Background Schistosomiasis is highly endemic in sub-Saharan Africa and frequently imported to Europe. male urogenital manifestations are often neglected. We aimed to ascertain the prevalence of genitourinary clinical signs and symptoms among long-term African migrants in a non-endemic European country using a serology test.methods We carried out a prospective, community-based cross-sectional study of adult male migrants from sub-Saharan Africa living in Spain.Schistosoma serology tests and microscopic urine examinations were carried out, and clinical data were obtained from an electronic medical record search and a structured questionnaire.Results We included 388 adult males, mean age 43.5 years [Standard Deviation (SD) = 12.0, range: 18–76]. The median time since migration to the European Union was 17 [Interquartile range (IQR): 11–21] years. The most frequent country of origin was Senegal (N = 179, 46.1%). Of the 338, 147 (37.6%) tested positive forSchistosoma. parasite eggs were present in the urine of only 1.3%. Nine genitourinary clinical items were significantly associated with positiveSchistosoma serology results: pelvic pain (45.2%;OR = 1.57, 95%CI: 1.0–2.4), pain on ejaculation (14.5%;OR = 1.85, 95%CI: 1.0–3.5), dyspareunia (12.4%;OR = 2.45, 95%CI: 1.2–5.2), erectile dysfunction (9.5%;OR = 3.10, 95%CI: 1.3–7.6), self-reported episodes of infertility (32.1%;OR = 1.69, 95%CI: 1.0–2.8), haematuria (55.2%;OR = 2.37, 95%CI: 1.5–3.6), dysuria (52.1%;OR = 2.01, 95%CI: 1.3–3.1), undiagnosed syndromic STIs (5.4%), and orchitis (20.7%;OR = 1.81, 95%CI: 1.0–3.1). Clinical signs tended to cluster.Conclusions Urogenital clinical signs and symptoms are prevalent among male African long-term migrants with a positiveSchistosoma serology results. Genital involvement can be frequent even among those with long periods of non-residence in their sub-Saharan African countries of origin. Further research is needed to develop diagnostic tools and validate therapeutic approaches to chronic schistosomiasis.

O_(3) based advanced oxidation for ibuprofen degradation

The degradation of the anti-inflammatory ibuprofen(IBp)was evaluated by several advanced oxidation processes.IBp was treated by single ozonation and oxidation with hydrogen peroxide(H_(2)O_(2)),as well as a combination of these treatments.In order to improve the efficiency,the presence of catalysts such as original carbon nanotubes,labelled as CNT,and iron oxide supported on carbon nanotubes,named as Fe/CNT sample,was considered.The evolution of IBp degradation,mineralization and toxicity of the solutions was assessed.The formation of intermediates was also monitored.In the non-catalytic processes,IBp was faster removed by single ozonation,whereas no significant total organic carbon(TOC)removal was achieved.Oxidation with H_(2)O_(2) did not present satisfactory results.When ozone and H_(2)O_(2) were combined,a higher mineralization was attained(70%after 180 min of reaction).On the other hand,in the catalytic processes,this combined process allowed the fastest IBp degradation.In terms of mineralization degree,the presence of Fe/CNT increases the removal rate in the first hour of reaction,achieving a TOC removal of 85%.Four compounds were detected as by-products.All treated solutions presented lower toxicity than the initial solution,suggesting that the released intermediates during applied processes are less toxic.

