Diazoxide preconditioning plus subsequent hypothermia increased resistance of rat cultured hippocampal neurons against hypoxia-reoxygenation injury

Background Cerebral ischemia-reperfusion/hypoxia-reoxygenation insult triggers lots of pathophysiological and biochemical events that separately affect the evolution of cerebral damage. Accordingly, all known effective neuroprotective measures should be taken to get the optimal efficacy of therapy. This study was undertaken to investigate whether diazoxide （DZ） preconditioning combined with the following hypothermia could contribute to synergistic neuroprotection compared with either hypothermia or DZ preconditioning alone. Methods Cultured for 9-10 days in vitro, the hippocampal neurons of SD rats were preconditioned with DZ 0 pmol/L or DZ 250 pmol/L for 1 hour per day and this treatment lasted for 3 days. Subsequently, neurons were subjected to deprivation of oxygen for 4 hours at 37°0, 34°C, 30℃ and 22℃, respectively. This experiment consisted of 8 groups （4 temperature groups and 4 combination groups） and each group contained 12-well or 2-dish cells. Survival rate, expression of Bcl-2, fluorescence magnitude of intracellular calcium, and concentration of malondialdehyde （MDA） were determined at 24 hours after reoxygenation. Results The survival rate and expression of Bcl-2 were both increased in individually hypothermic conditions compared with those at 370G （P〈0.05）, whereas intracellular calcium and MDA did the opposite exhibition simultaneously （P〈0.05）. 22℃ contributed to a higher survival rate and greater expression of Bcl-2 in comparison with other hypothermia （P〈0.05）. Preceding administration of 250 pmol/L DZ took the similar effects on the neurons like hypothermia. Moreover, compared with individual hypothermia or DZ preconditioning, the neuronal survival rate and expression of Bcl-2 in the combination group were increased significantly （P〈0.05）, whereas the calcium fluorescence density and concentration of MDA were reduced further （P〈0.05）. 250 Iamol/L DZ preconditioning combined with 22℃ provided a maximal neuroprotection. Ototoxic destruction by co-administration of kanamycin and ethacrynic acid in rats

It is well known that ethacrynic acid (EA) can potentiate the ototoxicity of aminoglycoside antibiotics (AmAn) such as kanamycin (KM),if they were applied at the same ***,to create the model of EA-KMinduced cochlear lesion in rats,adult rats received a single injection of EA (75 mg/kg,intravenous injection),or followed immediately by KM (500 mg/kg,intramuscular injection).The hearing function was assessed by auditory brainstem response (ABR) measurement in response to click and/or tone bursts at 4,8,12,16,20,24,and 32 *** static microcirculation status in the stria vascularis after a single EA injection was evaluated with eosin *** pathological changes in cochlear and vestibular hair cells were also quantified after co-administration of EA and *** a single EA injection,blood flow in vessels supplying the stria vascularis rapidly ***,the blood supply to the cochlear lateral wall partially recovered 5 h after EA *** changes in ABR were basically parallel to the microcirculation changes in stria vascularis after single EA ***,disposable co-administration of EA and KM resulted in a permanent hearing loss and severe damage to the cochlear hair cells,but spared the vestibular hair *** the cochlear lateral wall is the important part of the blood-cochlea barrier,EA-induced anoxic damage to the epithelium of stria vascularis may enhance the entry of KM to the ***,experimental animal model of selective cochlear damage with normal vestibular systems can be reliably created through co-administration of EA and KM.

Genome Alignment Spanning Major Poaceae Lineages Reveals Heterogeneous Evolutionary Rates and Alters Inferred Dates for KeyEvolutionary Events

Multiple comparisons among genomes can clarify their evolution, speciation, and functional innova- tions. To date, the genome sequences of eight grasses representing the most economically important Poaceae （grass） clades have been published, and their genomic-level comparison is an essential foundation for evolutionary, functional, and translational research. Using a formal and conservative approach, we aligned these genomes. Direct comparison of paralogous gene pairs all duplicated simultaneously reveal striking variation in evolutionary rates among whole genomes, with nucleotide substitution slowest in rice and up to 48% faster in other grasses, adding a new dimension to the value of rice as a grass model. We reconstructed ancestral genome contents for major evolutionary nodes, potentially contributing to understanding the divergence and speciation of grasses. Recent fossil evidence suggests revisions of the estimated dates of key evolutionary events, implying that the pan-grass polyploidization occurred ~96 million years ago and could not be related to the Creta- ceous-Tertiary mass extinction as previously inferred. Adjusted dating to reflect both updated fossil evidence and lineage-specific evolutionary rates suggested that maize subgenome divergence and maize-sorghum divergence were virtually simultaneous, a coincidence that would be explained if poly- ploidization directly contributed to speciation. This work lays a solid foundation for Poaceae transla- tional genomics.

