Cellular Cd^(2+)fluxes in roots confirm increased Cd availability to rice(Oryza sativa L.)induced by soil acidifications

Soil acidifications become one of the main causes restricting the sustainable development of agriculture and causing issues of agricultural product *** order to explore the effect of different acidification on soil cadmium(Cd)availability,soil pot culture and hydroponic(soil potting solution extraction)were applied,and non-invasive micro-test technique(NMT)was *** three different soil acidification processes were simulated,including direct acidification by adding sulfuric acid(AP1.,acid rain acidification(AP2)by adding artificial simulated acid rain and excessive fertilization acidification by adding(NH_(4))_(2)SO_(4)(AP3).The results showed that for direct acidification(AP1.,DTPA-Cd concentration in field soils in Liaoning(S1.and Zhejiang(S2)increased by 0.1.7-0.21. mg/kg and 0.1.1.0.346 mg/kg,respectively,compared with control *** soil pH decreased by 0.45 units in S1.the Cd content of rice stems,leaves and roots increased by 0.48 to 6.04 mg/kg and 2.58 to 1..84mg/kg,respectively,When the pH value of soil S1.and S2 decreased by 0.20 units,the average velocity of Cd^(2+)at 200μm increased by 1..03-33.1. pmol/cm~2/sec and 21.33-52.86pmol/cm^(2)/sec,respectively,and followed the order of AP3>AP2>*** summary,different acidification measures would improve the effectiveness of Cd,under the same pH reduction condition,fertilization acidification increased Cd availability most significantly.

Geothermal resources are increasingly gaining attention as a competitive,clean energy source to address the energy crisis and mitigate climate *** Wugongshan area,situated in the southeast coast geothermal belt of China,is a typical geothermal anomaly and contains abundant medium-and low-temperature geothermal *** study employed hydrogeochemical and isotopic techniques to explore the cyclic evolution of geothermal water in the western Wugongshan region,encompassing the recharge origin,water-rock interaction mechanisms,and residence *** results show that the geothermal water in the western region of Wugongshan is weakly alkaline,with low enthalpy and mineralization *** hydrochemistry of geothermal waters is dominated by Na-HCO_(3)and Na-SO_(4),while the hydrochemistry types of cold springs are all Na-HCO_(3).The hydrochemistry types of surface waters and rain waters are NaHCO_(3)or Ca-HCO_(3).The δD and δ^(1.)O values reveal that the geothermal waters are recharged by atmospheric precipitation at an altitude between 550.0 and 1.1..6 *** ratios of maj or solutes and isotopic compositions of^(87)Sr/^(86)Sr underscore the significant role of silicate weathering,dissolution,and cation exchange in controlling geothermal water ***,geothermal waters experienced varying degrees of mixing with cold water during their ***δ^(1.)C values suggest that the primary sources of carbon in the geothermal waters were biogenic and ***δ^(34)S value suggests that the sulfates in geothermal water originate from sulfide minerals in the surrounding *** dating using 3H and^(1.)C isotopes suggests that geothermal waters have a residence time exceeding 1.kaBP and undergo a long-distance cycling process.

A virtual thermometer for ultrahigh-temperature-pressure experiments in a large-volume press

Ultrahigh-temperature-pressure experiments are crucial for understanding the physical and chemical properties of *** recent development of boron-doped diamond(BDD)heaters has made such melting experiments possible in large-volume ***,estimates of temperatures above 2600 K and of the temperature distributions inside BDD heaters are not well constrained,owing to the lack of a suitable ***,we establish a three-dimensional finite element model as a virtual thermometer to estimate the temperature and temperature field above 2600 *** advantage of this virtual thermometer over those proposed in previous studies is that it considers both alternating and direct current heating modes,the actual sizes of cell assemblies after compression,the effects of the electrode,thermocouple and anvil,and the heat dissipation by the pressure-transmitting *** virtual thermometer reproduces the power-temperature relationships of ultrahigh-temperature-pressure experiments below 2600 K at press loads of 2.8-7.9 MN(~1. to 28 GPa)within experimental *** temperatures above 2600 K predicted by our virtual thermometer are within the uncertainty of those extrapolated from power-temperature relationships below 2600 ***,our model shows that the temperature distribution inside a BDD heater(1.-26 K/mm along the radial direction and1.0-200 K/mm).Our study thus provides a reliable virtual thermometer for ultrahigh-temperature experiments using BDD heaters in Earth and material sciences.