Gut region-specific TNFR expression:TNFR2 is more affected than TNFR1 in duodenal myenteric ganglia of diabetic rats

BACKGROUND Cytokines are essential in autoimmune inflammatory processes that accompany type 1 diabetes.Tumor necrosis factor alpha plays a key role among others in modulating enteric neuroinflammation,however,it has a dual role in cell degeneration or survival depending on different TNFRs.In general,TNFR1 is believed to trigger apoptosis,while TNFR2 promotes cell regeneration.The importance of the neuronal microenvironment has been recently highlighted in gut region-specific diabetic enteric neuropathy,however,the expression and alterations of different TNFRs in the gastrointestinal tract has not been reported.AIm To investigate the TNFR1 and TNFR2 expression in myenteric ganglia and their environment in different intestinal segments of diabetic rats.mETHODS Ten weeks after the onset of hyperglycemia,gut segments were taken from the duodenum,ileum and colon of streptozotocin-induced(60 mg/body weight kg i.p.)diabetic(n=17),insulin-treated diabetic(n=15)and sex-and age-matched control(n=15)rats.myenteric plexus whole-mount preparations were prepared from different gut regions for TNFR1/HuCD or TNFR2/HuCD double-labeling fluorescent immunohistochemistry.TNFR1 and TNFR2 expression was evaluated by post-embedding immunogold electron microscopy on ultrathin sections of myenteric ganglia.TNFRs levels were measured by enzyme-linked immunosorbent assay in muscle/myenteric plexus-containing(mUSCLE-mp)tissue homogenates from different gut segments and experimental conditions.RESULTS A distinct region-dependent TNFRs expression was detected in controls.The density of TNFR1-labeling gold particles was lowest,while TNFR2 density was highest in duodenal ganglia and a decreased TNFRs expression from proximal to distal segments was observed in mUSCLE-mp homogenates.In diabetics,the TNFR2 density was only significantly altered in the duodenum with decrease in the ganglia(0.32±0.02 vs 0.45±0.04,pmUSCLE-mp homogenates,both TNFRs levels significantly decre

Copper(I)selenide-nanocrystalline semiconductor was synthesized via one-step mechanochemical synthesis after 5 min milling in a planetary ball mill.The kinetics of synthesis was followed by X-ray powder diffraction analysis and specific surface area measurements of milled 2Cu/Se mixtures.The X-ray diffraction confirmed the orthorhombic crystal structure of Cu_(2)Se with the crystallite size∼25 nm.The surface chemical structure was studied by X-ray photoelectron spectroscopy,whereby the binding energy of the Cu 2p and Se 3d signals corresponded to Cu^(+)and Se^(2−)oxidation states.Transmission electron microscopy revealed agglomerated nanocrystals and confirmed their orthorhombic structure,as well.The optical properties were studied utilizing ultraviolet-visible spectroscopy and photoluminescence spectroscopy.The direct bandgap energy 3.7 eV indicated a blue-shift phenomenon due to the quantum size effect.This type of Cu_(2)Se synthesis can be easily adapted to production dimensions using an industrial vibratory mill.The advantages of mechanochemical synthesis represent the potential for inexpensive,environmentally-friendly,and waste-free manufacturing of Cu_(2)Se.

Background: The protective role of (poly)phenols against metabolic disorders has been extensively studied in adults but not in adolescents.Objective: To assess associations of dietary (poly)phenols and their subclasses with cardiometabolic health parameters in adolescents. methods: A cross-sectional study was conducted in 944 individuals aged 11 to 14 years enrolled in the SI! program for Secondary Schools trial (NCT03504059). (poly)phenol intake was assessed using semi-quantitative food frequency questionnaires and the phenol-Explorer database. The measured cardiometabolic parameters were waist circumference (WC) age-sex z-score， blood pressure (Bp) age-sex z-score， blood glucose (BG)， triglycerides (TG)， and high-density lipoprotein cholesterol (HDL-c). multilevel mixed-effect linear regression models were applied to examine the association between (poly)phenol quintiles and cardiometabolic health parameters.Results: Compared to the lowest quintile， adolescents in the highest quintile of total (poly)phenol intake had lower WC z-scores (β=-0.09， 95%CI=-0.09;-0.08， p-for trend=0.002)， mean arterial pressure z-scores (β =-0.11， 95%CI=-0.12;-0.09， p-for trend=0.206)， and HDL-c (β=-1.52， 95%CI=-2.90;-0.14， p-for trend=0.108)， after multivariable adjustment. The WC z-scores (β=-0.11， 95%CI=-0.19;-0.03， p-for trend=0.013) and HDL-c (β=-2.16， 95%CI=-3.78;-0.53， p-for trend=0.046) were lower in the highest quintile of flavonoid intake compared to the lowest quintile. The highest quintile of phenolic acid intake was associated with a lower WC z-score (β=-0.14， 95%CI=-0.26;-0.02， p-for trend=0.011) and TG levels (β=-8.83， 95%CI=-12.37;-5.30， p-for trend p-for trend p-for trend=0.034)， and with higher HDL-c (β=2.328， 95%CI= 1.40;3.17， p-for trend=0.466) compared to the lowest quintile. Cathepsin D overexpression in the nervous system rescues lethality and Aβ42 accumulation of cathepsin D systemic knockout in vivo

