Miniature microbial fuel cells have recently drawn lots of attention as portable power generation devices due to their short startup time and environmentally-friendly process which could be used for powering small integrated biosensors. We designed and fabricated a microbial fuel cell in a microfluidic platform. the device was made in polydimethylsiloxane with a volume of 4 μL and consisted of two carbon cloth electrodes and proton exchange membrane. Shewanella Oneidensis MR-1 was chosen to be the electrogenic bacterial strain and inoculated into the anode chamber. Ferricyanide was used as the catholyte and pumped into the cathode chamber at a constant flow rate during the experiment. the mi- niature microbial fuel cell generated a maximum current of 2.59 μA and had a significantly short startup time.

Chronic viral infections such as hepatitis B virus (HBV), hepatitis C virus (HCV) and human immunodeficiency virus (HIV) are major global health problems affecting more than 500 million people worldwide. Virus-specific CD8+ t cells play an important role in the course and outcome of these viral infections and it is hypothesized that altered or impaired differentiation of virus- specific CD8+ t cells contributes to the development of persistence and/or disease progression. A deeper understanding of the mechanisms responsible for functional differentiation of CD8+ t cells is essential for the generation of successful therapies aiming to strengthen the adaptive component of the immune system.

type VII secretion systems(t7SSs)are found in many disease related bacteria including Mycobacterium tuberculosis(Mtb).ESX-1[early secreted antigen 6 kilodaltons(ESAt-6)system 1]is one of the five subtypes(ESX-1?5)of t7SSs in Mfb,where it delivers virulence factors into host macrophages during ***,little is known about the molecular details as to how this ***,we provide high-resolution crystal structures of the C-terminal AtPase3 domains of EccC subunits from four different Mtb t7SS *** structures adopt a classic RecA-like a/p fold with a conserved Mg-AtP binding *** structure of EccCbl in complex with the C-terminal peptide of EsxB identifies the location of substrate recognition site and shows how the specific signaling module XxxxMxF"for Mtb ESX-1 binds to this site resulting in a translation of the bulge loop.A comparison of all the AtPase3 structures shows there are significant differences in the shape and composition of the signal recognition pockets across the family,suggesting that distinct signaling sequences of substrates are required to be specifically recognized by different t7SSs.A hexameric model of the EccC-AtPase3 is proposed and shows the recognition pocket is located near the central substrate translocation *** diameter of the channel is?25-A,with a size that would allow helix-bundle shaped substrate proteins to bind and pass ***,our work provides new molecular insights into substrate recognition for Mtb t7SS subtypes and also a possible transportation mechanism for substrate and/or virulence factor secretion.

Hepatic ischemia-reperfusion injury (IRI) limits access to transplantation. Heme oxygenase-1 (HO-1) is a powerful antioxidant enzyme which degrades free heme into biliverdin,free iron and carbon monoxide. HO-1 and its metabolites have the ability to modulate a wide variety of inflammatory disorders including hepatic IRI. Mechanisms of this protective effect include reduction of oxygen free radicals,alteration of macrophage and t cell phenotype. Further work is required to understand the physiological importance of the many actions of HO-1 identified experimentally,and to harness the protective effect of HO-1 for therapeutic potential.

Running device drivers as unprivileged user-level code, encapsulated into their own process, has often been proposed as a technique for increasing system robustness. However, in the past, systems based on user-level drivers have generally exhibited poor I/O performance. Consequently, user-level device drivers have never caught on to any significant degree. In this paper we demonstrate that it is possible to build systems which employ user-level device drivers, without significant performance degradation, even for high-bandwidth devices such as Gigabit Ethernet.

男性不育常伴随精子数量减少。Pygo2基因在染色质重塑的伸长精细胞中表达,其功能受损会导致精子形成阻滞和精子生成减少而引发不育。文章旨在检测引起人特发性少精子症和无精子症的Pygo2基因突变。从77例正常生育力男性和195例特发性少精子症和无精子症患者静脉血提取DNA,采用聚合酶链式反应-测序方法对Pygo2基因3个蛋白质编码区进行测序对比,非同义单核苷酸多态性(Single nucleotide polymorphisms,SNPs)位点分别用SIFt、Polyphen-2和Mutation taster软件进行诱发蛋白质结构和表型改变的检测和分析。结果表明,195例患者中,178例(30例轻度或中度少精子症,57例重度少精子症和91例无精子症)基因序列分析报告完好,无精子症中3例患者分别在2个位点(rs61758740,rs141722381)发生了非同义突变SNPs,重度少精子症中1例患者在位点rs61758741发生了非同义突变,3个突变位点在SNPs基因数据库都已有报道,轻度或中度少精子症患者以及正常生育力男性中不存在SNPs。rs61758740可使PYGO2蛋白第141位蛋氨酸(M)变为异亮氨酸(I),rs61758741使PYGO2蛋白第261位碱性赖氨酸(K)变为酸性谷氨酸(E),rs141722381使PYGO2蛋白第240位亲水侧链天冬酰胺(N)变为疏水侧链异亮氨酸(I)。软件分析表明,在所发现的3个SNP非同义突变位点中,rs141722381引起的单个氨基酸改变会导致PYGO2蛋白空间结构破坏和诱发相关疾病。因此,Pygo2基因蛋白质编码序列区SNPs可能是特发性少精子症和无精子症的诱发因素之一,导致男性不育。

Early disease detection is extremely important in the treatment and prognosis of many diseases, especially cancer. Often, proteomic fingerprints and a pattern recognition algorithm are used to classify the pathological condition of a given individual. It has been argued that accurate classification of the existing data implies an underlying biological significance. two fingerprint-based classifiers, decision tree and medoid classification algorithm, and a biomarker-based classifier were examined using a published dataset of mass spectral peaks from 81 healthy individuals and 78 individuals with benign prostate hyperplasia (BPH). For all three methods, classifiers were constructed using the original data and the data after permuting the labels of the samples (BPH and healthy). the fingerprint-based classifiers produced accurate results for the original data, though the peaks used in a given classifier depended upon which samples were placed in the training set. Accurate results were also obtained for the dataset with permuted labels. In contrast, only three unique peaks were identified as putative biomarkers, producing a small number of reasonably accurate biomarker-based classifiers. the dataset with permuted labels was poorly classified. Since fingerprint-based classifiers accurately classified the dataset with permuted labels, the argument for biological significance from a fingerprint-based classifier must be questioned.

A novel dinuclear complex [Co2(atr)3(mal)2(H2O)2]·4H2O(1) (atr = 4-amino-1,2,4-triazole and mal = malonic acid) containing mixed N and O-donor atoms has been prepared and structurally characterized. the structure is made up of the symmetrically 1, 2, 4-triazole bridged [Co2(atr)3(mal)2(H2O)2] moiety and four lattice water molecules. the single crystal X-ray analysis reveals that the complex has a distorted octahedral structure. Magnetic susceptibility measurements reveals that antiferromagnetic interactions exists between the high spin Co(II) ions within the dinuclear unit.
