目的明确明桑降脂茶的降脂作用及其发挥降脂作用的物质基础。方法制备大鼠空白血清、明桑降脂茶全方和桑叶减味含药血清,以油酸诱导人正常肝细胞(LO2细胞)建立脂肪变性细胞模型,将细胞分为对照组(含空白血清)、模型组(含空白血清)、明桑降脂茶全方血清处理组、桑叶减味血清处理组。培养4 h后,显微镜下观察各组脂肪积累情况;酶标仪测定细胞内脂质含量;按试剂盒说明测定细胞内甘油三酯(triglyceride,TG)、总胆固醇(totalcholesterlo,TC)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)含量,以及过氧化物酶体增殖物启动受体α(peroxisome proliferator-activated receptor alpha,PPARα)、固醇调节元件结合蛋白-1c(sterol regulatory element binding protein 1c,SREBP-1c)表达。高效液相色谱法(high performance liquid chromatography,HPLC)研究各组指纹图谱、绿原酸和橙黄决明素含量的差异。结果与模型组比较,明桑降脂茶全方血清组、桑叶减味血清组均能显著降低细胞内脂滴数量和脂质含量,降低TG、TC、LDL-C含量和SREBP-1c表达,升高HDL-C含量和PPARα表达,全方血清组的降脂作用优于桑叶减味血清组;全方血清组、桑叶减味血清组、空白血清组共有峰分别为53、35、23个,三者共有峰18个;剔除共有峰后,全方血清组有22个不同于桑叶减味血清组的差异峰,桑叶减味血清组有4个不同于全方血清组的差异峰;全方血清组中绿原酸和橙黄决明素含量均高于桑叶减味血清组,绿原酸含量在二者之间差异显著。结论明桑降脂茶具有明显的降脂作用,桑叶在其中发挥重要降脂作用。

Background： SH3TC2, PMP2, and BSCL2 genes are related to autosomal recessive （AR） Charcot-Marie-Tooth （CMT） disease type 1, autosomal dominant （AD）-CMTI, and AD-CMT2, respectively. Pathogenic variants in these three genes were not well documented in Chinese CMT patients. Therefore, this study aims to detect SH3TC2, PMP2, and BSCL2 pathogenic variants in a cohort of 315 unrelated Chinese CMT families. Methods： A total of 315 probands from 315 unrelated Chinese CMT families were recruited from the Department of Neurology of Third xiangya Hospital and xiangya Hospital. We screened for SH3TC2 pathogenic variants in 84 AR or sporadic CMT probands, PMP2 pathogenic variants in 39 AD or sporadic CMTI probands, and BSCL2 pathogenic variants in 50 AD or sporadic CMT2 probands, using polymerase chain reaction and Sanger sequencing. All these patients were out of 315 unrelated Chinese CMT families and genetically undiagnosed after exclusion of pathogenic variants of PMP22, MFN2, MPZ, GJB 1, GDAP1, HSPB1, HSPB8, EGR2, NEFL. and RABT. Candidate variants were analyzed based on the standards and guidelines of American College of Medical Genetics and Genomics （ACMG）. Clinical features were reevaluated. Results： We identified three novel heterozygous variants such as p.L95V （c.283C〉G）, p.L 1048P （c.3143T〉C）, and p.V 1105 M （c.3313G〉A） of SH3TC2 gene and no pathogenic variants of PMP2 and BSCL2 genes. Although evaluation in silico and screening in the healthy control revealed that the three SH3TC2 variants were likely pathogenic, no second allele variants were discovered. According to the standards and guidelines of ACMG, the heterozygous SH3TC2 variants such as p.L95V, ***048P, and p.V1105M were considered to be of uncertain significance. Conclusions： SH3TC2, PMP2, and BSCL2 pathogenic variants might be rare in Chinese CMT patients. Further studies to confirm our findings are needed.

Simulated Microgravity Conditions and Carbon Ion Irradiation Induce Spermatogenic Cell Apoptosis and Sperm DNA Damage

Objective To investigate the effect of simulated microgravity and carbon ion irradiation （CIR） on spermatogenic cell apoptosis and sperm DNA damage to the testis of male Swiss Webster mice, and assess the risk associated with space environment. Methods Sperm DNA damage indicated by DNA fragmentation index （DFI） and high DNA stainability （HDS） was measured by sperm chromatin structure assay （SCSA）. Apoptosis of spermatogenic cells was detected by annexin V-propidium iodide assay. Bax （the expression levels of p53） and proliferating cell nuclear antigen （PCNAI were measured by immunoblotting; p53 and PCNA were located by immunohistology. Results HDS, DFI, apoptosis index, and the expression levels of p53 and Bax were detected to be significantly higher in the experimental groups （P〈0.05） compared with those in the control group, however, the PCNA expression varied to a certain degree, p53- and PCNA- positive expression were detected in each group, mainly in relation to the spermatogonic cells and spermatocytes. Conclusion The findings of the present study demonstrated that simulated microgravity and CIR can induce spermatogenic cell apoptosis and sperm DNA damage. Sperm DNA damage may be one of the underlying mechanisms behind male fertility decline under space environment. These findings may provide a scientific basis for protectint~ astronauts and space traveler＇s health and safety.