The lysosome is responsible for protein and organelle degradation and homeostasis and the cathepsins play a key role in maintaining protein quality control.Cathepsin D(CTSD),is one such lysosomal protease,which when deficient in humans lead to neurolipofuscinosis(NCL)and is important in removing toxic protein aggregates.prior studies demonstrated that CTSD germ-line knockout-CtsdKO(CDKO)resulted in accumulation of protein aggregates,decreased proteasomal activities,and postnatal lethality on Day 26±1.Overexpression of wildtype CTSD,but not cathepsin B,L or mutant CTSD,decreased a-synuclein toxicity in worms and mammalian cells.In this study we generated a mouse line expressing human CTSD with a floxed STOp cassette between the ubiquitous CAG promoter and the cDNA.After crossing with Nestin-cre,the STOp cassette is deleted in NESTIN+cells to allow CTSD overexpression-CTSDtg(CDtg).The CDtg mice exhibited normal behavior and similar sensitivity to subchronic 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(mpTp)induced neurodegeneration.By breeding CDtg mice with CDKO mice,we found that over-expression of CTSD extended the lifespan of the CDKO mice,partially rescued proteasomal deficits and the accumulation of Aβ42 in the CDKO.This new transgenic mouse provides supports for the key role of CTSD in protecting against proteotoxicity and offers a new model to study the role of CTSD enhancement in vivo.

plants are exposed to environmental stress,in natural and agricultural conditions.Nitric oxide(NO),a small gaseous molecule which plays important roles in plants,has been involved in many physiological processes,and emerged as an important endogenous signaling molecule in the adaptation of plants to biotic and abiotic stress.NO is produced from a variety of enzymatic and non enzymatic sources,which are not yet fully understood.Also,NO and reactive nitrogen species(RNS)can produce posttranslational modifications affecting protein function.Nitrate reductase,a key enzyme in the nitrogen metabolism,is a proposed source of NO in plants which could be affected by posttranslational modifications.Thus,different pathways seem to be involved and can also regulate NO synthesis in the plant cell under physiological or stress conditions.However,how the levels of NO are reached in such time and place to fulfill its functions,are still puzzles to elucidate.

Is autoimmune hepatitis a frequent finding among HCV patients with intense interface hepatitis?

