The highly efficient chemoselectivity,stereoselectivity,and regioselectivity render enzyme catalysis an ideal pathway for the synthesis of various chemicals in broad applications.While the cofactor of an enzyme is necessary but expensive,the conversed state of the cofactor is not beneficial for the positive direction of the reaction.Cofactor regeneration using electrochemical methods has the advantages of simple operation,low cost,easy process monitoring,and easy product separation,and the electrical energy is green and sustainable.Therefore,bioelectrocatalysis has great potential in synthesis by combining electrochemical cofactor regeneration with enzymatic catalysis.In this review,we detail the mechanism of cofactor regeneration and categorize the common electron mediators and enzymes used in cofactor regeneration.The reaction type and the recent progress are summarized in electrochemically coupled enzymatic catalysis.The main challenges of such electroenzymatic catalysis are pointed out and future developments in this field are foreseen.

Immune-mediated liver injury (ILI) is a condition where an aberrant immune response due to various triggers causes the destruction of hepatocytes. Fibroblast growth factor 4 (FGF4) was recently identified as a hepatoprotective cytokine;however, its role in ILI remains unclear. In patients with autoimmune hepatitis (type of ILI) and mouse models of concanavalin A (ConA)- or S-100-induced ILI, we observed a biphasic pattern in hepatic FGF4 expression, characterized by an initial increase followed by a return to basal levels. Hepatic FGF4 deficiency activated the mitochondria-associated intrinsic apoptotic pathway, aggravating hepatocellular apoptosis. This led to intrahepatic immune hyper-reactivity, inflammation accentuation, and subsequent liver injury in both ILI models. Conversely, administration of recombinant FGF4 reduced hepatocellular apoptosis and rectified immune imbalance, thereby mitigating liver damage. The beneficial effects of FGF4 were mediated by hepatocellular FGF receptor 4, which activated the Ca^(2+)/calmodulin-dependent protein kinasekinase 2 (CaMKKβ) and its downstream phosphatase and tensin homologue-induced putative kinase 1 (PINK1)-dependent B-cell lymphoma 2-like protein 1-isoform L (Bcl-XL) signalling axis in the mitochondria. Hence, FGF4 serves as an early response factor and plays a protective role against ILI, suggesting a therapeutic potential of FGF4 and its analogue for treating clinical immune disorder-related liver injuries.

Here,we report a previously unrecognized syndromic neurodevelopmental disorder associated with biallelic loss-of-function variants in the RBM42 gene.The patient is a 2-year-old female with severe central nervous system(CNs)abnormalities,hypotonia,hearing loss,congenital heart defects,and dysmorphic facial features.Familial whole-exome sequencing(WEs)reveals that the patient has two compound heterozygous variants,c.304C>T(p.R102*)and c.1312G>A(p.A438T),in the RBM42 gene which encodes an integral component of splicing complex in the RNA-binding motif protein family.The p.A438T variant is in the RRM domain which impairs RBM42 pro-tein stability in vivo.Additionally,p.A438T disrupts the interaction of RBM42 with hnRNP K,which is the causa-tive gene for Au-Kline syndrome with overlapping disease characteristics seen in the index patient.The human R102*or A438T mutant protein failed to fully rescue the growth defects of RBM42 ortholog knockout△FgRbp1 in Fusarium while it was rescued by the wild-type(WT)human RBM42.A mouse model carying Rbm42 compound heterozygous variants,c.280C>T(p.Q94*)and c.1306_1308delinsACA(p.A436T),demonstrated gross fetal develop-mental defects and most of the double mutant animals died by E13.5.RNA-seq data confirmed that Rbm42 was involved in neurological and myocardial functions with an essential role in alternative splicing(As).Overall,we present clinical,genetic,and functional data to demonstrate that defects in RBM42 constitute the underlying etiology of a new neurodevelopmental disease which links the dysregulation of global AS to abnormal embryonic development.

There are many problems in Social Internet of Things(IoTs),such as complex topology information,different degree of association between nodes and overlapping communities.The idea of set pair information grain computing and clustering is introduced to solve the above problems so as to accurately describe the similarity between nodes and fully explore the multi-community structure.A Set Pair Three-Way Overlapping Community Discovery Algorithm for Weighted Social Internet of Things(WSIoT-SPTOCD)is proposed.In the local network structure,which fully considers the topological information between nodes,the set pair connection degree is used to analyze the identity,difference and reverse of neighbor nodes.The similarity degree of different neighbor nodes is defined from network edge weight and node degree,and the similarity measurement method of set pair between nodes based on the local information structure is proposed.According to the number of nodes'neighbors and the connection degree of adjacent edges,the clustering intensity of nodes is defined,and an improved algorithm for initial value selection of k-means is proposed.The nodes are allocated according to the set pair similarity between nodes and different communities.Three-way community structures composed of a positive domain,boundary domain and negative domain are generated iteratively.Next,the overlapping node set is generated according to the calculation results of community node membership.Finally,experiments are carried out on artificial networks and real networks.The results show that WSIoT-SPTOCD performs well in terms of standardized mutual information,overlapping community modularity and F1.

