Pancreatic cancer is one of the most lethal of human malignancies ranking 4th among cancer-related death in the western world and in the United States,and potent therapeutic options are *** during the last few years there have been important advances in the understanding of the molecular events responsible for the development of pancreatic cancer,currently specific mechanisms of treatment resistance remain poorly understood and new effective systemic drugs need to be developed and *** vivo models to study pancreatic cancer and approach this issue remain limited and present different molecular features that must be considered in the studies depending on the purpose to fit special research *** the last few years,several genetically engineered mouse models of pancreatic exocrine neoplasia have been *** models mimic the disease as they reproduce genetic alterations implicated in the progression of pancreatic *** alterations such as activating mutations in KRas,or TGfb and/or inactivation of tumoral suppressors such as p53,INK4A/ARf BRCA2 and Smad4 are the most common drivers to pancreatic carcinogenesis and have been used to create transgenic *** mouse models have a spectrum of pathologic changes,from pancreatic intraepithelial neoplasia to lesions that progress histologically culminating in fully invasive and metastatic disease and represent the most useful preclinical model *** models can characterize the cellular and molecular pathology of pancreatic neoplasia and cancer and constitute the best tool to investigate new therapeutic approaches,chemopreventive and/or anticancer ***,we review and update the current mouse models that reproduce different stages of human pancreatic ductal adenocarcinoma and will have clinical relevance in future pancreatic cancer developments.

Glacier reconstruction of La Covacha Massif in Sierra de Gredos(central Spain) during the Last Glacial Maximum

A paleoglacier reconstruction during the Last Glacial Maximum(LGM) is presented for a high mountain area of La Covacha massif in Sierra de Gredos(Iberian Central System) in central Spain. The moraines that, according with the previous literature, had formed during the global LGM, were mapped through photo interpretation of digital aerial photographs and 3D images and detailed field surveys. The topography of the paleoglaciers was estimated using a simple steady-state model that assumed a perfectly plastic ice rheology, reconstructing the theoretical ice profiles and obtaining the ice thickness of the glaciers during the LGM. The reconstruction of the glaciers was carried out using automated and semi-automated physically-based models, obtaining more realistic results with semi-automated models. According to our study, the paleoglaciers in the study area covered an area of 34.79 km^2 during the global LGM, with a maximum ice thickness of 366 meters in La Vega gorge, a total volume of 34.25 × 108 m^3 and a mean paleoELA of 1932 meters. Most of the ice(~82%) was in paleoglaciers facing north, and the rest was in paleoglaciers with other orientations. This emphasizes the importance of orientation in relation to glacier dynamics and ice accumulation. The results obtained in the calculation of paleoELAs during the global LGM in Gredos are average compared to the Iberian mountains of the Northwest where the values were much lower, and with respect to those of the Southwest, much higher. This demonstrates the importance of the exposure of Mediterranean mountains to the humidity sources coming from the NW during the global LGM, as is the case at present.

Pancreatic cancer is the deadliest solid cancer and currently the fourth most frequent cause of cancer-related deaths. Emerging evidence suggests that cancer stem cells (CSCs) play a crucial role in the development and progression of this disease. The identification of CSC markers could lead to the development of new therapeutic targets. In this study, the authors explore the functional role of c-Met in pancreatic CSCs, by analyzing self-renewal with sphere assays and tumorigenicity capacity in NOD SCID mice. They concluded that c-Met is a novel marker for identifying pancreatic CSCs and c-Met high in a higher tumorigenic cancer cell population. Inhibition of c-Met with XL184 blocks self-renewal capacity in pancreatic CSCs. In pancreatic tumors established in NOD SCID mice, c-Met inhibition slowed tumor growth and reduced the population of CSCs, along with preventing the development of metastases.

In non-cyanogenic species, the main source of cyanide derives from ethylene and camalexin biosyntheses. In mitochondria, cyanide is a potent inhibitor of the cytochrome c oxidase and is metabolized by the β-cyanoalanine synthase CYS-C1, catalyzing the conversion of cysteine and cyanide to hydrogen sulfide and β-cyanoalanine. The hydrogen sulfide released also inhibits the cytochrome c oxidase and needs to be detoxified by the O-acetylserine（thiol）lyase mitochondrial isoform, OAS-C, which catalyzes the incorporation of sulfide to O-acetylserine to produce cysteine, thus generating a cyclic pathway in the mitochondria. The loss of functional OAS-C isoforms causes phenotypic characteristics very similar to the loss of the CYS-C1 enzyme, showing defects in root hair formation. Genetic complementation with the OAS-Cgene rescues the impairment of root hair elongation, restoring the wild-type phenotype. The mitochondria compromise their capacity to properly detoxify cyanide and the resulting sulfide because the latter cannot re-assimilate into cysteine in the oas-c null mutant. Consequently, we observe an accumulation of sulfide and cyanide and of the alternative oxidase, which is unable to prevent the production of reactive oxygen species probably due to the accumulation of both toxic molecules. Our results allow us to suggest that the significance of OAS-C is related to its role in the proper sulfide and cyanide detoxification in mitochondria.

Using adiabatic invariance and the Bohr-Sommerfeld quantization rule we investigate the entropy spectroscopy of two black holes of heterotic string theory,the charged GMGHS and the rotating Sen *** is shown that the entropy spectrum is equally spaced in both cases,identically to the spectrum obtained before for Schwarzschild,Reissner-Nordstr？m and Kerr black *** the adiabatic invariance method does not use quasinormal mode analysis,there is no need to impose the small charge or small angular momentum limits and there is no confusion on whether the real part or the imaginary part of the modes is responsible for the entropy spectrum.
