Characterization of Pro-embryogenic Calli and Somatic Embryogenesis of Byrsonima intermedia A. Juss.

Byrsonima intermedia A. Juss. is a species from the Brazilian Cerrado that produces edible fruits and, in common with other species from the Byrsonima genus, has pharmacological potential. Previous attempts to propagate the species through conventional methods showed difficulties. Thus, the purpose of this work was to characterize pro-embryogenic masses of Byrsonima intermedia callus, aiming for their in vitro propagation through somatic embryogenesis. Leaf segments from in vitro germinated seedlings were employed as explants for callus production. The calli were then subcultured and exposed to dyes to fulfill their embryogenic potential. Digitalizations of the cytological preparations were made in order to measure the area that was stained by both Aceto-Carmine and Evans-Blue, using image tool software. Somatic embryos were induced after treatments with l-naphthaleneacetic acid （NAA）. The percentages of double-colored areas （by Aceto-Carmine and Evans-Blue） were calculated and the data were analyzed by using the Skott-Knott test （P ≤ 0.05） and, the embryogenic callus, as well as the formation of somatic embryos were analyzed by using the Krsuskal-Wallis rank test （P ≤0.05）. The results show that double coloration is effective at identifying cells showing embryogenic potential. Early callus subculture phases show a larger percentage ofembryogenic area （83%） Somatic embryos were induced by using high auxin level.

Enteroparasitoses in Patients with Rheumatoid Arthritis

Background: Patients with rheumatic diseases have profound alterations in the immune system as a result of underlying diseases and the treatments used, which increases the risk of occurrence and severity of infections, including enteroparasitosis. The current treatment for rheumatoid arthritis involves immunosuppressive therapies powerfully needed for screening infectious processes. The prevalence of parasitic infections in rheumatoid arthritis patients is not currently documented in literature. The objective of the current study was to determine the prevalence of parasitic diseases in a population with RA. Methods: We collected demographic and socioeconomic data from 67 patients at the Hospital Universitário de Brasília from July 2015 to April 2016. All patients underwent a parasitological examination of their stool and multiple variables were analyzed using Poisson regression method. Results: The mean age of patients was 53.9 years. They were predominantly in women (94%) and caucasian (47.8%). The mean disease duration was 9.2 years and most patients had the disease in remission or light activity. The prevalence of parasitic infections in these patients was 11.9%, all cases being that of protozoa of the following species: Endolimax nana, Entamoeba histolytica and Entamoeba coli. The final multivariate analysis indicated that the presence of disease had a significant statistical relationship between the presence of enteroparasitosis and rheumatoid arthritis with no fatigue by VAS (p = 0.0488) and best current health index by VAS (p = 0.0012). Conclusion: This study indicates that the prevalence of enteroparasitoses in a population with Rheumatoid Arthritis was 11.9% and all cases were found of protozoa.

ECF Bleaching with Molybdenum Activated Acid Peroxide and its Impact on Eucalyptus Pulp Properties and Effluent Quality

Acid hydrogen peroxide catalyzed with molybdenum (PMo stage) is effective to decrease pulp kappa number and potentially minimize chlorine dioxide demand in subsequent ECF bleaching. This study aimed at developing cost-effective methods for applying PMo as the first stage bleaching for eucalyptus kraft pulp and evaluating their impact on pulp properties and effluent quality in relation to current ECF bleaching techniques. The PMo(EP)DP and A/PMo(EP)DP sequences proved to be most cost-effective for PMo stage application. The pulp properties and effluent quality derived from bleaching with these two sequences were compared with those of the conventional bleaching with the DHT(EP)DP sequence. The PMo(EP)DP sequence produced pulp of refinability and physical properties similar to that of the reference but the A/PMo(EP)DP one resulted in pulp of 20% lower refinability and 10% lower tearing strength. Untreated effluents of the PMo(EP)DP and A/PMo(EP)DP sequences presented similar COD, but lower AOX and color than the DHT(EP)DP effluents. None of the effluents exhibited acute toxicity. The effluents from the PMo(EP)DP sequence showed much lower BOD and BOD/COD ratio than that of the A/PMo(EP)DP one. All effluents were readily treated in bench-scale reactors, and achieved COD removal efficiencies of greater than 70%.