RAP1 is a well-known telomere-binding protein, but its functions in human stem cells have remained unclea匚 Here we generated RAP1 -deficient human embryonic stem cells (hESCs) by using CRISPR/Cas9 technique and obtained RAP1-deficient human mesenchymal stem cells (hMSCs) and neural stem cells (hNSCs) via directed differentiation. In both hMSCs and hNSCs, RAP1 not only negatively regulated telomere length but also acted as a transcriptional regulator of RELN by tuning the methylation status of its gene promoter. RAP1 deficiency enhanced self-renewal and delayed senescence in hMSCs, but not in hNSCs, suggesting complicated lineage-specific effects of RAP1 in adult stem ***, these results demonstrate for the first time that RAP1 plays both telomeric and nontelomeric roles in regulating human stem cell homeostasis.

Application of constructed wetlands for wastewater treatment in tropical and subtropical regions(2000–2013)

Constructed wetlands（CWs） have been successfully used for treating various wastewaters for decades and have been identified as a sustainable wastewater management option for developing countries. With the goal of promoting sustainable engineered systems that support human well-being but are also compatible with sustaining natural（environmental）systems, the application of CWs has become more relevant. Such application is especially significant for developing countries with tropical climates, which are very conducive to higher biological activity and productivity, resulting in higher treatment efficiencies compared to those in temperate climates. This paper therefore highlights the practice,applications, and research of treatment wetlands under tropical and subtropical conditions since 2000. In the present review, removal of biochemical oxygen demand（BOD） and total suspended solid（TSS） was shown to be very efficient and consistent across all types of treatment wetlands. Hybrid systems appeared more efficient in the removal of total suspended solid（TSS）（91.3%）, chemical oxygen demand（COD）（84.3%）, and nitrogen（i.e.,80.7% for ammonium（NH）4-N, 80.8% for nitrate（NO）3-N, and 75.4% for total nitrogen（TN））as compared to other wetland systems. Vertical subsurface flow（VSSF） CWs removed TSS（84.9%）, BOD（87.6%）, and nitrogen（i.e., 66.2% for NH4-N, 73.3% for NO3-N, and 53.3% for TN）more efficiently than horizontal subsurface flow（HSSF） CWs, while HSSF CWs（69.8%）showed better total phosphorus（TP） removal compared to VSSF CWs（60.1%）. Floating treatment wetlands（FTWs） showed comparable removal efficiencies for BOD（70.7%）,NH4-N（63.6%）, and TP（44.8%） to free water surface（FWS） CW systems.

黄曲霉毒素M1(aflatoxin M1,AFM1)对人类和动物具有致癌性、诱变性、致畸性等,广泛存在于哺乳动物乳汁及其乳制品中。因此,制备特异性强、灵敏度高的抗体来检测AFM1至关重要。该研究以AFM1-BSA为抗原免疫双峰驼,采血分离淋巴细胞,提取总RNA反转录为cDNA,通过巢式PCR获得重链可变区(variable domain of heavy chain,VHH),与pComb3XSS载体连接电击转化至Escherichia coli TG1,构建AFM1特异性噬菌体展示文库。以AFM1-OVA为靶向抗原,使用4轮酸洗脱和竞争洗脱相结合的策略对文库进行淘选,并对抗AFM1纳米抗体进行初步鉴定,最后预测了精准的三维模型结构。结果显示,抗AFM1纳米抗体文库库容量为2.27×10^(9)CFU,多样性丰富,插入率为100%。经淘选共获得6条独特序列的纳米抗体。其中VHH-M4和VHH-M6具有较高的灵敏度,初步测定半抑制浓度分别为0.415 ng/mL和1.877 ng/mL,线性范围分别为0.141~1.652 ng/mL和0.800~3.811 ng/mL。一级和二级结构结果显示VHH-M4和VHH-M6均为亲水性蛋白,框架区(framework region,FR)和互补决定区(complementarity determining region,CDR)区符合典型纳米抗体的结构特征。该研究为AFM1免疫学检测提供核心元件,并为探究AFM1与VHH的分子结合机制奠定基础。

