In silico identification and characterization of com..n epitope-based peptide vaccine for Nipah and Hendra viruses

Objective: To explore a com..n B-and T-cell epitope-based vaccine that can elicit an im..ne response against encephalitis causing genus Henipaviruses, Hendra virus(He V) and Nipah virus(Ni V). m..hods: m..brane proteins F, G and m..f He V and Ni V were retrieved from..he protein database and subjected to different bioinform..ics tools to predict antigenic B-cell epitopes. Best B-cell epitopes were then analyzed to predict their T-cell antigenic potentiality. Antigenic B-and T-cell epitopes that shared m..im.. identity with He V and Ni V were selected. Stability of the selected epitopes was predicted. Finally, the selected epitopes were subjected to m..ecular docking sim..ation with HLA-DR to confirm..heir antigenic potentiality in silico. Results: One epitope from.. proteins, one from.. proteins and none from.. proteins were selected based on their antigenic potentiality. The epitope from..he G proteins was stable whereas that from.. was unstable. The m..pitope was m..e stable by adding flanking dipeptides. The 15-m.. G-epitope(VDPLRVQWRNNSVIS) showed at least 66% identity with all Ni V and He V G protein sequences, while the 15-m.. m..pitope(GKLEFRRNNAIAFKG) with the dipeptide flanking residues showed 73% identity with all Ni V and He V m..rotein sequences available in the database. m..ecular docking sim..ation with m..t frequent m.. class-II(m.. II) and class-I(m.. I) m..ecules showed that these epitopes could bind within HLA binding grooves to elicit an im..ne response. Conclusions: Data in our present study revealed the notion that the epitopes from.. and m..roteins m..ht be the target for peptide-based subunit vaccine design against He V and Ni V. Red Chittagong Cattle (RCC) is one of the renowned varieties of cattle which is indigenous and m..t potential type of dom..tic anim.. genetic resource in Bangladesh with distinct phenotypic characteristics. Reproductive m..agem..t m..ht be a process for m..tiplying this cattle variety. Therefore, the goals of this study were to characterize the follicular waves of RCC and to determ..e the Progesterone (P4), Follicle Stim..ating Horm..e (FSH) and Luteinizing Horm..e (LH) profile during three consecutive estrus cycle. Follicular wave em..gence (Day 0) was identified using trans-vaginal ultrasound technique and follicular growth was then m..itored from..ay 0 to Day 23 during three consecutive estrus cycle and blood sam..es were collected. After m..suring the growth and diam..er of the follicles, horm..al assays of collected blood sam..es were perform.. using Enzym..Linked Im..nosorbent Assay (ELISA). We found that, both RCC cows and heifers showed two waves inter-ovulatory intervals. The length of inter-ovulatory intervals and the diam..er of the ovulatory follicle of RCC cows and heifers were slightly different from..ach other. The identification of the dom..ant follicle was confirm.. from..he start of its deviation from..ther subordinate follicles till it reached to the m..im.. diam..er and identified as dom..ant follicle which increases linearly for approxim..ely six days (growing phase). This dom..ant follicle is rem..ned approxim..ely sam..size for five days (static phase), and then begins to regress (regressing phase) decreasing the antral diam..er. The follicular waves are first detectable as 4 - 5 m..follicles approxim..ely at day 0 and day 10 both for RCC cows and heifers which showed two waves inter-ovulatory intervals. The circulating varied concentrations of LH, FSH and P4 were determ..ed using ELISA in case of both cows and heifers and the concentration curves followed the sim..ar pattern of change during the estrus cycle. Com..ned Therapy of Pioglitazone and Atorvastatin Alleviate Diabetes in Rats m..e Effectively than That of m..o Therapy

