Targeted therapy has become one of the standards of care for advanced lung *** than 10 genetic aberrations have been discovered that are actionable and several tyrosine kinase inhibitors(TKIs)have been approved to target each of *** several genetic aberrations that are actionable in non-small cell lung cancer(NSCLC),ROS1 translocations also known as gene fusion proteins,are found in only 1%-2%of the patient ***1 mutations can usually be detected using a combination of techniques such as immunohistochemistry(IHC),Fluorescence in-situ testing(FISH),polymerase chain reaction(PCR),and nextgeneration sequencing(NGS).However,RNA NGS and ctDNA NGS(liquid biopsies)also contribute to the *** are currently numerous FDA-approved agents for these tumors,including crizotinib and entrectinib;however,there is in-vitro sensitivity data and clinical data documenting responses to ceritinib and *** responses and survival rates with these agents are frequently among the best compared to other TKIs with genetic aberrations;however,intrinsic or extrinsic mechanisms of resistance may develop,necessitating research for alternative treatment *** combat the mechanisms of resistance,novel agents such as repotrectenib,cabozantinib,talotrectinib,and others are being *** this article,we examine the literature pertaining to patients with ROS1 tumors,including epidemiology,clinical outcomes,resistance mechanisms,and treatment options.

estimation of Contraction Coefficient of Gorlin equation for Assessment of Aortic Valve Area in Aortic Stenosis

Background: The Gorlin equation is the reference method for the assessment of aortic valve area in aortic stenosis and is calculated using a constant, called the coefficient of contraction, which is empirically assumed to be 1. This coefficient is the ratio of effective aortic area to anatomic aortic area, and a value of 1 indicates that both are the same. The purpose of this study was to estimate the actual coefficient of contraction in patients with aortic stenosis and to evaluate its impact on aortic area as calculated by the Gorlin equation. Methods: We studied 17 patients with moderate to severe aortic stenosis. effective aortic area was calculated using the continuity equation. Anatomic aortic area was obtained by planimetry with transesophageal echocardiography. Aortic valve area by the Gorlin equation was calculated from echocardiography data. The coefficient of contraction was derived as above. Results: The coefficient of contraction was inversely related to the pressure recovery. effective area was correlated with anatomic area (r = 0.86, P 2). Aortic area by the Gorlin equation was not correlated with anatomic area, but the correlation became significant when the Gorlin equation was corrected for coefficient of contraction and pressure recovery. Conclusions: Using a coefficient of contraction of 1 in the Gorlin equation gives a poor correlation with anatomic area. Using the calculated coefficient of contraction for each patient and the mean gradient for pressure recovery improves the correlation with anatomic area. These facts could be taken in account when Gorlin equation is considered as the reference method.

Nowadays, the number of cases in which malignant colorectal polyps are removed is increasing due to colorectal cancer screening programmes. Cancerous polyps are classified into non-invasive high grade neoplasia (NHGN), when the cancer has not reached the muscularis mucosa, and malignant polyps, classed as T1, when they have invaded the submucosa. NHGN is considered cured with polypectomy, while the prognosis for malignant polyps depends on various morphological and histological factors. The prognostic factors include, sessile or pedunculated morphology of the polyp, whether partial or en bloc resection is carried out, the degree of differentiation of the carcinoma, vascular or lymphatic involvement, and whether the polypectomy resection margin is tumor free. A malignant polyp at T1 is considered cured with polypectomy ifit is a pedunculated polyp (Ip of the Paris classification), it has been completely resected, it is not poorly differentiated, the resection edge is not affected by the tumor and there is no vascular or lymphatic involvement. The sessile malignant polyp (Is of the Paris classification) at T1 is considered not cured with polypectomy. Only in some cases (e.g. older people with high surgical risk) local excision(polypectomy or endoscopic submucosal dissection or conventional endoscopic mucosal resection) is considered the definitive treatment.

Plant root-nodule symbiosis(RNS)with mutualistic nitrogen-fixing bacteria is restricted to a single clade of angiosperms,the Nitrogen-Fixing Nodulation Clade(NFNC),and is best understood in the legume *** species share many commonalities,explained either by divergence from a common ancestor over 100 million years ago or by convergence following independent origins over that same time ***,comparative analyses of diverse nodulation syndromes can provide insights into constraints on nodulation—what must be acquired or cannot be lost for a functional symbiosis—and the latitude for Plant Communications Genomic landscape of nodulation variation in the ***,much remains to be learned about nodulation,especially outside of ***,we employed a large-scale phylogenomic analysis across 88 species,complemented by 151 RNA-seq libraries,to elucidate the evolution of *** phylogenomic analyses further emphasize the uniqueness of the transcription factor NIN as a master regulator of nodulation and identify key muta-tions that affect its function across the *** transcriptomic assessment revealed nodule-specific upregulated genes across diverse nodulating plants,while also identifying nodule-specific and nitrogen-response *** 70%of symbiosis-related genes are highly conserved in the four representative species,whereas defense-related and host-range restriction genes tend to be lineage *** study also identified over 900000 conserved non-coding elements(CNes),over 300000 of which are unique to sampled NFNC ***-specific CNes are enriched with the active H3K9ac mark and are correlated with accessible chromatin regions,thus representing a pool of candidate regula-tory elements for genes involved in ***,our results provide novel insights into the evolution of nodulation and lay a foundation for engineering of RNS traits in agriculturally important crops.

