Immune response pattern varies with the natural history of chronic hepatitis B

BACKGROUND Chronic hepatitis B is a highly heterogeneous disease that can be divided into four phases: Immune tolerant(IT), immune active(IA), inactive carrier(IC) and hepatitis B envelope antigen(HBeAg)-negative hepatitis(ENEG).AIM To investigate the immune status of natural killer(NK) and T cells in different phases of chronic hepatitis *** The frequency, phenotype and function of circulating NK cells, as well as nonantigen-specific and hepatitis B virus(HBV)-specific T cell responses were detected by flow cytometry in healthy and HBV-infected *** The ability of NK cells to produce IFN-γ was markedly attenuated in HBVinfected patients overall but was less compromised in IC patients. Patients in the IT and IA phases also displayed significantly lower TNF-α production compared to healthy subjects. NK cells were phenotypically activated in the IA and ENEGphases, as evidenced by the upregulation of NKp44 in CD56^(bright) NK cells and CD69 in CD56^(dim) NK cells. Furthermore, global T-cells from the ENEG phase displayed a proinflammatory cytokine profile with upregulated IFN-γ and TNF-αexpression, while this profile was suppressed in IT and IA patients. Finally, core and S antigen-specific T cell responses were significantly stronger after in vitro expansion in the IC phase compared to other *** Our findings demonstrate the changes in immune response pattern during the natural history of HBV infection. Both NK and T cells are functionally impaired in the IT and IA phases. With the spontaneous clearance of HBeAg and hepatitis B surface antigen decline, NK cell cytokine production and HBV-specific T responses are partially restored in IC phase, and the ENEG phase is dominated by nonantigen-specific T cell responses.

The ecological consequences of precipitation change and increased atmospheric nitrogen(N)deposition have profound impacts on ecosystem CO2 exchange in grassland *** and N can largely influence grassland productivity,community composition and ecosystem ***,the influences of water and N addition on the ecosystem CO2 exchange of alpine grassland ecosystems remain unclear.A field manipulative experiment with water and N additions was conducted in an alpine meadow on the Tibetan Plateau over 4 years with contrasting precipitation *** were four treatments:control(Ctrl),N addition(N),water addition(W)and N and water addition(NW),each replicated three times.N addition,but not water addition,increased gross ecosystem productivity(GEP),plant biomass,community cover and community-weighted mean *** responses of ecosystem CO2 exchange to water and N addition varied between the wet and dry *** addition had a positive effect on net ecosystem carbon exchange(NEE)due to a larger increase in GEP than in ecosystem respiration(ER)only in the dry *** the contrary,N addition significantly enhanced ecosystem CO2 exchange only in the wet *** increased GEP in N addition was attributed to the larger increase in NEE than ***,N addition stimulated NEE mainly through increasing the cover of dominant *** observations highlight the important roles of precipitation and dominant species in regulating ecosystem CO2 exchange response to global environmental change in alpine grasslands.

liver metabonomics study on the protective effect of glycyrrhetinic acid against realgar-induced liver injury

Glycyrrhetinic acid(GA)is the bioactive ingredient in Glycyrrhizae Radix et *** previous study has reported that GA has protective effect on realgar-induced ***,the details of the hepatoprotective mechanisms of GA on realgar-induced liver injury remain to be *** the study,mice were divided into control,GA-control,realgar,and co-treated *** liver tissues were used for metabonomics study by ultra-performance liquid chromatography-mass spectrometry(UPLCMS)*** results illustrate that GA significantly ameliorate the liver injury and metabolic perturbations caused by *** metabolites,such as phenylalanine,pyroglutamic acid(PGA),proline,carnitine,nicotinamide,choline,lysophosphatidylcholine(LPC)16:0 and LPC 18:2 were found responsible for the hepatoprotective effect of *** metabolites are associated with the methylation metabolism of arsenic,cell membrane structure,energy metabolism and oxidative *** the results of this study,we infer that the potential hepatoprotective mechanism of GA on realgar-induced liver injury may be associated with reducing arsenic accumulation and its methylation metabolism in the liver,promoting the conjugation of arsenic and GSH to play detoxification effect,and ameliorating the liver metabolic perturbations caused by realgar.

The interaction between a functionalized single-wailed carbon nanotube (f-SWCNT) and the YAP65WW protein domain is investigated by using molecular dynamics *** is found that the f-SWCNT binds onto the active site of the YAP65WW domain and leads to a substantial conformational change of the protein domain,which may securely affect the original function of *** the hydrophobic interaction and the long lifetime hydrogen bonds play important roles in the binding.

Porous Si/C composite as anode materials for high-performance rechargeable lithium-ion battery

A novel porous silicon was synthesized through a magnesiothermic reduction method of molecular sieve for the first time, Si/C composite was synthesized by using pitch as carbon source. The porous Si/C composite shows a high initial specific capacity of 2018.5 mAh/g with current density of 0.1 A/g. When the current density increases to 2 A/g, it still exhibits high average specific capacity of 640.3 mAh/g. The porous structure can remit the Si particle pulverization during the lithiation]delithiation process. This article can provide a reference for the research of the porous Si anode for the high performance rechargeable lithium-ion battery.

Poly(meta-phenylene) Derivative with Rigid Twisted Biphenyl Units in Backbone:Synthesis,Structural Characterization,Photophysical Properties and Electroluminescence

A soluble poly（meta-phenylene） derivative with rigid twisted biphenyl unit was synthesized by the Yamamoto coupling reaction. The polymer is soluble in common organic solvents, and the number-average molecular weight is about 6500. The UV-Vis and quantum chemical calculation indicate that the different conformation segments named ＂conformers＂ exist in the polymer backbones; it was also further confirmed by the single crystal X-ray diffraction study of the dimeric model compound. The π-π^＊ transition of biphenyl segments of twisted and planar conformations made the polymer exhibit a strong absorption around 256 nm and a weak absorption at about 300 nm. Furthermore, the polymer exhibits a strong UV photoluminescence at 372 nm when the excitation wavelengths are longer than 300 run. The ultraviolet-emitting electroluminescence（EL） device with the single layer structure shows EL λmax of the derivative at 370 nm.

Leymus chinensis（Trin.） Tzvel.,widely distributed at eastern Eurasian steppe and divided into gray-green type and yellow-green type,has different stress resistance to *** the present study,the water-soluble polysaccharides from two ecotypes of *** were analyzed in detail,and the differences between polysaccharides from the two ecotypes of *** in the yield,monosaccharide composition,molecular weight and structure were *** polysaccharides of *** were composed of both neutral and acidic *** neutral polysaccharides contained mannose,glucose,galactose,xylose and arabinose,and mainly consisted of β-1,4-Glcp,α-1,3-Galp and α-1,2-Xylp *** acidic polysaccharides contained mannose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose,xylose and ***,the yields,monosaccharides contents and the molecular weights of the polysaccharides from the two ecotypes of *** were ***,the resistance type（gray-green type） of *** contained a number of α-1,3-Manp and reducing end of β-Glcp residues,and much more O-methyl groups than normal type（yellow-green type） of *** differences of the polysaccharides of the two ecotypes of *** might be due to the long-term environmental adaptability of plant,and the differences of the polysaccharides might influence the stress resistance of ***.

The effect of lactic acid bacteria inoculums on in vitro rumen fermentation, methane production, ruminal cellulolytic bacteria populations and cellulase activities of corn stover silage