心算是生活中常使用到的技能,涉及到多种引起大脑活动变化认知加工环节,对于心算的脑电(Electroencephalogram,EEG)研究有助于提高对认知任务的研究水平。本文提出了一种全局图振幅排列熵(Amplitude permutation entropy for global graph,APEGG)应用于脑电心算研究,以弥补传统图排列熵(Permutation entropy for graph,PEG)无法全面反映脑网络节点周边邻居节点变化的缺陷,克服了脑电信号幅值不敏感的问题。首先采用相位锁定值构建了EEG脑网络,分析多导联脑电信号之间的同步性和相关性,然后计算了不同频段下脑网络的全局图振幅排列熵,最后运用支持向量机(Support vector machine,SVM)进行分类。使用脑电心算公开数据集进行仿真,分析了不同频段的心算状态与静息状态的熵值散点图,两种状态的熵值散点图表现出较大差异。心算状态与静息状态分类结果与其他算法比较表现出更好的效果。

现有导电凝胶材料在实际使用时存在诸多缺陷,因此设计出一种兼具高强度、高黏附性和高导电性的凝胶材料具有重要的应用意义。本文以丙烯酸(AAc)和丙烯酸-N-羟基琥珀酰亚胺酯(AAc-NHS ester)为基体,聚乙二醇二丙烯酸酯(PEGDA)为交联剂,引入氧化石墨烯(GO)作为增效成分,通过光引发一步聚合法制备了复合凝胶材料(PAA-NHS/GO)。GO的引入提高了体系的交联度,增强了凝胶的稳定性并赋予了凝胶高导电特性。PAA-NHS/GO的拉伸强度、断裂伸长率、电导率分别可达0.336MPa、302.6%和0.93S/m。复合凝胶的功能基团能与组织表面产生强黏附效果,黏附强度超过40kPa,同时还具备高生物安全性和优异的生物相容性。在外力作用下,复合凝胶表现出稳定的应变传感特性。而在用于心电信号监测时,复合凝胶的灵敏度高,并可实现稳定清晰的信号传输。PAA-NHS/GO作为新型导电黏附凝胶,有望用于柔性可穿戴、生物传感、电子皮肤等领域。

Evaluation of optimized continuous venovenous hemodiafiltration therapy efficiency in severe burn patients with sepsis

As an initial factor,sepsis and multiple organ dysfunction syndrome(MODS)caused by sepsis are the principal causes of death in burned patients. In this report, we measured the levels of tumor necrosis factor (TNF)-α, interleukin (IL)-6 and IL-8 in severely burned patients with sepsis after the initiation of continuous vein-vein hemodiafiltration (CVVHDF) to evaluate the clinical usefulness of CVVHDF on the removal of key mediators. The vital sign indices, such as the heart rate (HR), respiration (R) and central venous pressure (CVP), were recorded at 0 and 42 h in each group. Further, the laboratory examinations indexes, such as the white blood cel count, blood sugar, se-rum sodium, blood urea nitrogen and serum creatinine, were detected in venous blood samples. Twenty-two severely burned patients suffering from sepsis were randomized into the control group (A, n = 11) and the experimental group (B, n = 11). The patients in group A underwent conventional treatment, and those in group B received conventional+CVVHDF treatment. The vital signs, such as the HR, R, and CVP, and laboratory examination indices, such as the blood cel count, blood sugar, serum sodium, blood urea nitrogen, and serum creatinine, dropped significantly in group B compared with those in group A at 42 h (P < 0.05). The plasma levels of TNF-α, IL-6 and IL-8 were measured at 0, 12, 18, 24, 36 and 42 h after the start of CVVHDF and at the same time points after the patients were diagnosed with sepsis in group A. The plasma levels of TNF-α in group B decreased by 32% at 18 h after the start of CVVHDF and decreased by 43%at 42 h after the start of CVVHDF;however, these levels were increased compared with the normal values (P < 0.01). The plasma levels of IL-6 decreased at 18 h after the start of CVVHDF (0.274 ± 0.137 ng/ml). Fol owing a brief increase at 24 h, the plasma levels of IL-6 again decreased continuously until the end of the investigation (0.192 ± 0.119 ng/ml). 透射电镜生物样品制备过程中,因青稞籽粒结构致密、细胞壁厚、试剂渗透性差,使用常规样品制备方法会造成制样失败,无法对籽粒超微结构进行观察。为解决青稞籽粒制样问题,将高压处理引入戊二醛固定和树脂浸透两个步骤中,并对常规样品制备方法中戊二醛固定液、锇酸固定液、树脂浸透液浓度进行相应调整。通过对不同试剂浓度和不同压强处理的结果进行观察和比较,选定实验范围,通过正交试验确定优化实验条件。结果表明,青稞籽粒透射电镜样品制备的较优前处理条件:体积分数为4.5%的戊二醛,质量浓度为15 g/L的锇酸,过夜处理的树脂浸透液中丙酮与环氧树脂体积比为1∶1,戊二醛固定和树脂浸透的高压处理压强为50 MPa,处理时间为10 min。在该处理条件下制备的青稞籽粒观测样品表皮细胞壁完整、无断裂,糊粉层各组织完整、结构清晰、无空洞,胚乳层细胞结构完整、形状规则,淀粉粒无缺失和褶皱。将该样品用于实验,结果稳定、可重复,可用于青稞籽粒透射电镜样品制备。

“双碳”视野下的乡村景观研究与实践是当代风景园林高质量发展的重要面向,乡村景观碳源碳汇量化解析与制图分析是其研究基础与技术途径,以面积系数法构建的乡村景观碳模型及其测度技术研究亟待开展。基于“三生空间”景观要素进行乡村碳源碳汇因子聚类,建立碳源碳汇清单并计算景观要素基础碳量,依托GIS平台关联对应面积、获取碳系数,建立乡村景观碳模型。以直山村为例,根据景观要素SHP图形数据库将碳模型进行制图表达,实现乡村景观要素的碳量集成与测度解析。相关研究为乡村景观要素碳源碳汇联立分析提供了一种可借鉴、可发展的技术框架与测度方法,为乡村景观碳模型系统建构与纵深发展提供研究基础。
