The effects of bavistin, cefotoxime and kanamycin along with the ethylene inhibitor silver thiosulphate on in vitro regeneration using the axillary buds of Solanum viarum (D.) have been investigated. Axillary bud explants when cultured on mS medium supplemented with bavistin (100 mg/l) maximum number of shoots (4.21 ± 0.83) were observed. in a separate experiment, bavistin along with growth regulators such as BAP and TDZ was added to mS medium containing sucrose (3%). maximum number of shoots was obtained (5.55 ± 0.28) in mS medium supplemented with bavistin (100 mg/l) and BAP (2.0 mg/l). Cefotoxime and kanamycin (10-150 μm/L) were added to mS medium supplemented with BAP (2.0 mg/l) with sucrose (3%) having shown the maximum shoot regeneration (4.38 ± 0.45 and 3.4 ± 0.48) at 80 μm/L and 50 μm/L respectively. Similarly the regeneration medium was supplemented with (5.0-100 μm/L) of STS. maximum number of shoots (4.75 ± 0.43) was seen with 40 μm/L. These plantlets were further maintained for root emergence on a rooting medium supplemented with growth regulators such as (0.5-2.0 mg/l) iAA, iBA and NAA. in mS medium supplemented with iBA (1.0 mg/l), maximum number of roots (24.3 ± 0.31) was seen with 100% regeneration. The rooted plants were acclimatized and transferred to field plots with (95%) of plant surviving in the field.

The paricá [Schizolobium amazonicum Huber ex Ducke] is a native species of the Amazon region, belonging to the Fabaceae family and is a legume that has great social and economic importance, raw material for the manufacture of wood panels. Heliophilous, with wood of light density and good workability. Amazonian soils are highly weathered and characterized by high acidity, so Al harms the growth and development of sensitive plants, as well as the presence of resistant and/or tolerant plants. Thus, in this work, we sought to study the biochemical metabolism alterations generated in young plants of Schizolobium amazonicum Huber ex Ducke affected at different dosages of AlCl3. The research was conducted in a greenhouse of the Federal Rural University of Amazonia, using pre-scarified paricá seeds, which were seeded in plastic containers of 3.5 L capacity, with black soil fertilized with NPK 15-15-20. On the 8th day after sowing (DAS), thinning of 5 seedlings was performed for 2, and on day 12 of 2 for 1 seedling. The treatments started after 1 month and 22 days of sowing and the plants were collected 24 days later. A completely randomized experimental design (RED) was used, with 6 treatments (Control, Al 15 mg/L-1, Al 30 mg/L-1, Al 45 mg/L-1, Al 60 mg/L-1 and Al 75 mg/L-1) with 5 replicates each (one plant/pot) totaling 30 experimental units. The AlCl3 dosages resulted in considerable changes in the biochemical variables evaluated, especially when the 75 mg·L-1 dosage was applied. The considerable reduction in some variables such as nitrate reductase, glycine betaine and total soluble carbohydrates of these compounds to the applied AlCl3·6H2O dosages, is a way to create resistance mechanisms to try to overcome stress or as a way of showing limitations in the applied dosages. Traditionally the evaluation of the cellular infiltrate and protein expression in skin tissue sections is done by manual quantification. However, for reliable evaluation of histology in the development of new anti-psoriatic treatments there is a need for a more time-efficient and reproducible method. To test the use of digital image analysis (dia) in this situation we compared the assessment of immunohistochemically stained skin sections with themore traditionalmanual quantification (mQ) and semiquantitative analysis (SQA). The number of CD3+T cells and the expression of E-selectin were evaluated in stained paired skin biopsie s from 11 patients with chronic plaque psoriasis before and after initiation of anti-psoriasis therapy. We observed significant correlations between mQ and dia for the number of T cells (epiderm is: r=0.88, P ≤0.01, dermis r=0.87, P ≤0.01). Both dia and mQ were equally eff ective in detecting reductions of T-cell numbers in active-treated patients. m Q took 20 h, compared to 6 h for *** also observed significant correlations b etween SQA and dia for the expression of E-selectin (r=0.88, P ≤0.01), althoug h dia was more sensitive than SQA to detect (early) changes. SQA took 10 h, comp ared to 4 h for dia. in conclusion, the quantification of the inflammatory infil trate in psoriatic lesional skin by dia generated similar results as mQ and SQA in a reliable, reproducible and higher time efficient fashion.

impact of Different Light Characteristics on the Growth and Lipid Content of diatom Phaeodactylum tricornutum Transconjugant Strains

