Phagocytosis: A Practical Approach for Evaluating Neutrophil Function in Health and Disease

Neutrophils, crucial players in the effector phase of the immune response, are recognized as important mediators of both innate and adaptive immune responses. Through the production of pro- and anti-inflammatory cytokines, they modulate the function of T and other lymphoid cells. Countless reports have highlighted the importance of these cells as efficient antimicrobial agents and annotated their involvement in the pathology of infectious and noninfectious diseases. The development of modern, sophisticated technologies has allowed the study of the functions of these cells in clinical settings. These advanced technologies include fluorescence-activated cell sorters, confocal microscopy, automated cell image analyzers, and live cell analysis instruments. Unfortunately, the cost of these modern instruments, maintenance, reagents, and the need for qualified technicians prohibit their use in low-income laboratories and universities in developing countries. With this in mind, we propose a series of basic tests that can be used in low-input clinical laboratories and universities to evaluate the function of neutrophils in health and disease. Our methodology allows us to assess in a practical and low-cost manner the functions of neutrophils in the phagocytic process, including opsonization, ingestion, ROI production (NBT reduction), myeloperoxidase content, phagosome-lysosome fusion, microbicidal activity, and NET production. Thus, under a disadvantageous ambiance, this may guide physicians in deciding whether a patient’s illness involves phagocytic defects without imposing a heavy financial *** Abstract[-rId13-]

Local renin angiotensin system and sperm DNA fragmentation

The renin angiotensin system(RAs)appears to influence male fertility at multiple *** this work,we analyzed the relationship between the RAs and DNA *** male volunteers were divided into two groups(25 each):control(DNA fragmentation≤20%)and pathological(DNA fragmentation>20%)*** of five peptidases controlling RAs were measured fluorometrically:prolyl endopeptidase(which converts angiotensin[A]I and A II to A 1–7),neutral endopeptidase(NEP/CD10:A I to A 1–7),aminopeptidase N(APN/CD13:A III to A IV),aminopeptidase A(A II to A III)and aminopeptidase B(A III to A IV).Angiotensin-converting enzyme(A I to A II),APN/CD13 and NEP/CD10 were also assessed by semiquantitative cytometry and quantitative flow cytometry assays,as were the receptors of all RAs components:A II receptor type 1(AT1R),A II receptor type 2(AT2R),A IV receptor(AT4R or insulin-regulated aminopeptidase[IRAP]),(pro)renin receptor(PRR)and A 1–7 receptor or Mas receptor(MasR)None of the enzymes that regulate levels of RAs components,except for APN/CD13(decrease in fragmented cells),showed significant differences between both *** of RAs receptors revealed no significant differences in immunolabeling patterns between normozoospermic and fragmented *** of AT1R(94.3%normozoospermic vs 84.1%fragmented),AT4R(96.2%vs 95.3%)and MasR(97.4%vs 87.2%)was similar between the ***2R(87.4%normozoospermic vs 63.1%fragmented)and PRR(96.4%vs 48.2%)were higher in non-fragmented *** findings suggest that fragmented DNA spermatozoa have a lower capacity to respond to bioactive RAs peptides.

This research explores the subject of territory,understood based on the life world of Andean inhabitants of the Colca Valley(Arequipa,Peru),who are descendants of ancient local Indigenous groups that,since prehistoric times,have known how to adapt to the rugged geographical reality of the *** a phenomenological approach,which assesses the subjective experience,it was possible to corroborate the existence of routine patterns of spatiality that preserve the emotional essence of past territorial conceptions,which have prevailed despite the hegemonic impositions systematically forced upon the land ever since the spanish conquest in the 16th *** carrying out coexistence fieldwork in situ,five layers of territorial sense have been registered,which,besides being very useful in order to optimize and give coherence to their agricultural and animal husbandry tasks,are periodically represented through verbal and corporal narratives,which have been mapped in this work in order to make their dynamics of symbolization visible and to conclude in the existence of an emotional Andean territory.

This paper reports on the ablation process of a pure Ti solid target immersed in a C-enriched acetone solution, leading to the production of titanium carbide (TiC) and Ti-C core-shell nanostructures. The used route of synthesis is generally called pulsed laser ablation in liquid (PLAL). The presence of carbon structures in the solution contributed to the carbon content in the produced Ti-based nanomaterials. The atomic composition of the produced nanostructures was analyzed using sEM-EDs, while TEM micrographs revealed the formation of spherical TiC and core-shell nanostructures ranging from 40 to 100 nm. The identification of atomic planes by HRTEM confirmed a 10 nm diameter C-shell with a graphite structure surrounding the Ti-core. Raman spectroscopy allowed for the identification of D and G peaks for graphite and a Raman signal at 380 and 600 cmsup>−1sup>, assigned to TiC. The results contribute to the state-of-the-art production of TiC and Ti-C core-shell nanostructures using the PLAL route.

This study presents an analysis of sediments in an Andean river impacted by both natural conditions and anthropogenic activities. Fifty samples were collected from selected sites throughout the Moquegua River drainage basin, and Tambo River headwaters at Pasto Grande, in Peru, and analysed with X-ray diffraction, energy dispersive X-ray spectroscopy, scanning electron microscopy, Fourier-transform infrared spectroscopy and thermogravimetric analysis. si, Ca, Al, Fe, and O, common constituents of soils, were dominant, along with K, N, and P. The latter originating as primary macronutrients or chemical residuals from fertilizers used in agriculture. Higher concentrations of macronutrients and organic components were found in agricultural and urban areas, respectively. Arsenic minerals were also detected, which occur naturally, but possibly at levels augmented by anthropogenic activity. The application of cluster analysis revealed clustering between arsenic, arsenolite, and potassium but no significant geospatial correlation between sample sites in the drainage basin.

Apomorphine effects on the hippocampus

Apomorphine is a non-specific dopamine receptor agonist that has been used in the treatment of some diseases and mental disorders. Its use has particularly well documented in Parkinson＇s disease（PD）. The dopaminergic agonists like apomorphine are related to oxidative processes that could induce cell damage and the functional impairment of some structures in the brain. However, most information about apomorphine in literature is focused on the improvement of the motor problems characteristic of PD, but little is known about the effects on cognitive behaviors and brain structures indirectly related to motor function. The presence of dopaminergic receptors in the hippocampus has recently been discovered, in connection with cognitive behaviors like learning and memory, these receptors are needed in neuronal plasticity. There has been a growing interest to know if this structure could be compromised by the effect of apomorphine and elucidate if part of the cognitive impairment present in the PD is due to the effect of apomorphine. In this mini-review, we summarized how apomorphine has been used since its creation, we discuss the latest information about its effect on the hippocampus and also the future perspectives to fully understand the effects of this compound.
