Nutrition, gastrointestinal microorganisms and metabolites in mastitis occurrence and control

Mastitis affects almost all mammals including humans and dairy cows. In the dairy industry, bovine mastitis is a disease with a persistently high incidence, causing serious losses to the health of cows, the quality of dairy products, and the economy of dairy farms. Although local udder infection caused by the invasion of exogenous pathogens into the mammary gland was considered the main cause of mastitis,evidence has been established and continues to grow, showing that nutrition factors and gastrointestinal microbiome(GM) as well as their metabolites are also involved in the development of mammary inflammatory response. Suboptimal nutrition is recognized as a risk factor for increased susceptibility to mastitis in cattle, in particular the negative energy balance. The majority of data regarding nutrition and bovine mastitis involves micronutrients. In addition, the dysbiotic GM can directly trigger or aggravate mastitis through entero-mammary gland pathway. The decreased beneficial commensal bacteria, lowered bacterial diversity, and increased pathogens as well as proinflammatory metabolites are found in both the milk and gastrointestinal tract of mastitic dairy cows. This review discussed the relationship between the nutrition(energy and micronutrient levels) and mastitis, summarized the role of GM and metabolites in regulating mastitis. Meanwhile, several non-antibiotics strategies were provided for the prevention and alleviation of mastitis, including micronutrients, probiotics, short-chain fatty acids, highfiber diet, inulin, and aryl hydrocarbon receptor.

To this end, an overview of systems science is first presented, followed by the conclusion that the development of artificial intelligence(AI) has promoted the derivation and development of intelligent systems(ISs).

Dietary supplementation with inulin improves lactation performance and serum lipids by regulating the rumen microbiome and metabolome in dairy cows

This study investigated the effects of inulin on rumen fermentation parameters,ruminal microbiome and metabolites,as well as lactation performance and serum indexes in dairy *** Holstein dairy cows with similar body conditions were randomly divided into 2 groups(n=8 per group),with inulin addition at 0 and 200 g/d *** experiment lasted for 6 weeks,including a 1-week adaptation period and a 5-week treatment *** the end of the experimental period,the milk,serum and rumen fluid were sampled and *** microbiome and metabolome in the rumen fluid were analyzed via 16 S rRNA sequencing and untargeted metabolomics,*** results showed that supplementation with inulin(200 g/d per cow)increased the milk yield(P=0.001),milk protein(P=0.032),lacto se rate(P=0.004)and proportion of saturated fatty acids(SFA)in milk(P<0.001),but decreased the proportion of unsaturated fatty acids(USFA)(P=0.041).Rumen pH(P=0.040)and the concentration of NH3-N(P=0.024)were decreased;however,acetate(P<0.001),propionate(P=0.003),butyrate(P<0.001)and lactic acid(LA)(P=0.043)were *** total cholesterol(TC)(P=0.008)and triglycerides(TG)(P=0.01)in serum were also ***,inulin addition elevated the relative abundance of several beneficial symbiotic and short-chain fatty acid(SCFA)-producing bacteria,such as Muribaculaceae(false discovery rate[FDR]-adjusted P<0.01),Acetitomaculum(FDR-adjusted P=0.043),and Bu tyrivibrio(FDR-adjusted P=0.036),while elevating the levels of L-lysine(FDR-adjusted P=4.24×10^(-3)),L-proline(FDR-adjusted P=0.0158),and L-phenylalanine(FDRadj usted P=0.027).In contrast,several pathogens and ruminal bacteria abu ndant in high-fat diets,such as Escherichia-Sh igella(FD R-adj usted P=0.022),Erysipelo trichaceaeUCG-004(FD R-adjusted P<0.01)and RF39(FDR-adjusted P=0.042)were decreased along with the reduction of lysophosphatidylcholine(LysoPC)(18:1(9 Z))(FDR-adjusted P=1.03×10^(-3)),LysoPC(16:0)(FDR-adjusted P=0.0108),LysoP

Flavones are among the major colorless pigments synthesized through branches of the flavonoid pathway in ***,due to the absence of a gene encoding flavone synthase(FNS)in the model plant Arabidopsis thaliana species,the regulatory mechanism of FNS-catalyzed flavone biosynthesis has rarely been studied in ***,it was found that flavones play a predominant role in the elimination of excess reactive oxygen species(ROS)at high temperatures in colorless plant organs.A novel atypical subgroup 7(SG7)R2R3-MYB transcription factor,CmMYB012,was found to be induced in response to prolonged high temperatures and to inhibit flavone biosynthesis by directly regulating ***,CmMYB012 was also found to inhibit anthocyanin biosynthesis by suppressing the expression of CmCHS,CmDFR,CmANS,and ***012 overexpression exerted a negative in fluence on plant fitness and pink flower color formation,while CmMYB012 suppression had the opposite effect in response to high *** findings provide new insights into the mechanisms by which high temperatures regulate the metabolism of flavones and anthocyanins to affect plant fitness and flower color formation.

