A novel GPCR mediates pancreatic cancer associated fibroblast-cancer cell interaction

OBJECTIVE Pancreatic ductal adenocarcinoma(PDAC),a lethal cancer in need of new,effective therapies,has a unique tumor microenvironment characterized by a dense fibrotic stroma(desmoplasia)that is generated by pancreatic cancer-associated fibroblasts(PCAFs)derived from pancreatic stellate cells(PSCs)and pancreatic fibroblasts(PFs).mETHEDS and RESULTS Hypothesizing that G protein-coupled receptors(GPCRs)may regulate PCAFs,we used an unbiased GPCRomic array approach to compare GPCR expression in PCAFs,PFs and PSCs and identified 82 GPCRs commonly expressed by PCAFs derived from primary tumors of five PDAC *** discovered that PCAFs have increased expression of numerous GPCRs,in particular a GPCR with much higher expression in PCAFs compared to both PFs and *** revealed increased expression of this GPCR in PDAC ***-culture of PSCs with PDAC cells or incubation with TNFαinduced its *** of the GPCR in PCAF sincreased expression of interleukin-6(IL-6)via a cA mP/PKA/CREB signaling *** knockdown with siR NA diminished IL-6 production and secretionby PCAFs and ability of PCAF conditioned media to enhance proliferation of PDAC *** We conclude that PDAC cells induce expression by PCAFs of a novel GPCR,resulting in increased IL-6 production by PCAFs and promotion of PDAC cell *** PCAF-expressed GPCR thus contributes to PDAC cell-PCAF interaction and as such,may be a novel therapeutic target for PDAC tumors.

Contact with environmental microbes are arguably the most common species interaction in which any animal *** have noted diverse relationships between hosts and resident microbes,which can have strong consequences for host development,physiology,and behavior. many of these studies focus specifically on pathogens or beneficial microbes,while the benign microbes,of which the majority of bacteria could be described,are often ***,we explore the nature of the relationships between the grass spider Agelenopsis pennsylvanica and bacteria collected from their cuticles in ***,using culture-based methods,we identified a portion of the cuticular bacterial communities that are naturally associated with these ***,we topically exposed spiders to a subset of these bacterial monocultures to estimate how bacterial exposure may alter 3 host behavioral traits:boldness,aggressiveness,and activity *** conducted these behavioral assays 3 times before and 3 times after topical application,and compared the changes observed in each trait with spiders that were exposed to a sterile control *** identified 9 species of bacteria from the cuticles of 36 spiders and exposed groups of 20 spiders to 1 of 4 species of cuticular *** found that exposure to Dermacoccus nishinomiyaensis and Staphylococcus saprophyticus was associated with a lO-fold decrease in the foraging aggressiveness of spiders toward prey in their *** bacterial exposure did not have survival consequences for hosts,these data suggest that interactions with cuticular bacteria,even non- pathogenic bacteria,could alter host behavior.

In today’s world, Cloud Computing (CC) enables the users to accesscomputing resources and services over cloud without any need to own the infrastructure. Cloud Computing is a concept in which a network of devices, located inremote locations, is integrated to perform operations like data collection, processing, data profiling and data storage. In this context, resource allocation and taskscheduling are important processes which must be managed based on the requirements of a user. In order to allocate the resources effectively, hybrid cloud isemployed since it is a capable solution to process large-scale consumer applications in a pay-by-use manner. Hence, the model is to be designed as a profit-driven framework to reduce cost and make span. With this motivation, the currentresearch work develops a Cost-Effective Optimal Task Scheduling model(CEOTS). A novel algorithm called Target-based Cost Derivation (TCD) modelis used in the proposed work for hybrid clouds. moreover, the algorithm workson the basis of multi-intentional task completion process with optimal resourceallocation. The model was successfully simulated to validate its effectivenessbased on factors such as processing time, make span and efficient utilization ofvirtual machines. The results infer that the proposed model outperformed theexisting works and can be relied in future for real-time applications.

Analysis of Trends of Sleep-Related Breathing Disorders in Patients with Systemic Essential Hypertension

Background: Studies have revealed a strong link between sleep-related breathing disorders and essential hypertension. However, the trends of sleep-disordered breathing in hypertensive patients have not been analyzed frequently. methods: In this prospective, single-center and observational study, random patients with systemic essential hypertension, who were visiting GSVm medical College, Kanpur, India between December 2012 and July 2014, were selected. After obtaining informed consent, patients were analyzed for symptoms of sleep-disordered breathing and Epworth Sleepiness Scale. Subsequently, all patients underwent overnight polysomnography to estimate the events of apnea and hypopnea. Observed data were compared between stage I and stage II hypertensive patients. Results: A total of 51 patients (age: 45.7 ± 13.43 years), comprising 36 stage I hypertensives and 15 stage II hypertensives, were enrolled in the study. About 47% of these patients displayed no symptoms of sleep-disordered breathing. Compared to stage I hypertensives, higher percentage of stage II hypertensives reported excessive day time sleepiness. Sleep latency time and sleep architecture were comparable between stage I and stage II hypertensives. Stage II hypertensives displayed shorter total sleep time and higher snore time than stage I hyper-tensives. Apnea Hypopnea Index (AHI) values increased with the severity of hypertension. Nocturnal oxygen desaturation was present in 25.0% and 53.3% of stage I and II hypertensives respectively. mean lowest oxygen saturation was lower and the mean C-reactive protein levels were higher in stage II hypertensives as compared to stage I hypertensives. The mean C-reactive protein levels increased with increasing AHI scores, while the mean lowest oxygen saturation decreased with increasing AHI scores. Electromagnetic and Thermal Simulations of Human Neurons for SAR Applications

The impact of the electromagnetic waves (Em) on human neurons (HN) has been under investigation for decades, in efforts to understand the impact of cell phones (radiation) on human health, or radiation absorption by HN for medical diagnosis and treatment. Research issues including the wave frequency, power intensity, reflections and scattering, and penetration depths are of important considerations to be incorporated into the research study. In this study, computer simulation for the Em exposure to HN was studied for the purpose of determining the upper limits of the electric and magnetic field intensities, power consumption, reflections and transmissions, and the change in temperature resulting from the power absorption by human neurons. Both high frequency structural simulators (HFSS) from ANSYS software, and COmSOL multi-physics were used for the simulation of the Em transmissions and reflections, and the temperature profile within the cells, respectively. For the temperature profile estimation, the study considers an electrical source of 0.5 watt input power, 64 mHz. The Em simulation was looking into the uniformity of the fields within the sample cells. The size of the waveguide was set to be appropriate for a small animal model to be conducted in the future. The incident power was fully transmitted throughout the waveguide, and less than 1% reflections were observed from the simulation. The minimum reflected power near the sample under investigation was found to be with negligible reflected field strengths. The temperature profile resulting from the COmSOL simulation was found to be near 0.25 m°K, indicating no change in temperature on the neuro cells under the Em exposure. The paper details the simulation results for the Em response determined by HFSS, and temperature profile simulated by COmSOL.