Present research was designated to investigate the hypoglycem.., hypolipidem.. and antioxidant activity of the com..nation of pioglitazone and atorvastatin on long-term..lloxan-induced diabetes rats (AIDRs). The experim..ts were carried out to determ..e blood glucose level, lipid profile, free radial scavenging activities, superoxide dism..ase (SOD) and catalase in liver tissue. In addition, left ventricular (LV) hypertrophy and cardiom..cyte size were also determ..ed by histological analysis. It was found that in short-term..nduction, pioglitazone significantly reduced the blood glucose level without having any considerable effect on lipid profile and antioxidant enzym.. (SOD and catalase) in rats. On the other hand, atorvastatin significantly reduced total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterol (LDL-C) with m..ked increase in the level of high density lipoprotein cholesterol (HDL-C) and im..oved activity of SOD and catalase enzym... However, pathological changes of heart and pancreas were not observed after short-term..xposure to alloxan in rats. Long-term..iabetes induction resulted in LV hypertrophy and prom..ent shrinkage of islets of Langerhans cells. Treatm..t with atorvastatin in com..nation with pioglitazone significantly reduced the LV hypertrophy, TC, TG and LDL level whereas they noticeably increased HDL level, DPPH (1,1-Diphenyl-2-picryl-hydrazyl) free radical scavenging activity, SOD and catalase activity with satisfactory recovery of Langerhans cells. The result dem..strated that com..nation therapy was m..e effective than that of m..o-therapy for preventing diabetes with cardiovascular diseases (CVD) in rats.

Worldwide prevalence of diabetes m..litus has becom..an issue of great concern in current decades. This life threatening disease is associated with worsening of glycem.. control and progressive m..abolic dysfunctions. Objective: Current study aim.. to investigate the effect of hydroxychloroquine (HCQ) as an adjunct to glibenclam..e or m..form.. on glycem.. control in alloxan induced diabetic rats. m..hods: HCQ was com..ned separately with two conventional anti-diabetic drugs;glibenclam..e and m..form... At first, alloxan (120 m..kg) induced diabetic rats were treated with single dose of m..form.. (850 m..70 kg BW), glibenclam..e (10 m..70 kg BW) and HCQ (300 m..70 kg BW) intraperitoneally once daily for two weeks. Then non fixed dose com..nations of glibenclam..e (5 m..70 kg BW) with HCQ (150 m..70 kg BW) and m..form.. (425 m..70 kg BW) with HCQ (150 m..70 kg BW) were injected along with those of the three drugs alone once daily for four weeks. Results: In alloxan induced diabetic rats, glibenclam..e, m..form.. and their com..nation therapies reduced blood glucose level significantly but com..nation therapies are the m..t effective. Glibenclam..e or m..form.. in com..nation with HCQ also significantly (P m..eover, HCQ potentiates the liver glycogen synthesis of m..form.. or glibenclam..e. Conclusion: Outcom.. of this investigation indicate that com..nation of glibenclam..e or m..form.. with HCQ im..oves glycem.. control and provides additional m..abolic benefits, not achieved with either glibenclam..e or m..form.. alone.

Fully fashioned knitwear has a problem..hat the texture and dim..sion of the garm..t, consum..ion and costing cannot be decided until the pattern is sincerely knitted and carried out environs of stipulations involved in the knitting process. The study finds out a total regular consum..ion, costing system..nd its param..ers that are required to m..e a cost sheet of knitwear (sweater). The analysis gives the com..ete m..ner of sweater costing with the recom..nded aid of inspecting the elem..ts that have an im..ct on costs. Required consum..ion and costing techniques such as yarn, trim.. accessories, testing, rate of m..ufacturing and FOB are on hand in this study about exhibiting a special m..el of costing. In term..of hardware, it utilizes a cam..a, a sm..l em..dded processor board, and an Alexa Echo Dot. The software involves an open source text detection library called Tesseract along with Leptonica and OpenCV. The system..n its current version can only work with English text. By using the Am..on cloud web services, a skill set was deployed, which would read aloud the detected text utilizing a OpenCV program..ia the Alexa Echo Dot. For this developm..t, a Raspberry Pi was utilized as the em..dded processor system.
