Taxifolin stability: In silico prediction and in vitro degradation with HpLC-UV/UpLCe ESI-MS monitoring

Taxifolin has a plethora of therapeutic activities and is currently isolated from the stem bark of the tree Larix gmelinni(Dahurian larch). It is a flavonoid of high commercial interest for its use in supplements or in antioxidant-rich functional foods. However, its poor stability and low bioavailability hinder the use of flavonoid in nutritional or pharmaceutical formulations. In this work, taxifolin isolated from the seeds of Mimusops balata, was evaluated by in silico stability prediction studies and in vitro forced degradation studies(acid and alkaline hydrolysis, oxidation, visible/UV radiation, dry/humid heating) monitored by high performance liquid chromatography with ultraviolet detection(HpLC-UV) and ultrahigh performance liquid chromatography-electrospray ionization-mass spectrometry(UpLC-ESI-MS). The in silico stability prediction studies indicated the most susceptible regions in the molecule to nucleophilic and electrophilic attacks, as well as the sites susceptible to oxidation. The in vitro forced degradation tests were in agreement with the in silico stability prediction, indicating that taxifolin is extremely unstable(class 1) under alkaline hydrolysis. In addition, taxifolin thermal degradation was increased by *** the other hand, with respect to photosensitivity, taxifolin can be classified as class 4(stable).Moreover, the alkaline degradation products were characterized by UpLC-ESI-MS/MS as dimers of taxifolin. These results enabled an understanding of the intrinsic lability of taxifolin, contributing to the development of stability-indicating methods, and of appropriate drug release systems, with the aims of preserving its stability and improving its bioavailability.

The intertwined challenges of climate change, resource scarcity, and conflict require innovative integrated solutions that address both environmental and societal vulnerabilities. Technological innovation offers a transformative pathway for climate change adaptation and peacebuilding, with emphasis on a holistic approach to managing resource conflicts and environmental challenges. This paper explores the synergies between emerging technologies and strategic framework to mitigate climate-induced tensions and foster resilience. It focuses on the application of renewable energy systems to reduce dependence on contested resources, blockchain technology to ensure transparency in climate finance, equitable resource allocation and Artificial Intelligence (AI) to enhance early warning systems for climate-related disaster and conflicts. Additionally, technologies such as precision agriculture and remote sensing empower communities to optimize resource use, adapt to shifting environmental conditions, and reduce competition over scares resources. These innovations with inclusive governance and local capacity-building are very primordial. Ultimately, the convergence of technology, policy, and local participation offers a scalable and replicable model for addressing the dual challenges of environmental degradation and instability, thereby paving the way for a more sustainable and peaceful future.

In this study, we examined variability of sun-related energies, auroral electrojet current, ring current, and magnetopause current during solar cycles 23 and 24. The study revealed a dependence of sun-related energies to the Sun and Earth currents systems with solar activity from 1996 to 2019. A decrease in the correlation between sun-related energies and sunspot number was observed over solar cycles 23 and 24 (0.88 for the solar cycle 23 and 0.66 for the solar cycle 24), with a drop in the speed of magnetic disturbances in the solar wind. These results could be attributed to the decrease in Sun’s magnetic field toroidal component magnitude induced by a weak in sunspots number and solar flares during the solar cycle 24. A weak in the Earth currents systems (auroral electrojet current, ring current, and magnetopause current) is also observed. During the decrease in the Earth currents, several peaks are observed, indicating a nonlinear dependence in the Earth currents variation (ring current, auroral electrojet current, and magnetopause current) from solar cycle 23 to solar cycle 24. This could be attributed to the Corotating Interaction Regions (CIRs) observed during the declining phase of solar cycle 23 and the deep minimum preceding solar cycle 24.

This study investigates the variability in cancer diagnosis across different tissues and organs, with a focus on the role of diagnostic methods such as Hematoxylin and Eosin (H&E) staining and immunohistochemistry (IHC). The predominance of female breast cancer (30%) aligns with global trends, underscoring the need for robust diagnostic protocols, particularly in developing regions. Other prevalent cancers, including skin, stomach, and cervix uteri, reflect a mix of environmental, genetic, and infectious factors. The underrepresentation of gallbladder and thyroid cancers (potential underdiagnosis or lower prevalence. Age distribution data indicate peak cancer incidence in individuals aged 31 - 45 years, with gender-specific cancers like breast and cervical cancer predominantly affecting females (63.4%). The analysis also highlights significant diagnostic gaps, as 61.2% of cases did not undergo IHC testing due to resource constraints, leading to potential biases in cancer prevalence and diagnostic accuracy. The study emphasizes the complementary role of IHC in confirming ambiguous H&E findings, with strong alignment observed when both methods were used. However, the absence of IHC in many cases limits the robustness of conclusions, suggesting the need for increased access to IHC testing. The findings advocate for integrating IHC into routine diagnostics, expanding diagnostic capabilities, and improving sample sizes to ensure more reliable and comprehensive cancer data.

In this study we review the occurrence of different types (A, B, C, M, and X classes) of solar flares during different solar cycle phases from 1996 to 2019 covering the solar cycles 23 and 24. During this period, a total of 19,126 solar flares were observed regardless the class: 3548 flares in solar cycle 23 (SC23) and 15,668 flares in solar cycle 24 (SC24). Our findings show that the cycle 23 has observed the highest occurrences of M-class and X-class flares, whereas cycle 24 has pointed out a predominance of B-class and C-class flares throughout its different phases. The results indicate that the cycle 23 was magnetically more intense than cycle 24, leading to more powerful solar flares and more frequent geomagnetic storms, capable of generating significant electromagnetic emissions that can affect satellites and GpS signals. The decrease in intense solar flares during cycle 24 compared to cycle 23 reflects an evolution in solar activity patterns over time.

Developing reliable adaptation and mitigation strategies to combat climate change is necessary at regional and local scales. The present study analyses the ability of the multi-model ensemble (MME) composed of fourteen (14) CORDEX-Africa simulations to capture characteristics of the mean temperature for the present day (1979-2005) and associated extremes over Côte d’Ivoire. For this end, the analysis uses the mean variables of the temperature (i.e., minimum temperature (TMIN), mean temperature (TMEAN) and maximum temperature (TMAX)) as well as associated extremes such as intra-period extreme temperature range (ETR), warm spell duration index (HWFI) and warm days index (TX90p) during January-February-March (JFM), April-May-June (AMJ), July-August-September (JAS) and October-November-December (OND) seasons. The results indicate that mean temperature variables (TMIN, TMEAN and TMAX) are underestimated by CORDEX MME in general, except TMEAN in the centre of Côte d’Ivoire. On the other hand, extreme temperature indices are overestimated over Côte d’Ivoire, except ETR in JAS with an underestimation of about 2˚C and TX90p during JAS in the southern part of the country in JFM, AMJ and OND with an underestimation varying between 1% to 4%. In addition, CORDEX MME and observational datasets (CpC and NCEp) have a significant correlation in simulating temperature variables (TMIN, TMEAN, TMIN), while this correlation is not significant in general for extreme temperature, except ETR and HWFI. Furthermore, extreme temperatures (TX90p and HWFI) are characterized by more important interannual variability in the observations CpC and NCEp for ETR. Moreover, mean temperature variables (TMIN, TMEAN, TMAX) show slight interannual variability with respect to the observations CpC and NCEp, which are characterized by the most variability. Overall, CORDEX MME outperforms the seasonal and spatial variability of the temperature and associated extremes over Côte d’Ivoire, although some biases in
