CYP24A1 inhibition facilitates the anti-tumor effect of vitamin D3 on colorectal cancer cells

AIm:the effects of vitamin D3 have been investigated on various tumors, including colorectal cancer (CRC). 25-hydroxyvitamin-D3-24-hydroxylase (CYP24A1), the enzyme that inactivates the active vitamin D3 metabolite 1,25-dihydroxyvitamin D3 (1,25-D3), is considered to be the main enzyme determining the biological halflife of 1,25-D3. During colorectal carcinogenesis, the expression and concentration of CYP24A1 increases significantly, suggesting that this phenomenon could be responsible for the proposed efficacy of 1,25-D3 in the treatment of CRC. the aim of this study was to investigate the anti-tumor effects of vitamin D3 on the human CRC cell line Caco-2 after inhibition of the cytochrome P450 component of CYP24A1 activity. mEtHODS:We examined the expression of CYP24A1 mRNA and the effects of 1,25-D3 on the cell line Caco-2 after inhibition of CYP24A1. Cell viability and proliferation were determined by means of sulforhodamine-B staining and bromodeoxyuridine incorporation, respectively, while cytotoxicity was estimated via the lactate dehydrogenase content of the cell culture supernatant. CYP24A1 expression was measured by realtime reverse transcription polymerase chain reaction. A number of tetralone compounds were synthesized to investigate their CP24A1 inhibitory activity. RESULtS:In response to 1,25-D3, CYP24A1 mRNA expression was enhanced significantly, in a time- and dose-dependent manner. Caco-2 cell viability and proliferation were not influenced by the administration of 1,25-D3 alone, but were markedly reduced by coadministration of 1,25-D3 and KD-35, a CYP24A1-inhibiting tetralone. Our data suggest that the mechanism of action of co-administered KD-35 and 1,25-D3 does not involve a direct cytotoxic effect, but rather the inhibition of cell proliferation. CONCLUSION:these findings demonstrate that the selective inhibition of CYP24A1 by compounds such as KD-35 may be a new approach for enhancement of the anti-tumor effect of 1,25-D3 on CRC.

Breast cancer is the predominant form of carcinoma among women worldwide,with 70%of advanced patients developing bone metastases,with a high mortality *** this sense,the bone marrow(Bm)mesenchymal stem/stromal cells(mSCs)are critical for Bm/bone homeostasis,and failures in their functionality,transform the Bm into a premetastatic niche(PmN).We previously found that Bm-mSCs from advanced breast cancer patients(BCPs,infiltrative ductal carcinoma,stage III-B)have an abnormal *** work aims to study some of the metabolic and molecular mechanisms underlying mSCs shift from a normal to an abnormal profile in this group of patients.A comparative analysis was undertaken,which included self-renewal capacity,morphology,proliferation capacity,cell cycle,reactive oxygen species(ROS)levels,and senescence-associatedβ‑galactosidase(SA‑β‑gal)staining of Bmderived mSCs isolated from 14 BCPs and 9 healthy volunteers(HVs).Additionally,the expression and activity of the telomerase subunit tERt,as well as telomere length,were *** levels of pluripotency,osteogenic,and osteoclastogenic genes(OCt-4,SOX-2,m-CAm,RUNX-2,BmP-2,CCL-2,m-CSF,and IL-6)were also *** results showed that mSCs from BCPs had reduced,self-renewal and proliferation *** cells also exhibited inhibited cell cycle progression and phenotypic changes,such as an enlarged and flattened ***,there was an increase in ROS and senescence levels and a decrease in the functional capacity of tERt to preserve telomere *** also found an increase in pro-inflammatory/pro-osteoclastogenic gene expression and a decrease in pluripotency gene *** conclude that these changes could be responsible for the abnormal functional profile that mSCs show in this group of patients.