The multi-physics instrument(MPI)is the first user cooperative instrument at the China Spallation Neutron Source(CSNS).It was designed to explore the structures of complex materials at multiple scales based on the neutron total scattering *** imposes the requirements for the detector,including a high detection efficiency to reduce the measurement time and a large solid angle coverage to cover a wide range of momentum *** satisfy these demands,a large-area array of 3He-filled linear position-sensitive detectors(LPSDs)was constructed,each with a diameter of 1 inch and pressure of 20 *** uses an orbicular layout of the detector and an eight-pack module design for the arrangement of 3He LPSDs,covering a range of scattering angles from 3°to 170°with a total detector area of approximately 7 *** detector works in air,which is separated from the vacuum environment to facilitate installation and *** characteristics of the MPI detector were investigated through Monte Carlo(MC)simulations using Geant4 and experimental *** results suggest that the detectors are highly efficient in the wavelength range of the MPI,and an efficiency over 25%is achievable for above 0.1 A neutrons.A minimal position resolution of 6.4 mm full width at half maximum(FWHM)along the tube length was achieved at a working voltage of 2200 V,and a deviation below 2 mm between the real and measured positions was attained in the beam *** detector module exhibited good consistency and an excellent counting rate capacity of up to 80 kHz,which satisfied the requirements of experiments with a high event *** of its operation over the past year have shown that the detector works steadily in sample experiments,which allows the MPI to serve the user program successfully.

BACKGROUND The efficacy and safety of proprotein convertase subtilisin/kexin type 9(PCSK-9) inhibitors were confirmed by several clinical trials, but its effectiveness in routine clinical practice in China has not been evaluated. This study aims to describe the real world effectiveness of PCSK-9 inhibitors combined with statins compared with statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease(ASCVD).METHODS This is a multi-center observational study, enrolled patients from 32 hospitals who underwent percutaneous coronary intervention(PCI) from January to June in 2019. There are 453 patients treated with PCSK-9 inhibitors combined with statins in PCSK-9 inhibitor group and 2,610 patients treated with statins-based lipid lowering therapies in statins-based group. The lipid control rate and incidence of major adverse cardiovascular events(MACE) over six months were compared between two groups.A propensity score-matched(PSM) analysis was used to balance two groups on confounding factors. Survival analysis using Kaplan-Meier methods was applied for *** In a total of 3,063 patients, 89.91% of patients had received moderate or high-intensity statins-based therapy before PCI, but only 9.47% of patients had low-density lipoprotein cholesterol(LDL-C) levels below 1.4 mmol/L at baseline. In the PSM selected patients, LDL-C level was reduced by 42.57% in PCSK-9 inhibitor group and 30.81%(P < 0.001) in statins-based group after six months. The proportion of LDL-C ≤ 1.0 mmol/L increased from 5.29% to 29.26% in PCSK-9 inhibitor group and 0.23% to 6.11% in statins-based group, and the proportion of LDL-C ≤ 1.4 mmol/L increased from 10.36% to 47.69% in PCSK-9 inhibitor group and 2.99% to 18.43% in statins-based group(P < 0.001 for both). 针对渤海某油田面临的SRB细菌全面控制难题,评选了9种杀菌剂,优选出长链烷基胍盐杀菌剂SJ-03,现场评价结果表明,当加剂浓度为30mg/L时,长链烷基胍盐杀菌剂SJ-03的杀菌率达100%,综合对比SJ-03的杀菌抑菌效果均优于其他药剂;配伍性实验表明,加入杀菌剂后均未对原油脱水速度与脱水体积产生明显影响;现场应用效果表明,以一定浓度冲击加注杀菌剂SJ-03后,监测周期内SRB细菌含量始终保持在低于6个/mL,说明杀菌剂SJ-03对水系统内SRB细菌的杀菌抑菌能力明显,同时水工艺舱出口、缓冲罐入口、缓冲罐出口和注水出口点位的水中含油OIW整体较为平稳,未发生大范围波动,因此杀菌剂的改变不会对平台生产流程造成影响。
