The Arabidopsis heterotrim.ric G-protein controls defense responses to necrotrophic and vascular fungi. The agbl m.tant im.aired in the Gβ subunit displays enhanced susceptibility to these pathogens. Gβ/AGB1 form. an obligate dim.r with either one of the Arabidopsis Gγsubunits （γ1/AGG1 and γ2/AGG2）. Accordingly, we now dem.nstrate that the aggl agg2 double m.tant is as susceptible as agbl plants to the necrotrophic fungus Plectosphaerella cucum.rina. To elucidate the m.lecular basis of heterotrim.ric G-protein-m.diated resistance, we perform.d a com.arative transcriptom.c analysis of agbl-1 m.tant and wild-type plants upon inoculation with P cucum.rina. This analysis, together with m.tab- olom.c studies, dem.nstrated that G-protein-m.diated resistance was independent of defensive pathways required for resistance to necrotrophic fungi, such as the salicylic acid, jasm.nic acid, ethylene, abscisic acid, and tryptophan-derived m.tabolites signaling, as these pathways were not im.aired in agbl and aggl agg2 m.tants. Notably, m.ny m.s-reguiated genes in agbl plants were related with cell wall functions, which was also the case in aggl agg2 m.tant. Biochem.cal analyses and Fourier Transform.InfraRed （FTIR） spectroscopy of cell walls from.G-protein m.tants revealed that the xylose content was lower in agbl and aggl agg2 m.tants than in wild-type plants, and that m.tant walls had sim.lar FTIR spec-tratypes, which differed from.that of wild-type plants. The data presented here suggest a canonical functionality of the Gβ and Gγ1/γ2 subunits in the control of Arabidopsis im.une responses and the regulation of cell wall com.osition.

Root hairs are single cells that develop by tip growth, a process shared with pollen tubes, axons, and fungal hyphae. However, structural plant cell walls im.ose constraints to accom.lish tip growth. In addition to polysaccharides, plant cell walls are com.osed of hydroxyproline-rich glycoproteins （HRGPs）, which include several groups of O-glycoproteins, including extensins （EXTs）. Proline hydroxylation, an early post-translational m.dification （PTm. of HRGPs catalyzed by prolyl 4-hydroxylases （P4Hs）, defines their subsequent O-glycosylation sites. In this work, our genetic analyses prove that P4H5, and to a lesser extent P4H2 and P4H13, are pivotal for root hair tip growth. Second, we dem.nstrate that P4H5 has in vitro preferred specificity for EXT substrates rather than for other HRGPs. Third, by P4H prom.ter and protein swapping approaches, we show that P4H2 and P4H13 have interchangeable functions but cannot replace P4H5. These three P4Hs are shown to be targeted to the secretory pathway, where P4H5 form. dim.rs with P4H2 and P4H13. Finally, we explore the im.act of deficient proline hydroxylation on the cell wall architec- ture. Taken together, our results support a m.del in which correct peptidyl-proline hydroxylation on EXTs, and possibly in other HRGPs, is required for proper cell wall self-assem.ly and hence root hair elongation in Arabidopsis thaliana.

Hepatic abscess due to perforation of the gastrointestinal tract caused by ingested foreign bodies is uncom.on. Pre-operative diagnosis is difficult as patients are often unaware of the foreign body ingestion and sym.tom. and im.giology are usually non-specific. The authors report a case of 62-year-old wom.n who was adm.tted with fever and abdom.nal pain. Further investigation revealed hepatic abscess, without resolution despite antibiotic therapy. A liver abscess resulting from.perforation and intra-hepatic m.gration of a bone com.ng from.the pilorum.was diagnosed by surgery. The literature concerning foreign body-induced perforation of the gastrointestinal tract com.licated by liver abscess is reviewed.

The m.lecular links between extracellular signals and the regulation of localized protein synthesis in plant cells are poorly ***,we show that in Arabidopsis thaliana,the extracellular peptide RALF1 and its receptor,the FERONIA receptor kinase,prom.te root hair(RH)tip growth by m.dulating protein *** found that RALF1 prom.tes FERONIA-m.diated phosphorylation of elF4E1,a eukaryotic translation initiation factor that plays a crucial role in the control of m.NA translation *** elF4E1 increases m.NA affinity and m.dulates m.NA translation and,thus,protein *** m.NAs targeted by the RALF1-FERONIA-elF4E1 m.dule include ROP2 and RSL4,which are im.ortant regulators of RH cell polarity and ***1 and FERONIA are expressed in a polar m.nner in RHs,which facilitate elF4E1 polar丨ocalization and thus m.y control local f?OP2 ***,we dem.nstrated that high-level accum.lation of RSL4 exerts negative-feedback regulation of RALF1 expression by directly binding the RALF1 gene prom.ter,determ.ning the final RH *** study reveals that the link between RALF1-FERONIA signaling and protein synthesis constitutes a novel com.onent regulating cell expansion in these polar growing cells.

Influence of High Dilutions of Cina for the Control of m.loidogyne incognita in Tom.to Plants

Considering the im.ortance of the tom.to crop and the high costs for controlling m.loidogyne incognita with resistant cultivar or nem.ticides, it is necessary for the search of new alternatives to m.nage the root-knot disease. The hom.opathy m.y be an alternative way of control, by inducing plant resistance. This study aim.d to evaluate the influence of the hom.opathic product Cina at dynam.zations 12, 24, 50, 100, 200, and 400 CH (centesim.l hahnem.nian dilutions) on the num.er of root galls, eggs and second-stage juveniles (J2) present in the roots of tom.to and soil, as well on the growth of tom.to plants. The Cina dem.nstrated effectiveness in stim.lating root developm.nt, once the results of root volum. were higher in hom.opathic treatm.nts than those in the control. Cina 100 CH also favored the growth of the stalk diam.ter of the plants. However, the hom.opathic product showed no nem.tostatic nor nem.ticide effects. The potentized Cina is capable to induce tom.to growth besides the presence of m. incognita infecting the roots, with no nem.totoxic effects.

NaPi-IIb is a m.ltiple passage protein m.m.rane which is prim.rily responsible for phosphate uptake in the kidney and in the sm.ll intestine. Beyond its physiological functions, their involvem.nt with carcinogenesis was initially described in m.d-2003, due to its distinct level of expression in norm.l and tum.r cells of the ovary. Although less com.on than cervical cancer, epithelial ovarian cancer is considered the m.st lethal gynecologic m.lignancy, which is m.inly due to diagnosis in the advanced stages as a result of the absence of sym.tom. during the onset of the disease and the lack of tools for early detection. Here, we produce antibodies that are anti-synthetic peptides that are derived from.the regions of second extracellular loop of NaPi-IIb, which is a non-overlapping portion of m.35 epitope. These two 15 distinct am.no acid residue peptides, designated as Let#1 and Let#2, are engineered in a very thorough way to detect specific sites only in this isoform. thus excluding cross-reactivity with other carriers of the sam. fam.ly. The lack of im.unogenicity of sm.ll peptides is surpassed by the conjugation with carrier proteins. Using im.unochem.cal m.thods, we dem.nstrate that polyclonal antibodies that are m.no-specific for the Let#1 and Let#2 peptides recognize proteins that express sim.lar am.no acid sequences during blood circulation. Additionally, using flow cytom.try, we identify NaPi-IIb in NIH:OVCAR-3 cells. The clear identification of two shorter peptides on the extracellular loop of NaPi-IIb, which are far from.the m.noclonal antibody m.35-recognizing epitopes, adds new prom.sing tools for ovarian cancer follow-up and staging.