目的比较^(68)Ca-前列腺特异性膜抗原(PSMA)PET/CT与多参数磁共振(mpMRI)对初诊前列腺癌分期诊断的效能。方法回顾性分析2021年12月至2023年6月复旦大学附属肿瘤医院收治的30例前列腺癌患者的病例资料。中位年龄68(67,76)岁。前列腺特异性抗原(PSA)17.91(9.41,39.53)ng/ml。患者术前均接受^(68)Ga-PSMAPET/CT和mpMRI检查,两项检查中位间隔时间21.00(2.75,35.50)d。影像学检查共发现50处病灶。患者术前均经穿刺活检病理确诊为前列腺腺癌,均未接受任何抗肿瘤治疗。所有患者均接受根治性前列腺切除术,手术切除标本术后制作病理大切片。将术后病理大切片作为金标准,比较^(68)Ga-PSMAPET/CT与mpMRI在诊断前列腺癌包膜外侵犯(EPE)、精囊腺侵犯(SVI),以及前列腺内肿瘤病灶方面的敏感性和特异性。结果本组30例,术后病理结果显示12例出现EPE,8例出现SVI。术前发现的50处病灶中,术后病理检查诊断为有临床意义前列腺癌病灶38处(76.0%)。将mpMRI、^(68)Ga-PSMAPET/CT检查图像与术后病理大切片行三重融合比较。^(68)Ga-PSMAPET/CT和mpMRI诊断EPE的敏感性分别为66.7%(8/12)和50.0%(6/12)(P=0.500),特异性分别为88.9%(16/18)和83.3%(15/18)(P=1.000);诊断SVI的敏感性分别为50.0%(4/8)和75.0%(6/8)(P=0.687),特异性分别为86.3%(19/22)和90.9%(20/22)(P=1.000);诊断有临床意义前列腺癌病灶的敏感性分别为89.5%(34/38)和63.2%(24/38)(P=0.031),特异性分别为50.0%(6/12)和33.3%(4/12)(P=0.750)。结论^(68)Ga-PSMA PET/CT诊断前列腺癌病灶的敏感性显著高于mpMRI,两种检查方法的特异性无显著差异.

TWO MODELS FOR ROLL WAVES IN A MUD LAYER

As has been extensively reported by hydraulic engineers in China, the flow of hyperconcentrated fluid mud in rivers is often pulsating and appears as roll waves. We report here two theories for such pulsating flows besed on two constitutive laws: the power law and Bingham plastic law. In order to account for the nonlinear bores convective inertia is kept in the approximate long wave equations. Kar man's momentum integral method with an assumed velocity profile is then employed. Results of the linearized instability analyses are *** bores are obtained either as stationary wave solutions or as the long- time limit of unstable disturbances which are initially infinitesimal. Implications on ho scour is examined.

纳米抗体(nanobody,Nb)具有稳定性高、特异性强、易于表达等优势,已经成为小分子免疫分析的重要工具。为了监测乳制品中黄曲霉毒素M1(aflatoxin M1,AFM1)的污染情况,以先前从双峰驼AFM1免疫文库中淘选得到的抗AFM1型纳米抗体M6为研究对象,评估其理化性质并建立间接竞争酶联免疫吸附测定法(indirect competitive enzyme-linked immunosorbent assay,icELISA)。优化反应体系中封闭方法、甲醇溶液、pH值和离子强度等参数后,建立竞争抑制标准曲线,并进行方法学验证特异性和准确性。结果表明,Nb-M6具有良好的亲和力和热稳定性;Nb-icELISA的检测限(limit of detection,LOD)为0.111 ng/mL,半数抑制浓度(half maximal inhibitory concentration,IC_(50))为1.498 ng/mL,乳制品加标回收率为89.8%~104.1%,与高效液相色谱法的检测结果趋于一致。该方法操作简单、便捷,可应用于乳制品中AFM1的初步批量筛查。