Acoustic emission characterization of microcracking in laboratory-scale hydraulic fracturing tests

Understanding microcracking near coalesced fracture generation is critically important for hydrocarbon and geothermal reservoir characterization as well as damage evaluation in civil engineering structures. Dense and sometimes random microcracking near coalesced fracture formation alters the mechanical properties of the nearby virgin material. Individual microcrack characterization is also significant in quantifying the material changes near the fracture faces （i.e. damage）. Acoustic emission （Ae） monitoring and analysis provide unique information regarding the microcracking process temporally, and infor- mation concerning the source characterization of individual microcracks can be extracted. In this context, laboratory hydraulic fracture tests were carried out while monitoring the Aes from several piezoelectric transducers. In-depth post-processing of the Ae event data was performed for the purpose of under- standing the individual source mechanisms. Several source characterization techniques including moment tensor inversion, event parametric analysis, and volumetric deformation analysis were adopted. Post-test fracture characterization through coring, slicing and micro-computed tomographic imaging was performed to determine the coalesced fracture location and structure. Distinct differences in fracture characteristics were found spatially in relation to the openhole injection interval. Individual microcrack Ae analysis showed substantial energy reduction emanating spatially from the injection interval. It was quantitatively observed that the recorded Ae signals provided sufficient information to generalize the damage radiating spatially away from the injection wellbore.

Attosecond twisted beams from high-order harmonicgeneration driven by optical vortices

Optical vortices are structures of the electromagnetic field with a spiral phase ramp about a point-phase singularity,carrying orbital angular momentum(OAM). Recently, OAM has been imprinted to short-wavelength radiation through high-order harmonic generation(HHG), leading to the emission of attosecond twisted beams in the extreme-ultraviolet(XUV) regime. We explore the details of the mapping of the driving vortex to its harmonic spectrum. In particular, we show that the geometry of the harmonic vortices is convoluted, arising from the superposition of the contribution from the short and long quantum paths responsible of HHG. Finally, we show how to take advantage of transverse phase-matching to select twisted attosecond beams with different spatiotemporal properties.

Intruder familiarity and not duet similarity influences the territory defense in a year-round territorial bird species

To avoid unnecessary energy expenditures in territorial defense,many species(e.g.,insects,amphibians,birds,and mammals)have developed the capability to distinguish between different intruder types using visual,acoustic,and/or chemical *** the mechanism used for intruder recognition is key to understanding the dynamics of territorial *** birds that use vocalizations for territorial defense,the frequency and duration of vocalizations or the familiarity with the intruder may be the main mechanism used for intruder ***,we conducted a playback experiment with territorial White-eared Ground-sparrows(Melozone leucotis),to analyze if territorial pairs recognize intruders using frequency and duration characteristics(a bird’s response is based on how structurally similar the intruders’duets are to their own)or by familiarity with the intruders(neighbors ***-neighbors).We focused on duets because this species uses duets exclusively for territorial *** broadcasted a duet from a territorial neighbor,two duets from non-neighbors(with different frequency and duration characteristics),and a duet from a control species in 39 territorial pair from three *** playback we measured five behavioral responses:latency of the first vocalization,latency of the approach to the speaker,time spent close the speaker(within 5 m),number of individuals that approached the speaker,and the number of *** found that territorial White-eared Ground-sparrow pairs responded stronger to neighbors than non-neighbors,and this response is not influenced by duet duration or frequency *** result suggests that neighbors represent a greater territorial threat for White-eared Ground-sparrows than *** work is necessary to understand how common this observation is for tropical species that defend small territories year-round.

Background:To finish an endurance race,athletes perform a vigorous effort that induces the release of cardiac damage *** are several factors that can affect the total number of these markers,so the aim of this review was to analyze the effect of endurance running races on cardiac damage markers and to identify the factors that modify the levels of segregation of these cardiac damage ***:A systematic search of PubMed,Web of Science,and the Cochrane Library databases was *** analysis included studies where the acute effects of running races on cardiac damage markers(troponin I and troponin T)were analyzed,assessing the levels of these markers before and after the ***:The effects of running races on troponin I(mean difference=0.0381 ng/mL)and troponin T(mean difference=0.0256 ng/mL)levels were *** ages(R^(2)=14.4%,p=0.033)and body mass indexes(R^(2)=14.5%,p=0.045)of the athletes had a significant interaction with troponin *** addition,gender,mean speed,time to finish the race,and type of race can affect the level of cardiac damage ***:endurance running races induce the release of cardiac-damage markers that remain elevated for at least 24 h after the *** addition,young male athletes with high body mass indexes who perform races combining long duration and moderate intensity(i.e.,marathons)release the highest levels of cardiac damage *** should take into consideration these results in the diagnosis and treatment of patients admitted to the hospital days after finishing endurance running races.
