AIMTo assess KRAS G12D mutation detection by droplet digital PCR (ddPCR) in stool-derived DNA from colorectal cancer (CRC) *** this study, tumor tissue and stool samples were collected from 70 patients with stage I-IV CRC diagnosed by preoperative biopsy. KRAS mutational status was determined by pyrosequencing analysis of DNA obtained from formalin-fixed paraffin-embedded (FFPE) tumor tissues. The KRAS G12D mutation was then analyzed by ddPCR in FFPE tumors and stool-derived DNA from patients with this point mutation. Wild-type (WT) tumors, as determined by pyrosequencing, were included as controls; analysis of FFPE tissue and stool-derived DNA by ddPCR was performed for these patients as *** the total 70 patients included, KRAS mutations were detected by pyrosequencing in 32 (45.71%), whereas 38 (54.29%) had WT tumors. The frequency of KRAS mutations was higher in left-sided tumors (11 located in the right colon, 15 in the left, and 6 in the rectum). The predominant point mutation was KRAS G12D (14.29%, n = 10), which was more frequent in early-stage tumors (I-IIA, n = 7). In agreement with pyrosequencing results, the KRAS G12D mutation was detected by ddPCR in FFPE tumor-derived DNA, and only a residual number of mutated copies was found in WT controls. The KRAS G12D mutation was also detected in stool-derived DNA in 80% of all fecal samples from CRC patients with this point *** is a reliable and sensitive method to analyze KRAS G12D mutation in stool-derived DNA from CRC patients, especially at early stages. This non-invasive approach is potentially applicable to other relevant biomarkers for CRC management.

Background： Leptin has a strong relation to important traits in animal production, such as carcass composition,feed intake, and reproduction. It is mainly produced by adipose cells and acts predominantly in the *** this study, circulating leptin and its gene expression in muscle were evaluated in two groups of young Nellore bulls with divergent feed efficiency. Individual dry matter intake（DMI） and average daily gain（ADG） of 98 Nellore bulls were evaluated in feedlot for 70 d to determinate the residual feed intake（RFI） and select 20 animals for the high feed efficient（LRFI） and 20 for the low feed efficient（HRFI） groups. Blood samples were collected on d 56 and at slaughter（80 d） to determine circulating plasma leptin. Samples of Longissimus dorsi were taken at slaughter for leptin gene expression ***： DMI and RFI were different between groups and LRFI animals showed less back fat and rump fat thickness,as well as less pelvic and kidney fat weight. Circulating leptin increased over time in all animals. Plasma leptin was greater in LRFI on 56 d and at slaughter（P = 0.0049）. Gene expression of leptin were greater in LRFI animals（P = 0.0022） in accordance with the plasma levels. The animals of the LRFI group were leaner, ate less, and had more circulating leptin and its gene ***： These findings demonstrated that leptin plays its physiological role in young Nellore bulls, probably controlling food intake because feed efficient animals have more leptin and lower residual feed intake.

Triptolide (PG-490) induces apoptosis of dendritic cells through sequential p38 MAP kinase phosphorylation and caspase 3 activation

Dendritic cells (DCs) are the most potent antigen-presen ting cells that play crucial roles in the regulation of immune response. Triptol ide, an active component purified from the medicinal plant Tripterygium wilfor dii Hook F., has been demonstrated to act as a potent immunosuppressive drug c apab le of inhibiting T cell activation and proliferation. However, little is known a bout the effects of triptolide on DCs. The present study shows that triptolide d oes not affect phenotypic differentiation and LPS-induced maturation of murine DCs. But triptolide can dramatically reduce cell recovery by inducing apoptosis of DCs at concentration as low as 10 ng/ml, as demonstrated by phosphatidylserin e exposure, mitochondria potential decrease, and nuclear DNA condensation. Tript olide induces activation of p38 in DCs, which precedes the activation of caspase 3. SB203580, a specific kinase inhibitor for p38, can block the activation of caspase 3 and inhibit the resultant apoptosis of DCs. Our results suggest that t he anti-inflammatory and immunosuppressive activities of triptolide may be due, in part, to its apoptosis-inducing effects on DCs.