Introduction: Neisseria is a large genus of bacteria that colonize mucosal surfaces. Of the 11 species that colonize humans, only two are pathogens, N. meningitidis and N. gonorrhoeae. Both are obligate human pathogens;the last is causal agent of gonorrhea disease. Although curable with timely antibiotic treatment, an annual incidence of more than 62 million cases is estimated by the World Health Organization and there are no successful vaccines available. In contrast, several prophylactic vaccine options for Neisseria meningitidis meningitis do exist. Of note, there is trace of cross parenteral response induced between N. meningitidis and N. gonorrhoeae, and Proteoliposome (PL, also named outer membrane vesicles, OmV) vaccine has shown high impact on this response. Objective: to determine effect of VAmENGOC-BC? and its derivates (AFPL1 and AFCo1) at mucosal and systemic level and possible cross response against Neisseria gonhorroeae in Balb/c and C57Bl/6 mice. methods: We evaluated cross response against N. gonorrhoeae in mouse serum IgG and saliva IgA after mucosal immunization with VA-mENGOC-BC or its derivatives (AF, Adjuvant Finlay PL1 or AFCo (cochleate) 1). Results: Immunizations with AFPL1 or AFCo1 induce anti N. gonorrhoeae at saliva IgA and serum IgG responses similar to VA-mENGOC-BC? vaccine. Conclusions: Such data confirms a new possible window of prime-boost vaccination strategy against gonorrhea and extends our knowledge regarding the effect of PL vaccines on cross responses.

Copolymer-1（Cop-1） is a peptide with immunomodulatory properties, approved by the Food and Drug Administration of United States in the treatment of multiple sclerosis. Cop-1 has been shown to exert neuroprotective effects and induce neurogenesis in cerebral ischemia models. Nevertheless, the mechanism involved in the neurogenic action of this compound remains unknown. the choroid plexus（CP） is a network of cells that constitute the interphase between the immune and central nervous systems, with the ability to mediate neurogenesis through the release of cytokines and growth factors. therefore, the CP could play a role in Cop-1-induced neurogenesis. In order to determine the participation of the CP in the induction of neurogenesis after Cop-1 immunization, we evaluated the gene expression of various growth factors（brain-derived neurotrophic factor, insulin-like growth factor 1, neurotrophin-3） and cytokines（tumor necrosis factor alpha, interferon-gamma, interleukin-4（IL-4）, IL-10 and IL-17）, in the CP at 14 days after ischemia. Furthermore, we analyzed the correlation between the expression of these genes and neurogenesis. Our results showed that Cop-1 was capable of stimulating an upregulation in the expression of the genes encoding for brain-derived neurotrophic factor, insulin-like growth factor 1, neurotrophin-3 and IL-10 in the CP, which correlated with an increase in neurogenesis in the subventricular and subgranular zone. As well, we observed a downregulation of IL-17 gene expression. this study demonstrates the effect of Cop-1 on the expression of growth factors and IL-10 in the CP, in the same way, presents a possible mechanism involved in the neurogenic effect of Cop-1.

Cystic lymphangioma of the pancreas

Lymphangioma of the pancreas is an extremely rare benign tumour of lymphatic origin, with fewer than 60 published cases. Histologically, it is polycystic, with the cysts separated by thin septa and lined with endothelial cells. though congenital, it can affect all age groups, and occurs more frequently in females. Patients usually present with epigastric pain and an associated palpable mass. Complete excision is curative, even though, depending on the tumour location, surgery may be simple or involve extensive pancreatic resection and anastomoses. the authors present a 49-year-old woman in whom a polycystic septated mass, 35 mm× 35 mm in size, was discovered by ultrasonography （US） in the body of the pancreas during investigations for epigastric pain and nausea. At surgery, a well circumscribed polycystic tumor was completely excised, with preservation of the pancreatic duct. the postoperative recovery was uneventful. Histology confirmed a microcystic lymphangioma of the pancreas. Immunohistochemistry showed cystic endothelial cells reactivity to factor Ⅷ -ra （＋＋）, CD31 （＋＋＋） and CD34 （-）. Postoperatively, abdominal pain disappeared and the patient remained symptomfree for 12 mo until now. Although extremely rare, lymphangioma of the pancreas should be taken into consideration as a differential diagnosis of a pancreatic cystic lesion, especially in women.