In Silico Evaluation of the Potential Interference of Boceprevir, Calpain Inhibitor II, Calpain Inhibitor XII, and GC376 in the Binding of SARS-CoV-2 Spike Protein to Human Nanobody Nb20

Virtual screening can be a helpful approach to propose treatments for COVID-19 by developing inhibitors for blocking the attachment of the virus to human cells. This study uses molecular docking, recovery time and dynamics to analyze if potential inhibitors of main protease (Mpro) of SARS-CoV-2 can interfere in the attachment of nanobodies, specifically Nb20, in the receptor binding domain (RBD) of SARS-CoV-2. The potential inhibitors are four compounds previously identified in a fluorescence resonance energy transfer (FRET)-based enzymatic assay for the SARS-CoV-2 Mpro: Boceprevir, Calpain Inhibitor II, Calpain Inhibitor XII, and GC376. The findings reveal that Boceprevir has the higher affinity with the RBD/Nb20 complex, followed by Calpain Inhibitor XII, GC376 and Calpain Inhibitor II. The recovery time indicates that the RBD/Nb20 complex needs a relatively short time to return to what it was before the presence of the ligands. For the RMSD the Boceprevir and Calpain Inhibitor II have the shortest interaction times, while Calpain Inhibitor XII shows slightly more interaction, but with significant pose fluctuations. On the other hand, GC376 remains stably bound for a longer duration compared to the other compounds, suggesting that they can potentially interfere with the neutralization process of Nb20.

Comparative Analysis of Two Methods for the Assessment of Aesthetic Impairment Involving Teeth

Objective: To analyze and compare the effectiveness of two methods designed for the assessment of esthetic impairment in patients with missing teeth. Material and Methods: Two methods were addressed in this study: 7-level scale (7LS) and AIPE-Odonto/Brasil. Both tools are designed to assess esthetic dental impairment in civil and criminal Courts. One-hundred and thirteen dentists used the tools to assess dental esthetic impairment in four mock cases (one male and three females). The outcomes of their analyses were compared with pre-established gold-standard scores. Chi-square test was used to investigate the influence of dentists’ sex, age, time of experience and forensic knowledge with their outcomes. Results: dentists’ outcomes were closer to the expected gold-standard scores when AIPE-Odonto/Brasil was used compared to 7LS (p dentists’ sex, time of experience and forensic knowledge were significantly associated with the scores obtained with both tools (p dental impairment involving missing teeth compared to the 7LS.

Marigold(Tagetes erecta): The Potential Value in the Phytoremediation of Chromium

Environmental pollution with chromium （Cr） is harmful to humans, animals and plants, while in plants it causes diminished growth, anatomical alterations and death. In the present study, the potential value of marigold （Tagetes erecta） in the phytoremediation of Cr has been investigated. The randomized experimental design involved the exposure of plants to nutrient solutions containing 0.00, 0.04, 0.08, 0.12, 0.16 or 0.24 mmol L-1 Cr（III）. Chromium toxicity was observed at Cr（III） concentrations ≥ 0.12 mmol L-1 as demonstrated by diminished growth of the aerial parts and reduced density of the root system. Increasing Cr（III） concentrations in the nutrient solution resulted in a higher bioaccumulation of total Cr in the tissues, although translocation from roots to aerial parts was not efficient （maximum value of 25% at 0.12 mmol L-1 Cr（III））. The Cr bioaccumulation was up to 11-fold greater in roots than in the aerial parts. Tagetes erecta exhibited leaf plasticity when exposed to Cr, indicating the existence of a tolerance mechanism to Cr in this species. Chromium caused a reduction in xilem vases, resulting in a plastic effect in T. erecta leaves that increased the metal tolerance in culture solution. Tagetes spp. are potential Cr hyperaccumulators; at Cr（III） concentrations up to 0.12 mmol L-1, the plants accumulated levels above that proposed for hyperaccumulators and still maintained a considerable growth and even flourished. However, this study was conducted in nutrient solution, and studies on species confirmation as Cr hyperaccumulator should be conducted in soils for further clarification.