The Desierto de los Leones National Park is a place that features recreational activities for residents and visitors to Mexico City. The Presa de los Patos (Ducks Dam) is one of the many bodies of water that are present in the park. By means of physico-chemical and microbiological analysis, it was determined that the water of the Presa de los Patos cannot be considered potable. The pH range was between 5.5 - 5.9. The turbidity exceeded the values established in the standards (10.7 - 32.3 Nephelometric Turbidity Units (NTU’s)). Chemical Oxygen Demand (COD) showed that large amounts of oxygen are needed to oxidize organic matter (106 - 450 mg/L). The amount of coliforms and fecal coliforms exceeds the values established by the standards used (total coliforms 98 - 956 Colony Forming Units (CFU’s), fecal coliforms 78 - 807 CFU’s). Five bacterial genera (Escherichia coli, Pseudomona, Klebsiella, Shigella and Salmonella) were isolated and identified. Most of the bacteria isolated were resistant to ampicillin (90%), while 25% were resistant to ciprofloxacin. The water consumption of the Presa de los Patos is a great risk for the habitants and visitors of the park.

There are two species of filarial parasites with sheathless microfilariae known to commonly cause parasitaemias in humans:Mansonella perstans and Mtinsonella *** most contemporary accounts of the distribution of these parasites,neither is usually considered to occur anywhere in the Eastern ***,Sir Patrick Manson,who first described both parasite species,recorded the existence of sheathless sharp-tailed Mansonella ozzardilike parasites occurring in the blood of natives from New Guinea in each and every version of his manual for tropical disease that he wrote before his death in ***'s reports were based on his own identifications and were made from at least two independent blood sample collections that were taken from the *** region Mansonella perstans parasitaemias were also later(in 1923) reported to occur in New Guinea and once before this(in 1905) in *** Mansonella-parasilaernias are generally regarded as benign,they are thought to be of public health importance because they can affect the epidemiological monitoring of other filarial *** this article,we reviewed the historic literature concerning Pacific-origin Mansonella-parasitaemias in an attempt to explain how,despite repeated reports of Pacificregion Mansonella-parasilaemias,by as early as the 1970 s,the WHO had arrived at the presentday view that Wuchereria bancrofti is the only cause of filarial parasitaemias in Papua New *** have also evaluated the evidence supporting the contemporary existence of Pacificarea parasitaemia-causing Mansonella parasites and assessed the relevance such parasites could have for present-day lymphatic filariasis elimination efforts in the region.

Stability of Hydroxytyrosol in Aqueous Solutions at Different Concentration, Temperature and with Different Ionic Content: A Study Using UPLC-MS

The work describes a stability study of hydroxytyrosol (HTyr) in aqueous solutions carried out varying the following parameters: HTyr concentration, temperature and ionic content in the aqueous solution. First, an analytical method based on the use of ultra performance liquid chromatography (UPLC-MS) as analytical technique was developed and validated. The equation of the linear model was A/AISTD= –0.0013 + 0.0117. [HTyr], with a very good linearity, since the R-Squared was 99.9% and the P value of the lack of fit test was higher than 5%. Limits of quantification estimated, CCα(α= 0.05%) and CCβ(β= 0.05%), were 0.2 and 0.4 g/L respectively. As well, the relative standard deviation in the intra-day precision was lower than 3%. Therefore, the method is reproducible. We have applied this methodology to study the stability of HTyr and have observed that the most critical aspect turned to be the ionic content of the aqueous solutions. A tap water with high contents of calcium and magnesium (52 and 21 mg/L) degraded HTyr up to 64% in 48 hours whereas a low mineralization water with low contents of calcium and magnesium (4 and 2 mg/L) produced only 19% degradation.

Effect of Ambient Temperature and Humidity Conditions on the Efficacy of Organic Treatments against Varroa destructor in Different Climatic Zones of Chile

Varroasis is a disease caused by the mite Varroa destructor, which affects honeybees worldwide. It is estimated that it entered Chile in 1992, and it must be controlled through different types of treatments such as thymol, formic and oxalic acid in different formulations and presentations. This kind of organic products has shown a variable acaricidal capacity, since different factors affect their efficacy such as ambient temperature and humidity, product presentation and location inside the hive, etc.. In Chile, beekeeping takes place in four different climatic zones, in which ambient temperature and humidity as well as the duration of the apicultural season are different. Therefore, the aim of this study was to determine how ambient temperature and humidity influence the efficacy of the organic treatments used in the beekeeping season. Different apiaries were installed in those climatic zones in three seasons between the years 2009 and 2012. All beehives received the same management in each apiary and it was evaluated the same organic treatments in different formulations and presentations. All the beehives were positive to Varroa, which was obtained using samples of adult bees and the soapy water test before and after each treatment. A thermo-hygrometer was installed in each apiary in order to monitor the ambient temperature （T°） and humidity （H°） conditions, which were registered daily for a period of 30 days since the beginning. It was obtained that the treatments＇ efficacy increases as the minimum T° and average T° rose, whereas on the contrary, the efficacy decreases when maximum H° was higher.