Light regulates important metabolic processes in microalgal cells, which can further impact the metabolism and the accumulation of biomolecules such as lipids, carbohydrates, and proteins. Different characteristics of light have been studied on various strains of the model diatom Phaeodactylum tricornutum, but not on transconjugant cells and information on wild-type strains is still limited. Therefore, we studied the impact of different light characteristics such as spectral quality, light intensity and light shift on the growth, and the composition in lipids and fatty acids of P. tricornutum cells to provide a comprehensive context for future applications. initially, we tested the impact of spectral quality and light intensity on P. tricornutum transformed with an episomal vector (Ptev), harboring the resistance gene Sh ble. Results indicated that Ptev cells accumulated more biomass and overall lipids in spectral quality Red 1 (R1: 34% > 600 nm > 66%) more effectively as compared to Red 2 (R2: 8% > 600 nm > 92%). it was also detected that cell granularity was higher in R1 as compared to R2. Furthermore, by testing two light intensities 65 μmol·m-2·s-1 and 145 μmol·m-2·s-1 light, it was observed that 145 μmol·m-2·s-1 led to an increase in growth trend, total biomass and lipid content. Combining spectral qualities and light intensities, we show that the lipid accumulation raised by 2.8-fold. Studying the light intensity and spectral quality allowed us to optimize the light conditions to R1 spectral quality and light intensity 145 μmol·m-2·s-1. These initial results showed that red light R1 at 145 μmol·m-2·s-1 was the best condition for biomass and total lipids accumulation in Ptev cells. Objectives To explore the optimal threshold values of waist circumference （WC） for detecting cardiovascular （CV） risk factors among Chinese children and adolescents. methods Association of WC with CV risk factors was studied among 65 898 children aged 7-18 years whose data were pooled from nine previous studies in China. CV risk factors in this study included hypertension （blood pressure above 95 percentile levels）, dyslipidemia （with one or more of the following three indexes： TG ≥ 1.7 mmol/L, TC ≥ 5.18 mmol/L, and HDL-C ≤ 1.04 mmol/L） and elevated glucose level （fasting plasma glucose ≥5.6 mmol/L）. Receive-operating characteristic analysis （ROC） and logistic regression were employed to derive optimal age- and sex-specific waist circumference references for predicting CV risk factors. Results A slight increasing trend of CV risk factors was observed starting from the 75th percentile of waist circumference in the study population, while a remarkable increasing trend occurred from the 90th percentile. The optimal waist circumference thresholds for predicting high blood pressures were at the 75th percentile for both boys and girls, which was at the 90th percentiles for detecting at least two of the above three CV risk factors. in comparison with children with waist circumference below the 75th percentile, the odds ratio of two CV risk factors doubled among children with waist circumference between the 75th and the 90th percentile, and increased by 6 times among children with waist circumference above the 90th percentile. The trend of high blood pressure increasing with waist circumference remained significant after having been stratified by Bmi category. Conclusion The 75th and the 90th percentiles of WC are the optimal cut-off points for predicting an increased and a substantially increased risk of CV factors in Chinese children and adolescents, respectively.

Context: The epidemic of heart failure has yet to be fully investigated , and d ata on incidence, survival, and sexspecific temporal trends in community-based populations are limited. Objective: To test the hypothesis that the incidence of heart failure has declined and survival after heart failure diagnosis has improved over time but that secular trends have diverged by sex. Design, Setting, and Participants: Population-based cohort study using the resources of the Rochester Epidemiology Project conducted in Olmsted County , minnesota. Patients were 4537 Olmsted County residents (57%women; mean > a ge, 74 > years) with a diagnosis of heart failure between 1979 and 2000. Fram ingham criteria and clinical criteria were used to validate the diagnosis main O utcome measures: incidence of heart failure and survival after heart failure dia gnosis. Results: The incidence of heart failure was higher among men (378/100000 persons; 95%confidence interval i>>, 361-395 for men; 289/100000 persons; 95 %Ci, 277-300 for women) and did not change over time among men or women. After a mean follow-up of 4.2 years (range, 0-23.8 years), 3347 deaths occurred, in cluding 1930 among women and 1417 among men. Survival after heart failure diagno sis was worse among men than women (relative risk, 1.33; 95%Ci, 1.24-1.43) but overall improved over time (5-year age-adjusted survival, 43%in 1979-1984 v s 52%in 1996-2000, Pmen and younger persons experienced larg er survival gains, contrasting with less or no improvement for women and elderly persons. Conclusion: in this community-based cohort, the incidence of heart fa ilure has not declined during 2 decades, but survival after onset of heart failu re has increased overall, with less improvement among women and elderly persons.

Background: While the role of EUS in the evaluation of pancreaticobiliary (PB) disorders in adults is well established, its utility in children remains unproven. This pros-pective study evaluates the feasibility, the safety, and the impact of EUS in the evaluation of PB disorders in children. methods: All children (ion of suspected PB disorders who underwent EUS before scheduled ERCP. The main outcome measure was to evaluate the impact of EUS in the evaluation of PB disorders in children. EUS was considered to have a significant impact if a new diagnosis was established or if the findings altered subsequent management. Results: Fourteen patients (mean age 13 years; range 5- 17 years) underwent 15 EUS procedures over a 3-year period. main indications were the following: acute or recurrent pancreatitis (6 patients), suspected biliary obstruction (5), and abdominal pain suggestive of PB origin (3). EUS diagnosed chronic pancreatitis (3 patients), idiopathic fibrosing pancreatitis (2), carcinoid tumor (1), pancreatic pse-udocyst (1), pancreas divisum (1), choledocholithiasis (1), duodenal duplication cyst (1), and normal (4). dia-gnosis of idiopathic fibrosing pancreatitis and carcinoid tumor was established by EUS-guided FNA. The procedure was successful in all patients, and no complications were encountered. EUS had an impact on patient management in 93% of cases: established new diagnosis (10), precluded need for ERCP (9), and provided additional information that facilitated focused endotherapy (4). A limitation was the small number of enrolled patients and absence of long-term clinical follow-up. Conclusions: EUS and EUS-guided FNA are feasible, safe, and have significant impact that alters subsequent management in the majority of children with PB disorders. Further studies and dissemination of information is required to facilitate its increased application in children.