Assessment of radiation Hazard from External and Internal Exposures at Adham and Surroundings in KSA

twenty-eight environmental samples (eight well water, sixteen granitic rocks and four soils) were collected from different parts of Adham governorate (Adham, Haqal and Al-Jaizah), to assess the radiological hazard and cancer risk from different perspectives. Adham is situated in a valley between two granitic mountain chains, where much of water supply for drinking, house use and irrigation comes from wells collecting water rains. the activity concentrations of naturally occurring 40K, 226ra and 232th and radionuclides were measured by gamma-ray spectrometry for all samples using RGK-1, RGU-1 and RGth-1, IAEA reference standards issued by the International Atomic Energy Agency, for detector efficiency calibration. the measured values were utilized to evaluate the internal and external exposures both outdoors and indoors. Different standard room models were adopted for this respect to evaluate the indoor gamma-rays exposure from construction materials as well as internal exposure to radon gas emanating from them. radon concentration indoors, exceeded the upper reference level in dwellings set at 300 Bq/m3 by the world health organization, in many scenarios. the mean value of the total excess lifetime cancer risk (due to external exposure from gamma-rays) was 2.29 × 10-3, above the world average value of 1.45 × 10-3. Furthermore, the measured radon concentrations in all water samples exceeded the EPA (Environmental Protection Agency) 11.1 Bq·L-1 standard for drinking water, ranging from 12 to 38 Bq·L-1 with a mean value of 27 Bq·L-1. the total annual effective dose (due to inhalation and ingestion) from radon in water, ranged from 58 to 192 μSv/y (for adults) exceeding the international permissible limit of 100 μSv/y, in seven out of eight samples. Destructive and Nondestructive Determination of 226ra and 228ra in Drinking Water by Gamma Spectrometry

the Safe Drinking Water Act (SDWA) mandates that the drinking water should be monitored for 226ra and 228ra isotopes and establishes the maximum Contaminant Level of 185 mBq/L (5 pCi·L-1) for the sum. In addition, SDWA regulates the Detection Limit (DL) of 37.0 mBq/L (1 pCi/L) for each isotope. the purpose of this work is to develop a working method for the determination of radium isotopes in drinking water satisfying the regulatory requirements of U.S. Environmental Protection Agency by utilizing our extensive experience in low-background gamma spectrometry at this laboratory. two versions of the method were studied: destructive and non-destructive. Destructive method used the BaSO4 coprecipitation as well as 133Ba tracer for chemical recovery. We have used three gamma spectrometers: low-background 102% and 134% efficient with top muon guards, as well as an ultralow-background 140% efficient with full muon guard. We obtained a range of DLs from 5.3 to 22.6 mBq/L for 226ra and from 7.4 to 30.4 mBq/L for 228ra using the destructive method. For non-destructive method, the DL range was 26.0 to 26.9 mBq/L for 226ra and 27.6 to 28.6 mBq/L for 228ra using the 140% detector. to verify the methods, 7 to 10 laboratory control samples were spiked with both 226ra and 228ra at two different activities of 37.0 and 185 mBq/L. the results were evaluated by performing a combined location/variance chi-square test at a right-tail significance of 0.01 (99% Confidence Level), as stipulated by EPA. the verification results passed the chi-square tests at both activity levels. the destructive method can be accomplished using low-background gamma spectrometry, whereas non-destructive method requires ultralow-background gamma spectrometry.

Interest of SPECt/Ct in Detection of Sentinel Lymph Node in Breast Cancer

Aim: Assess the role of hybrid modality SPECt/Ct versus planar scintigraphy in sentinel lymph node (SLN) identification in patients with breast cancer. methods: Planar scintigraphy and hybrid modality SPECt/Ct were performed in 23 women with breast cancer (mean age 59.5 years with range 25 - 82 years) with invasive breast cancer (t0, t1 and t2), without clinical evidence of axillary lymph node metastases (N0) and no remote metastases (m0), radiocolloid was injected in four subareolar sites. Planar and SPECt/Ct images were separately interpreted. Results: SLNs were detected on lymphoscintigraphy in all patients (100%), taking into consideration both techniques (planar and SPECt-Ct images). Planar images identified 45 SLNs in 23 women, with a mean of 1.95 per patient, whereas 56 SLNs were detected on SPECt/Ct, increasing this mean to 2.43 per patient. Drainage to internal mammary lymph nodes was seen in 4 patients (17.39%). However, two foci of uptake were identified on planar image as hot SLN in two patients (8.69%);while they have been found as a false positive non-nodal site of uptake on SPECt/Ct. Conclusion: SPECt/Ct is more focused than planar scintigraphy in the detection of SLN in patients with breast cancer. It detects some lymph nodes not visible on planar images, excludes false positive uptake and exactly locates axillary and non-axillary SLNs.
