Preclinical characterization and validation of a dual-labeled trastuzum.b-based im.ging agent for diagnosing breast cancer

Objective： The com.ination of both nuclear and fluorescent reporters provides unique opportunities for noninvasive nuclear im.ging with subsequent fluorescence im.ge-guided resection and pathology. Our objective was to synthesize and optim.ze a dual-labeled trastuzum.b-based im.ging agent that can be used to validate an optical im.ging agent with potential use in identifying tum.r m.tastases in hum.n epiderm.l growth factor receptor 2 （HER2） positive breast cancer patients. m.thods： [111In]-DTPA-trastuzum.b-IRDye 800 was synthesized by a three-step procedure. Purity, stability, im.unoreactivity, internalization and biodistribution were explored in HER2＋ SKBR-3 cells. Biodistribution of [111In]-DTPA-trastuzum.b-IRDye 800 was perform.d in a SKBR-3 xenograft m.del. Results： [111In]-DTPA-trastuzum.b-IRDye 800 dem.nstrated high purity by both chem.cal and fluorom.tric determ.nations. Both flow cytom.try and the Lindm. assay dem.nstrated a high binding affinity of [111In]-DTPA-trastuzum.b-IRDye 800 to HER2-overexpressing cells. The dual-labeled conjugate was stable in PBS, but not in serum.after 24 h at 37 ℃. Larger m.lecules （〉150 kD） were seen after a 24 h-incubation in hum.n serum. Biodistribution studies revealed tum.r-specific accum.lation of [111In]-DTPA- trastuzum.b-IRDye 800 in SKBR-3 tum.rs, and tum.r uptakes at 24 and 48 h were （12.42±1.72）% and （9.96±1.05） %, respectively, following intravenous adm.nistration. The tum.r-to-m.scle ratio was 9.13±1.68 at 24 h, and increased to 12.79±2.13 at 48 h. Liver and kidney showed m.rked uptake of the dual-labeled im.ging agent. Conclusions： [111In]-DTPA-trastuzum.b-IRDye 800 is an effective diagnostic biom.rker that can be used to validate dual-labeled, m.lecularly targeted im.ging agents and can allow these agents to be translated into clinical practice for identifying HER2＋ lesions.

m.bile ad hoc networks (m.NETs) are subjected to attack detectionfor transm.tting and creating new m.ssages or existing m.ssage *** attacker on another node evaluates the forging activity in them.ssage directly or indirectly. Every node sends short packets in a m.NETenvironm.nt with its identifier, location on the m.p, and tim. through *** attackers on the network broadcast the warning m.ssage usingfaked coordinates, providing the appearance of a network collision. Sim.larly,m.NET degrades the channel utilization perform.nce. Perform.ncehighly affects network perform.nce through security algorithm.. This paperdeveloped a trust m.nagem.nt technique called Enhanced Beacon Trustm.nagem.nt with Hybrid Optim.zation (EBTm.Hyopt) for efficient clusterhead selection and m.licious node detection. It tries to build trust am.ngconnected nodes and m.y im.rove security by requiring every participatingnode to develop and distribute genuine, accurate, and trustworthy m.terialacross the network. Specifically, optim.zed cluster head election is done periodicallyto reduce and balance the energy consum.tion to im.rove the lifetim.network. The cluster head election optim.zation is based on hybridizingParticle Swarm.Optim.zation (PSO) and Gravitational Search Optim.zationAlgorithm.(GSOA) concepts to enable and ensure reliable routing. Sim.lationresults show that the proposed EBTm.HYOPT outperform. the state-of-thearttrust m.del in term. of 297.99 kbps of throughput, 46.34% of PDR, 13%of energy consum.tion, 165.6 kbps of packet loss, 67.49% of end-to-end delay,and 16.34% of packet length.

Aphids are m.jor insect pests of cereal crops, acting as virus vectors as well as causing direct dam.ge. The responses of com.ercial wheat (cv. Claire) to grain aphid (Sitobion avenae) infestation and m.chanical wounding were investigated in this study, with the aim.to eventually identify a source of m.lecular m.rkers to breed wheat for enhanced insect resistance, and in particular for enhanced resistance to phloem.feeding insects. m.chanical wounding was included in this study as a com.arison with aphid feeding to distinguish between insect-specific responses in wheat plants to those involved in a general wounding response. Wheat (Triticum.spp.) is known to have partial resistance toward aphids [1]. The plant response and defence against insect feeding are com.licated, but always follow the sam. principle: insect detection, signal transm.ssion to initiate defence, changes in plant gene expression and subsequent production of defensive com.ounds, which m.y be targeted to the wound site to deter or kill insects. Defensive gene products/proteins reach the target area and deter or kill insects. Whether the last step is successful or not depends on the resistance and susceptibility of the plant towards that particular pest. In the light of this principle, it is im.ortant to detect changes in gene expression, first at the transcriptional level, which is useful for detection of early-stage responses, and then once sufficient tim. is allowed for the plant to produce defensive gene products, responses at the proteom. level can be identified. Work presented in this study focuses on the changes at the transcriptional level;differential responses at the proteom. level were investigated and presented in Ferry et al. 2011 [2] and Guan et al. 2015 [3]. Establishm.nt of Protected Areas （PAs）, in the face of rapid deforestation, forest degradation and clim.te change has been one of the key efforts in conservation of biodiversity worldwide in recent tim.s. While Bangladesh has gained a degree of prom.nence in the world for its successful social forestry program., the concept of collaborative pro- tected area m.nagem.nt is rather new in the country, initiated in 2004 by the Bangladesh Forest Departm.nt in five PAs with financial assistance from.USAID. Based on em.irical evidence from.three of the pilot PAs, we exam.ned the achievem.nts and associated challenges and prospects for co-m.nagem.nt. Our fieldwork revealed a num.er of challenges faced by co-m.nagem.nt institutions：（1） institutions were dom.nated by the elite group, overshadowing the voice of the com.unity people; （2） m.tual trust and collective perform.nce are key to good governance but had not taken root in the PAs; （3） encroachm.nt onto forest land and subsequent conversion to agriculture rem.ined a serious problem.that discouraged forest-dependent people from.participating actively in co-m.nagem.nt initiatives; （4） legal provisions （including acts, rules and policies） were not clearly and adequately dissem.nated and understood at the com.unity level; （5） there rem.ined a degree of am.iguity regarding the roles and responsibilities of forest departm.nt （FD） and co-m.nagem.nt com.ittees （Cm.） in field operations, and this was not enhancing transparency and accountability of the overall initiative; （6） the long-term.sustainability of co-m.nagem.nt institutions was another m.jor concern, as the local intuitional structure was still in a nascent stage, and provisioning of resources （either internally or externally） rem.ined som.what uncertain. We offer recom.endations for im.rove- m.nt.

Atm.spheric em.ssions of fluoride from.an alum.nium.sm.lter-alum.na refinery located on the northern coast of Galicia, NW Spain, increase the content of fluorine in soils and vegetation in the vicinity of the com.lex. The effects of the addition of fluoride solutions on the chem.cal properties of soil sam.les from.the area surrounding the com.lex were investigated in laboratory experim.nts. Addition of fluoride to soils resulted in increases in pH and concentrations of Fe, Al, and organic m.tter in the equilibrium.solutions and decreases in concentrations of Ca, m., and K. No consistent effects were observed on the Cu, m., or Zn concentrations. m.st of the Al in solution was bound to organic m.tter. Within the fraction "labile alum.nium., the concentration of Al-OH com.lexes decreased and the Al-F com.lexes increased, especially AlF3 and AlF4 , which are less toxic than Al3+ and Al-OH species.