Repurposing diacerein to suppress colorectal cancer growth by inhibiting the DCLK1.STAT3 signaling pathway

Double cortin-like kinase 1.DCLK1.exhibits high expression levels across various cancers,notably in human colorectal cancer(CRC).Diacerein,a clinically approved interleukin(IL)-1.inhibitor for osteoarthritis treatment,was evaluated for its impact on CRC proliferation and migration,alongside its underlying mechanisms,through both in vitro and in vivo *** study employed MTT assay,colony formation,wound healing,transwell assays,flow cytometry,and Hoechst 33342 staining to assess cell proliferation,migration,and ***,proteome microarray assay and western blotting analyses were conducted to elucidate diacerein’s specific mechanism of *** findings indicate that diacerein significantly inhibits DCLK1.dependent CRC growth in vitro and in *** high-throughput proteomics microarray and molecular docking studies,we identified that diacerein directly interacts with ***,the suppression of p-STAT3 expression following DCLK1.inhibition by diacerein or specific DCLK1.siRNA was ***,diacerein effectively disrupted the DCLK1.STAT3 signaling pathway and its downstream targets,including MCL-1.VEGF,and survivin,thereby inhibiting CRC progression in a mouse model,thereby inhibiting CRC progression in a mouse model.

Molecular mechanism of exopolysaccharide biosynthesis regulated by wze and wzd genes on the immunomodulatory effects of probiotic Lacticaseibacillus paracasei S-NB

The surface macromolecules of probiotic bacteria play crucial roles in modulating immune responses in the host. Exopolysaccharide (EPS) from lactic acid bacteria (LAB) have been widely reported to exhibit immunomodulatory activity. In this study， the EPS biosynthesis gene cluster of Lacticaseibacillus paracasei S-NB was analyzed and a deletion mutant S-NB△7576 (two genes S-NB＿21.5/wze and S-NB＿21.6/wzd were responsible for the chain length determination and export of EPS) was successfully constructed， resulting a 40.02% decrease in the production of EPS. The deletion of wze and wzd had little effect on the monosaccharide composition and major groups of the two EPS fractions (BEPS1.and BEPS2). Both BEPS1.and BEPS2 could inhibit the transcriptional level of tumor necrosis factor-α (TNF-α)， interleukin-1. (IL-1.)， inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) mRNA in RAW 264.7 cells induced by lipopolysaccharide (LPS)， and enhance host immune tolerance via suppressing NF-κB and MAPK signaling. Notably， the S-NB△7576 mutant supplied with the BEPS1.BEPS2 exhibited more significant inhibition of cytokines production and the phosphorylation of p65 and c-Jun N-terminal kinase (JNK) in LPS-stimulated cells compared with the S-NB△7576 mutant alone. Our study provided the immunomodulatory effect of BEPS1.and BEPS2 from L. paracasei S-NB， in which the wze and wzd genes associated with EPS biosynthesis may play an important role.

Lignans are a powerful weapon for plants to resist stresses and have diverse bioactive functions to protect human *** the mechanisms of stereoselective biosynthesis and response to stresses of lignans is important for the guidance of plant ***,we identified the complete pathway to stereoselectively synthesize antiviral(-)-lariciresinol glucosides in Isatis indigotica roots,which consists of three-step sequential stereoselective enzymes DIR1.2,PLR,and ***1.was further identified as the key gene in respoJanuary 2024nse to stresses and was able to trigger stress defenses by mediating the elevation in lignan ***,the phytohormone-responsive ERF transcription factor LTF1.colocalized with DIR1.in the cell periphery of the vascular regions in mature roots and helped resist biotic and abiotic stresses by directly regulating the expression of *** systematic results suggest that DIR1.as the first common step of the lignan pathway cooperates with PLR and UGT71.2 to stereoselectively synthesize(-)-lariciresinol derived antiviral lignans in *** roots and is also a part of the LTF1.mediated regulatory network to resist *** conclusion,the LTF1.DIR1.module is an ideal engineering target to improve plant Defenses while increasing the content of valuable lignans in plants.