Optimal Placement of Phasor Measurement Units Using a Modified Canonical Genetic Algorithm for Observability Analysis

This paper proposes a method for optimal placement of synchronized PMUs （phasor measurement units） in electrical power systems using a MCGA （modified canonical genetic algorithm）, which the goal is to determine the minimum number of PMUs, as well as the optimal location of these units to ensure the complete topological observability of the system. In case of more than one solution, a strategy of analysis of the design matrix rank is applied to determine the solution with the lower number of critical measurements. In the proposed method of placement, modifications are made in the crossover and mutation genetic operators, as well as in the formation of the subpopulation, and are considered restrictive hypotheses in the search space to improve the performance in solving the optimization problem. Simulations are performed using the IEEE 14-bus, IEEE 30-bus and New England 39-bus test systems. The proposed method is applied on the IEEE 118-bus test system considering the presence of observable zones formed by conventional measurements.

Prevalence of Low Levels of Vitamin D in Type 2 Diabetes at the City of Mangueirinha, Paraná, Southern Brazil

Background: Diabetes mellitus is a worldwide epidemic. In 2002 there were 173 million diabetic adults worldwide, and these numbers are expected to reach up to 300 million people by 2030. Meanwhile, vitamin D deficiency has its worldwide prevalence directly influenced by factors as solar radiation, skin color, latitude and seasons, cultural habits of populations such as clothing and food, and these factors are important to explain the different prevalences of vitamin D deficiency in the world. Methods: A prospective cross-sectional cohort study was conducted with patients in the outpatient clinic of the Health Unit of the city of Mangueirinha, Paraná, Southern Brazil. Fifty-four type 2 diabetic patients were evaluated (38 women and 16 men), aged 55.8 ± 12.6 years. The following variables were evaluated: age, ethnicity, presence of type 2 diabetes (DM2), hypertension, dyslipidemia, weight, BMI, WC, blood pressure, blood glucose, glycated hemoglobin, ionized calcium, PTH, 25-OH Vit, total cholesterol, HDL, LDL, urea, creatinine, uric acid and red cell/hematocrit. Results: mean BMI was 30.2 ± 4.4 kg/m2, indicating class 1 obesity in this population. Fasting glucose levels were approximately 169.8 ± 74.5 mg/dL. The 25-OH vitamin D values for this population were 23.4 ± 8.3 ng/mL, and 13% of them showed 25-OH vitamin D levels above 30 mg/dL. Fifty percent of those patients had vitamin D levels lower than 30 mg/dL, and 37% had less than 20 mg/dL. Conclusions: this study suggests that vitamin D is associated with low levels of vitamin D in type 2 diabetic patients. Supplementation of vitamin D should be considered in diabetic patients, when levels under 30 mg/mL are found.

The Role of Nitrogen Fertilizers in Sugarcane Root Biomass under Field Conditions

Sugarcane is used worldwide for sugar, ethanol and energy production. In Brazil, the shift from burned to unburned harvest systems resulted in increases in nitrogen fertilization rates, which can impact root architecture and biomass. The expectation is also an increase in sugarcane biomass. The study hypothesized that high N rates applied to sugarcane fields increases root growth and N stored in roots, promoting higher biomass and N accumulated in shoots. Two experiments were set up in Southeastern Brazil, on a Typic Kandiudox (TK) and Rhodic Eutrudox (RE). Four treatments were studied 1) N application in the plant-cane (0 and 120 kg·ha-1 N) and 2) N application in the ratoon (0 and 150 kg·ha-1 N). The shoot biomass and the root density (by the core method up to 0.6 m) were evaluated over the first ratoon crop cycle, and the N content in those compartments was also examined. There was no carry over effect on N applied at planting in root and shoot biomass in the ratoon crop cycle. At the RE site, the ratoon N fertilization increased root density in the superficial soil layer (0 - 0.2 m) and close to the plants (der Brazilian field conditions.
