage and transport easier. The phases obtained were characterized by IR spectrometry, X-ray, m?ssbauer, spectroscopy and thermogravimeric analysis.

An-najaf province is situated in south-western part of Iraq. It is 70 meters above the sea level in the dry desert environment. The city is considered as one of the most important cities in Iraq, facing a fast population growth and continuous development in constructions such as residential complexes, hotels, bridges and shopping malls. Soil investigation data for An-najaf Province (An-najaf and Kufa cities) from 464 boreholes drilled by the na-tional Centre for Construction Laboratories & Researches (NCCLR)/Babylon laboratory were used in this research. The data were analysed and possessed using Excel program then represented on the Geographical Information System (GIS) program by Inverse Distance Weighted (IDW) tool to create an allowable bearing capacity map for the soil at depths 0 - 2 meters. The allowable bearing capacity is one of the most important soil characteristics to be considered when making a database for An-najaf city soil. Geographical Information System GIS program enables to create reliable database for any characteristic and it is one of the best programs to produce an accurate map and allow ease in dealing with it. Those maps cover all the studied areas and by using contour lines, approximate values for no-data areas can be obtained. The results show that the allowable bearing capacity range is 5 - 20 Ton/m2 for both An-najaf and Kufa cities. Kufa city has the range 5 - 9 Ton/m2. An-najaf city has the range 7 - 18 at the center, 8 - 10 Ton/m2 at the north eastern part, 7 - 14 Ton/m2 for the north western part, 6 - 12 Ton/m2 at the south eastern and 12 - 19 Ton/m2 at the south western.

In the present investigation, two zeolite-rich tuffs from Guaymas and el Cajon（State of Sonora）, which were conditioned with a sodium chloride solution and subsequently modified with a thiourea solution, were chosen to evaluate the removal of Cd from aqueous systems. The zeolitic materials were characterized by scanning electron microscopy and X-ray diffraction. The surface area was also determined. The experiments were performed in a batch system, and the influences of p H, contact time between phases, and the concentration of Cd in the solution on the adsorption by sodium or thiourea-modified zeolite-rich tuff were investigated. It was found that the efficiency of Cd ion removal from aqueous solutions is influenced by the p H of the aqueous systems. The Cd adsorption kinetic data were well fitted to a pseudo-second-order model in all cases. The Langmuir and Langmuir–Freundlich isotherms adequately described the Cd sorption behavior by the zeolite from el Cajon pretreated with na Cl and the zeolite from Guaymas modified with thiourea, respectively.
