Efficacy and safety of tadalafil taken as needed for the treatment of erectile dysfunction in Asian men： results of an integrated analysis

We evaluated the efficacy and safety of as-needed tadalafil in a diverse clinical population （with varying patient demographics, disease severity, and comorbid medical conditions） of Asian men with erectile dysfunction （ED）. An integrated analysis of five double-blind, placebo-controlled trials （N = 1 046） was performed. patients were randomly assigned to receive 10 mg tadalafil （N = 185）, 20 mg tadalafil （N = 510）, or placebo （N = 351）. Efficacy assessments included the International Index of Erectile Function （IIEF）, Sexual Encounter profile （SEp） diary and Global Assessment Question （GAQ）. patients receiving 10 mg or 20 mg tadalafil showed significant improvement from baseline-to-end point on the Erectile Function domain of the International Index of Erectile Function （IIEF-EF） domain score in all clinical sub-populations analyzed, compared with patients receiving placebo （p 〈 0.001）. The 10-mg and 20-mg tadalafil groups showed a mean success rate of 64.1% and 70.5% for sexual intercourse attempts （SEp3, successful intercourse）, respectively, compared with 33.4% in the placebo group （p 〈 0.001）, and 85.5% and 85.4% reported improved erections at end point GAQ, respectively, versus 43.5% in the placebo group （p 〈 0.001）. Tadalafil was well tolerated across all groups studied. Headache and back pain were the most frequently reported adverse events. Overall, tadalafil was effective and well tolerated across a diverse clinical spectrum of Asian men with ED.

DNA methylation,a conserved epigenetic mark,is critical for tuning temporal and spatial gene expression.The Arabidopsis thaliana DNA glycosylase/lyase REpRESSOR OF SILENCING 1(ROS1)initiates active DNA demethylation and is required to prevent DNA hypermethylation at thousands of genomic loci.However,how ROS1 is recruited to specific loci is not well understood.Here,we report the discovery of Arabidopsis AGENET Domain Containing protein 3(AGDp3)as a cellular factor that is required to prevent gene silencing and DNA hypermethylation.AGDp3 binds to H3K9me2 marks in its target DNA via its AGD12 cassette.Analysis of the crystal structure of the AGD12 cassette of AGDp3 in complex with an H3K9me2 peptide revealed that dimethylated H3 K9 and unmodified H3 K4 are specifically anchored into two different surface pockets.A histidine residue located in the methyllysine binding aromatic cage provides AGDp3 with pH-dependent H3K9me2 binding capacity.Our results uncover a molecular mechanism for the regulation of DNA demethylation by the gene silencing mark H3K9me2.

Objective:To investigate the antihyperlipidemic effect of crude ethanolic extract of melothria maderaspatana(m.maderaspatana) leaf(CEEm) on deoxycorticosterone acetate(DOCA)-salt hypertensive rats.methods:A midscapular incision was made on each rat and the left kidney was excised after ligation of the renal artery.The surgical wound was closed using an absorbable suture.After one week recovery period,hypertension was induced by subcutaneous injection of DOCA-salt solution,twice a week,and the rats received a 1%sodium chloride solution as drinking water throughout the experimental period.CEEm or nifedipine was administered orally once a day for 6 weeks.Results:In DOCA-salt hypertensive rats,the level of plasma and tissues of total cholesterol(TC),triglycerides(TG),free fatty acids(FFA) and phospholipids (pL) significantly increased and administration of CEEm significantly reduced these parameters towards normality.Further,the levels of low density lipoprotein-cholesterol(LDL-C) and very low density lipoprotein-cholesterol(VLDL-C) significantly increased while high density lipoprotein-cholesterol (HDL-C) decreased in hypertensive rats and administration of CEEm brought these parameters to normality which proved their antihyperlipidemic action.Histopathology of liver, kidney and heart on DOCA-salt induced rats treated with CEEm showed reduced the damages towards normal histology.Conclusions:These findings provided evidence that CEEm was found to be protecting the liver,kidney and heart against DOCA-salt administration and the protective effect could attribute to its antihyperlipidemic activities.