AIm:To evaluate the overlap of autoimmune hepatitis in hepatitis C virus(HCV)-infected patients with intense interface hepatitis.mETHODS:Among 1759 patients with hepatitis C submitted to liver biopsy,92(5.2%) presented intense interface hepatitis.These patients were evaluated regarding the presence of antinuclear antibody(ANA),anti-smooth muscle antibody(SmA) and anti-liver/kidney microsomal antibody(LKm-1),levels of γ-globulin and histological findings related to autoimmune hepatitis(plasma cell infiltrate and presence of rosettes).RESULTS:Among patients with hepatitis C and intense interface hepatitis there was a low prevalence of autoantibodies(ANA=12%,SmA=5%,LKm-1=0%) and the median γ-globulin level was within the normal range.Typical histological findings of autoimmune disease were observed in only two cases(2%).After applying the score for diagnosis of autoimmune hepatitis,only one patient was classified with a definitive diagnosis of autoimmune hepatitis.Since overlap with autoimmune hepatitis was not the explanation for the intense necroinflammatory activity in patients with chronic hepatitis C we sought to identify the variables associated with this finding.The presence of intense interface hepatitis was associated with more advanced age,both at the time of infection and at the time of the biopsy,and higher prevalence of blood transfusion and alcohol abuse.CONCLUSION:Although possible,overlap with autoimmune hepatitis is a very rare association in HCV-infected patients with intense interface hepatitis,an unusual presentation which seems to be related to other host variables.

Region-dependent effects of diabetes and insulin-replacement on neuronal nitric oxide synthase-and heme oxygenase-immunoreactive submucous neurons

AIm To investigate the intestinal segment-specific effects of diabetes and insulin replacement on the density of different subpopulations of submucous neurons. mETHODS Ten weeks after the onset of type 1 diabetes samples were taken from the duodenum, ileum and colon of streptozotocin-induce diabetic, insulin-treated diabetic and sex-and age-matched control rats. Whole-mount preparations of submucous plexus were prepared from the different gut segments for quantitative fluorescent immunohistochemistry. The following double-immunostainings were performed: neuronal nitric oxide synthase(n NOS) and Hu C/D, heme oxygenase(HO) 1 and peripherin, as well as HO2 and peripherin. The density of n NOS-, HO1-and HO2-immunoreactive(IR) neurons was determined as a percentage of the total number of submucous neurons. RESULTS The total number of submucous neurons and the proportion of n NOS-, HO1-and HO2-IR subpopulations were not affected in the duodenal ganglia of control, diabetic and insulin-treated rats. While the total neuronal number did not change in either the ileum or the colon, the density of nitrergic neurons exhibited a 2-and 3-fold increase in the diabetic ileum and colon, respectively, which was further enhanced after insulin replacement. The presence of HO1-and HO2-IR submucous neurons was robust in the colon of controls(38.4%-50.8%), whereas it was significantly lower in the small intestinal segments(0.0%-4.2%, p pathophysiological conditions the only alteration detected was an increase in the ileum and a decrease in the colon of the proportion of HO-IR neurons in insulin-treated diabetic animals. CONCLUSION Diabetes and immediate insulin replacement induce the most pronounced region-specific alterations of n NOS-, HO1-and HO2-IR submucous neuronal density in the distal parts of the gut.

Background:In contrast with the negligible contribution of the forest understorey to the total aboveground phytobiomass of a forest,its share in annual litter production and nutrient cycling may be more important.Whether and how this functional role of the understorey differs across forest types and depends upon overstorey characteristics remains to be investigated.methods:We sampled 209 plots of the FunDivEUROpE Exploratory platform,a network of study plots covering local gradients of tree diversity spread over six contrasting forest types in Europe.To estimate the relative contribution of the understorey to carbon and nutrient cycling,we sampled non-lignified aboveground understorey biomass and overstorey leaf litterfall in all plots.Understorey samples were analysed for C,N and p concentrations,overstorey leaf litterfall for C and N concentrations.We additionally quantified a set of overstorey attributes,including species richness,proportion of evergreen species,light availability(representing crown density)and litter quality,and investigated whether they drive the understorey’s contribution to carbon and nutrient cycling.Results and conclusions:Overstorey litter production and nutrient stocks in litterfall clearly exceeded the contribution of the understorey for all forest types,and the share of the understorey was higher in forests at the extremes of the climatic gradient.In most of the investigated forest types,it was mainly light availability that determined the contribution of the understorey to yearly carbon and nutrient cycling.Overstorey species richness did not affect the contribution of the understorey to carbon and nutrient cycling in any of the investigated forest types.
