Electrospray ionization mass spectrometry（ESI-MS） was applied simultaneously in determining norditerpenoid alkaloids from the roots of Aconitum sinomantanum Nakai （RAS） based on molecular mass information. The tandem mass spectra（ ESI-MS^n） provided the alkaloidal structural information, through which the existence of these alkaloids was further confirmed. Accordingly, six known norditerpenoid alkaloids were simultaneously determined on the basis of their ESI-MS^n spectra. Furthermore, based on the diagnostic fragmentation pathways of alkaloidal MS^n, a rapid method for direct detection and characterization of alkaloids from an ethanolic extract of RAS was described.

The application of n-alkane as markers to estimate herbage intake, apparent digestibility and species composition of diet consumed by grazing sheep was studied. Six local Mongolian sheep were used to determine dry matter （DM） intake, apparent DM digestibility and species composition of diet during summer, autumn and winter. Animals were orally dosed twice daily with n-alkane gelatin capsules containing 60 mg C32-alkane as an external marker. Diet composition was estimated by comparing the odd-chain n-alkanes pattern profile （C27-C31） of the consumed plant species with the n-alkanes fecal concentrations of grazing animals, using a non-negative least squares algorithm called EATWHAT software package. The alkane pair C32：C33 and C33 alkane were used to estimate DM intake and diet apparent DM digestibility, respectively. The results showed that daily dry matter intake of the sheep were 1.77, 1.61 and 1.18 kg d-1 in summer, autumn and winter, respectively. Apparent DM digestibility, crude protein （CP）, metabolizable energy （ME） and neutral detergent fiber （NDF） intake of diet consumed by sheep decreased significantly （P〈0.01） from summer to winter, with no evident changes in ADF and ADL intake. Diet composition indicated Artemisia frigida Willd was the most dominant diet component, contributed 79.68, 68.12 and 86.26% of sheep＇s diets in summer, autumn and winter, respectively. Cleistogenes songorica Ohwi and Convolvulus arnmannii Desr were the important components of the diet. Although Stipa breviflora Griseb is one of the main plant species in the study area, the sheep rarely choosed it. The study indicated that CP and ME in diet consumed by sheep were deficient in winter. Therefore, appropriate supplementation strategies should be indispensable during this period.

Most materials can be easily corroded or ineffective in carbonaceous atmospheres at high temperatures in the reactor core of the high temperature gas-cooled reactor(HTGR).To solve the problem,a material performance test apparatus was built to provide reliable materials and technical support for relevant experiments of the *** apparatus uses a center high-purity graphite heater and surrounding thermal insulating layers made of carbon fiber felt to form a strong carbon reducing atmosphere inside the *** designed tungsten rhenium thermocouples which can endure high temperatures in carbonaceous atmospheres are used to control the temperature field.A typical experimental process was analyzed in the paper,which lasted 76 hours including seven *** results showed the test apparatus could completely simulate the carbon reduction atmosphere and high temperature environment the same as that confronted in the real reactor and the performance of screened materials had been successfully tested and *** temperature in the apparatus could be elevated up to 1600oC,which covered the whole temperature range of the normal operation and accident condition of HTGR and could fully meet the test requirements of materials used in the reactor.

Cereal cyst nematode（Heterodera avenae, CCN） distributes worldwide and has caused severe damage to cereal crops, and a model host will greatly aid in the study of this nematode. In this research, we assessed the sensitivity of 25 inbred lines of Brachypodium distachyon to H. avenae from Beijing, China. All lines of B. distachyon were infested by secondstage juveniles（J2s） of H. avenae from Daxing District of Beijing population, but only 13 inbred lines reproduced 0.2–3 cysts/plant, showing resistance. The entire root system of the infested B. distachyon appeared smaller and the fibrous roots were shorter and less numerous. We found that a dose of 1 000 J2s of H. avenae was sufficient for nematode infestation. We showed that Koz-1 of B. distachyon could reproduce more cysts than TR2A line. Line Koz-1 also supported the complete life cycles of 5 CCN geographical populations belonging to the Ha1 or Ha3 pathotype group. Our results suggest that B. distachyon is a host for CCN.

By using transport and magnetic measurement,the upper critical field H_(c2)(T)and the irreversibility line Hirr(T)has been determined.A big separation between H_(c2)(0)and Hirr(0)has been found showing the existence of a quantum vortex liquid state induced by quantum fluctuation of vortices in the new superconductor MgB_(2).Further investigation on the magnetic relaxation shows that both the quantum tunneling and the thermally activated flux creep depends weakly on ***,when the melting field Hirr is approached,a drastic rise of the relaxation rate is *** may indicate that the melting of the vortex matter at a finite temperature is also induced by the quantum fluctuation of vortices.

Powdery mildew, caused by Blumeria graminis f. sp. tritici, is one of the most devastating wheat diseases. Wild emmer wheat（Triticum turgidum ssp. dicoccoides） is a promising source of disease resistance for wheat. A powdery mildew resistance gene conferring resistance to B. graminis f. sp. tritici isolate E09, originating from wild emmer wheat, has been transferred into the hexaploid wheat line WE4 through crossing and backcrossing. Genetic analyses indicated that the powdery mildew resistance was controlled by a single dominant gene, temporarily designated Ml WE4. By mean of comparative genomics and bulked segregant analysis, a genetic linkage map of Ml WE4 was constructed, and Ml WE4 was mapped on the distal region of chromosome arm 5BL. Comparative genetic linkage maps showed that genes Ml WE4, Pm36 and Ml3D232 were co-segregated with markers XBD37670 and XBD37680, indicating they are likely the same gene or alleles in the same locus. The co-segregated markers provide a starting point for chromosome landing and map-based cloning of Ml WE4, Pm36 and Ml3D232.

Powdery mildew, caused by Blumeria graminis f. sp. tritici （Bgt）, is one of the most damaging diseases to wheat in the world. The cultivation of resistant varieties of wheat is essential for controlling the powdery mildew epidemic. Wheat landraces are important resources of resistance to many diseases. Mapping powdery mildew resistance genes from wheat landraces will promote the development of new varieties with disease resistance. The Chinese wheat landrace Baiyouyantiao possesses characteristic of disease resistance to powdery mildew. To identify the resistance gene in this landrace, Baiyouyantiao was crossed with the susceptible cultivar Jingshuang 16 and seedlings of parents and F1, BC1, F2, and F~：3 were tested with Bgt isolate E09. The genetic results showed that the resistance of Baiyouyantiao to E09 was controlled by a single recessive gene, tentatively designated PmBYYT. An Illumina wheat 90K single-nucleotide polymorphism （SNP） array was applied to screen polymorphisms between F2-resistant and F2-susceptible DNA bulks for identifying the chromosomal location of PmBYYT. A high percentage of polymorphic SNPs between the resistant and susceptible DNA bulks was found on chro- mosome 7B, indicating that PmBYYT may be located on this chromosome. A genetic linkage map of PmBYYTconsisting of two simple sequence repeat markers and eight SNP markers was developed. The two flanking markers were SNP markers W7BL-8 and W7BL-15, with genetic distances of 3 and 2.9 cM, respectively. The results of this study demonstrated the rapid characterization of a wheat disease resistance gene and SNP marker development using the 90K SNP assay. The flanking markers of gene PmBYYTwill benefit marker-assisted selection （MAS） and map-based cloning in breeding wheat cultivars with powdery mildew resistance.