The catalytic hydrogenation of nitrobenzene to aniline employing nickel impregnated on rutile,anatase,and high surface area titania supports has been investigated.The nickel is present in elemental state as fcc phase on the catalyst as evidenced by X-ray diffraction results.The Ni crystallite size was found to be greater for Ni/anatase.The temperature-programmed reduction results suggest a greater metal-support interaction for Ni/rutile.The observed order of catalytic activity for the hydrogenation of nitrobenzene is Ni/rutile > Ni/anatase > Ni/TiO2.A conversion of 99% was observed for Ni/rutile at 140 oC and hydrogen pressure of 1.96 mpa.Interestingly,aniline is the only product formed which demonstrates the catalytic hydrogenation of nitrobenzene proceeds with atom economy.Both Ni/rutile and Ni/anatase exhibited a better stability than Ni/TiO2.The hydrogenation proceeds with the preferential adsorption of hydrogen on nickel present in the catalyst surface,possibly assisted by TiOx species.

microRNAs(miRNAs)play important roles in rice response to magnaporthe oryzae,the causative agent of rice blast disease.Studying the roles of rice miRNAs is of great significance for the disease control.Osa-miR167d belongs to a conserved miRNA family targeting auxin responsive factor(ARF)genes that act in developmental and stress-induced responses.Here,we show that OsamiR167d plays a negative role in rice immunity against m.oryzae by suppressing its target gene.The expression of Osa-miR167d was significantly suppressed in a resistant accession at and after 24 h post inoculation(hpi),however,its expression was significantly increased at 24 hpi in the susceptible accession upon m.oryzae infection.Transgenic rice lines over-expressing Osa-miR167d were highly susceptible to multiple blast fungal strains.By contrast,transgenic lines expressing a target mimicry to block OsamiR167d enhanced resistance to rice blast disease.In addition,knocking out the target gene ARF12 led to hypersusceptibility to multiple blast fungal strains.Taken together,our results indicate that Osa-miR167d negatively regulate rice immunity to facilitate the infection of m.oryzae by downregulating ARF12.Thus,Osa-miR167d-ARF12 regulatory module could be valuable in improvement of blast-disease resistance.

Coronavirus disease 2019(COVID-19)is a global pandemic where several comorbidities have been shown to have a significant effect on mortality.patients with diabetes mellitus(Dm)have a higher mortality rate than non-Dm patients if they get COVID-19.Recent studies have indicated that patients with a history of diabetes can increase the risk of severe acute respiratory syndrome coronavirus 2 infection.Additionally,patients without any history of diabetes can acquire newonset Dm when infected with COVID-19.Thus,there is a need to explore the bidirectional link between these two conditions,confirming the vicious loop between“Dm/COVID-19”.This narrative review presents(1)the bidirectional association between the Dm and COVID-19,(2)the manifestations of the Dm/COVID-19 loop leading to cardiovascular disease,(3)an understanding of primary and secondary factors that influence mortality due to the Dm/COVID-19 loop,(4)the role of vitamin-D in Dm patients during COVID-19,and finally,(5)the monitoring tools for tracking atherosclerosis burden in Dm patients during COVID-19 and“COVID-triggered Dm”patients.We conclude that the bidirectional nature of Dm/COVID-19 causes acceleration towards cardiovascular events.Due to this alarming condition,early monitoring of atherosclerotic burden is required in“Diabetes patients during COVID-19”or“new-onset Diabetes triggered by COVID-19 in Non-Diabetes patients”.

一、前言铜由于具有优异的综合性能,而成为许多工程技术普遍应用的一种金属结构材料.例如,突出的电和热传导性,对腐蚀及氧化的强抵抗力,以及有良好的延展性和可预制成构件等.当然,也有其不足之处,特别在温度较高时,铜的机械强度的相对降低.在电气工业应用中,导电性是最重要的,但也要求在温度较高时,能有一定的强度.常规的增加铜材强度的方法,是采取冷加工和合金热处理,结果在较高温度下仍会因软化而失去强度或降低导电性.
