Enhancing m^(6)A modification in the motor cortex facilitates corticospinal tract remodeling after spinal cord injury

Spinal cord injury typically causes corticospinal tract disruption. Although the disrupted corticospinal tract can self-regenerate to a certain degree, the underlying mechanism of this process is still unclear. N6-methyladenosine(m^(6)A) modifications are the most common form of epigenetic regulation at the RNA level and play an essential role in biological processes. However, whether m^(6)A modifications participate in corticospinal tract regeneration after spinal cord injury remains unknown. We found that expression of methyltransferase 14 protein(METTL14) in the locomotor cortex was high after spinal cord injury and accompanied by elevated m^(6)A levels. Knockdown of Mettl14 in the locomotor cortex was not favorable for corticospinal tract regeneration and neurological recovery after spinal cord injury. Through bioinformatics analysis and methylated RNA immunoprecipitation-quantitative polymerase chain reaction, we found that METTL14 regulated Trib2 expression in an m^(6)A-regulated manner, thereby activating the mitogen-activated protein kinase pathway and promoting corticospinal tract regeneration. Finally, we administered syringin, a stabilizer of METTL14, using molecular docking. Results confirmed that syringin can promote corticospinal tract regeneration and facilitate neurological recovery by stabilizing METTL14. Findings from this study reveal that m^(6)A modification is involved in the regulation of corticospinal tract regeneration after spinal cord injury.

Convertible hydrogel supercapacitors have emerged as promising energy storage devices in switches,diodes, and transistors. However, inherent weaknesses in ionic conductivity, mechanical properties,and water retention of hydrogel electrolytes seriously hinder their development. Inspired by the hardness conversion of sea cucumber skin, a conductivity and mechanics dual-tunable salt gel electrolyte is successfully designed. The salt gel presents a reversible switching of conductors-insulators and a mechanical regulation between softness and hardness via the dissolution-crystallization transition of sodium acetate trihydrate（SAT）. Meanwhile, the salt gels spontaneously grow a layer of “armor” through saturated phase-change salt crystals effectively reducing water evaporation of hydrogel ***, this phase-change soft-rigid conversion strategy will expand the capabilities of gel-based flexible supercapacitors(area capacitance: 258.6 mF cm-2), and the capacitance retention rate could still reach 86.9% after 3000 cycles at high temperatures. Moreover, the salt gel supercapacitor is potentially used in over-heat alarm systems. It is anticipated that the strategy of conductivity and mechanics of dual-tunable salt gel would provide a new perspective on the development of energy storage devices,wearable electronics, and flexible robots.

Tanycytes, specialized ependymal cells located in the hypothalamus, play a crucial role in the generation of new neurons that contribute to the neural circuits responsible for regulating the systemic energy balance. The precise coordination of the gene networks controlling neurogenesis in naive and mature tanycytes is essential for maintaining homeostasis in adulthood. However, our understanding of the molecular mechanisms and signaling pathways that govern the proliferation and differentiation of tanycytes into neurons remains limited. This article aims to review the recent advancements in research into the mechanisms and functions of tanycyte-derived neurogenesis. Studies employing lineage-tracing techniques have revealed that the neurogenesis specifically originating from tanycytes in the hypothalamus has a compensatory role in neuronal loss and helps maintain energy homeostasis during metabolic diseases. Intriguingly,metabolic disorders are considered early biomarkers of Alzheimer's disease. Furthermore,the neurogenic potential of tanycytes and the state of newborn neurons derived from tanycytes heavily depend on the maintenance of mild microenvironments, which may be disrupted in Alzheimer's disease due to the impaired blood–brain barrier ***, the specific alterations and regulatory mechanisms governing tanycyte-derived neurogenesis in Alzheimer's disease remain unclear. Accumulating evidence suggests that tanycyte-derived neurogenesis might be impaired in Alzheimer's disease, exacerbating neurodegeneration. Confirming this hypothesis, however, poses a challenge because of the lack of long-term tracing and nucleus-specific analyses of newborn neurons in the hypothalamus of patients with Alzheimer's disease. Mangroves are crucial to the ecological security of the Earth and human well-being. Their management, conservation, and restoration are of great importance and necessitate the support of spatio-temporal information and multidisciplinary knowledge such as biology and ecology. Traditional knowledge services such as plant atlas provide illustrated textual knowledge of mangroves. However, this kind of service is oriented to information retrieval and is incapable of effectively mining and utilizing fragmented knowledge from multi-source heterogeneous data, facing the problem of “massive data, rare knowledge”. Knowledge graphs are capable of extracting, organizing, and fusing the knowledge contained in massive data into semantic networks that can be understood and computed by computers. They provide a solution for the realization of intelligent knowledge services. Focusing on the urgent need for mangrove knowledge acquisition, formal representation, and intelligent services, this paper proposes a research prospect on mangrove knowledge graphs and knowledge services. We first analyze the similarities and differences between various domain-specific concepts of Tupu. On this basis, we define the mangrove knowledge graph as a large-scale knowledge base that integrates multi-disciplinary knowledge and spatio-temporal information with mangrove ecosystems as the core. Then, we propose a research framework for mangrove knowledge services that can realize the transformation from multi-modal data to intelligent knowledge services, including multiple research levels such as ubiquitous data sensing and aggregation, knowledge organization and graph construction, and intelligent mangrove knowledge services. Subsequently, the methods and workflow for constructing mangrove knowledge graphs are introduced. An efficient quantum proactive incremental learning algorithm

In scenarios where a large amount of data needs to be learned, incremental learning can make full use of old knowledge, significantly reduce the computational cost of the overall learning process, and maintain high performance. In this paper, taking the MaxCut problem as our example, we introduce the idea of incremental learning into quantum computing, and propose a Quantum Proactive Incremental Learning algorithm(QPIL). Instead of a one-off training of quantum circuit, QPIL contains a multi-phase training on gradually-increased subgraphs of all vertices, proactively reducing large-scale problems to smaller ones to solve in steps, providing an efficient solution for MaxCut. Specifically, some vertices and corresponding edges are randomly selected for training to obtain optimized parameters of the quantum circuit at first. Then, in each incremental phase, the remaining vertices and corresponding edges are gradually added and the parameters obtained from the previous phase are reused in the parameter initialization of the current phase. We perform experiments on 120 different small-scale graphs, and it shows that QPIL performs superior to prevalent quantum and classical baselines in terms of approximation ratio(AR), time cost, anti-forgetting, and solving stability. In particular, QPIL's AR surpasses 20% of mainstream quantum baselines, and the time cost is less than 1/5 of them. The idea of QPIL is expected to inspire efficient and high-quality solutions in large-scale Max Cut and other combinatorial optimization problems.