Post-traumatic stress disorder(PTSD)is a psychiatric disease that seriously affects brain ***,selective serotonin reuptake inhibitors(SSRIs)are used to treat PTSD clinically but have decreased efficiency and increased side *** this study,nasal cannabidiol inclusion complex temperature-sensitive hydrogels(CBD TSGs)were prepared and evaluated to treat *** model of PTSD was established with conditional fear *** TSGs could significantly improve the spontaneous behavior,exploratory spirit and alleviate tension in open field box,relieve anxiety and tension in elevated plus maze,and reduce the freezing *** and eosin and c-FOS immunohistochemistry slides showed that the main injured brain areas in PTSD were the prefrontal cortex,amygdala,and hippocampus *** TSGs could reduce the level of tumor necrosis factor-a caused by *** blot analysis showed that CBD TSGs increased the expression of the 5-HT1 A *** administration of CBD TSGs was more efficient and had more obvious brain targeting effects than oral administration,as evidenced by the pharmacokinetics and brain tissue distribution of CBD ***,nasal CBD TSGs are safe and effective and have controlled *** are a novel promising option for the clinical treatment of PTSD.

Synthesis of silica/metatitanic acid nanocomposite and evaluation of its catalytic performance for aquathermolysis reaction of extra-heavy crude oil

A lipophilic silica/metatitantic acid（denoted as Si O2/H2 TiO 3） nanocomposite was synthesized by hydrothermal reaction with surface-modified Si O2 as the lipophilic carrier. As-synthesized Si O2/H2 TiO 3nanocomposite was used as a catalyst to promote the aquathermolysis reaction of extra-heavy crude oil thereby facilitating the recovering from the deep reservoirs at lowered temperature. The catalytic performance of the as-synthesized Si O2/H2 TiO 3catalyst for the aquathermolysis reaction of the heavy oil at a moderate temperature of 150 °C was evaluated in relation to the structural characterizations by TEM,FTIR,XRD and FESEM as well as the determination of the specific surface area by N2adsorption–desorption method. Findings indicate that as-synthesized Si O2/H2 TiO 3nanocomposite exhibits an average size of about 20 nm as well as good lipophilicity and dispersibility in various organic solvents; and it shows good catalytic performance for the aquathermolysis reaction of the extra-heavy oil extracted from Shengli Oilfield of China. Namely,the assynthesized Si O2/H2 TiO 3catalyst is capable of significantly reducing the viscosity of the tested heavy oil from58,000 c P to 16,000 c P（referring to a viscosity reduction rate of 72.41%） at a mass fraction of 0.5%,a reaction temperature of 150 °C and a reaction time of 36 h,showing potential application in downhole upgrading heavy crude oils.

Predatory bacterial hydrogels for topical treatment of infected wounds

Wound infection is becoming a considerable healthcare crisis due to the abuse of antibiotics and the substantial production of multidrug-resistant *** immersion wounds usually become a mortal trouble because of the infection of Vibrio *** bacteriovorus,one kind of natural predatory bacteria,is recognized as a promising biological therapy against intractable ***,we prepared a ***-loaded polyvinyl alcohol/alginate hydrogel for the topical treatment of the seawater immersion wounds infected by *** ***-loaded hydrogel(BG)owned highly microporous structures with the mean pore size of 90μm,improving the rapid release of *** from BG when contacting the aqueous *** showed high biosafety with no L929 cell toxicity or *** importantly,BG exhibited excellent in vitro *** *** highly effective infected wound treatment effect of BG was evaluated on mouse models,revealing significant reduction of local ***,accelerated wound contraction,and alleviated *** the high bacterial inhibition of BG,BG remarkably reduced inflammatory response,promoted collagen deposition,neovascularization and re-epithelization,contributing to wound *** is a promising topical biological formulation against infected wounds.

Short-chain fatty acids ameliorate spinal cord injury recovery by regulating the balance of regulatory T cells and effector IL-17^(+) γδ T cells

Spinal cord injury(SCI)causes motor,sensory,and autonomic *** gut microbiome has an important role in SCI,while short-chain fatty acids(SCFAs)are one of the main bioactive mediators of *** the present study,we explored the effects of oral administration of exogenous SCFAs on the recovery of locomotor function and tissue repair in ***’s method was utilized to establish an SCI model in Sprague-Dawley(SD)*** animals received water containing a mixture of 150 mmol/L SCFAs after *** 21 d of treatment,the Basso,Beattie,and Bresnahan(BBB)score increased,the regularity index improved,and the base of support(BOS)value *** cord tissue inflammatory infiltration was alleviated,the spinal cord necrosis cavity was reduced,and the numbers of motor neurons and Nissl bodies were ***-linked immunosorbent assay(ELISA),real-time quantitative polymerase chain reaction(qPCR),and immunohistochemistry assay revealed that the expression of interleukin(IL)-10 increased and that of IL-17 decreased in the spinal *** promoted gut homeostasis,induced intestinal T cells to shift toward an anti-inflammatory phenotype,and promoted regulatory T(Treg)cells to secrete IL-10,affecting Treg cells and IL-17^(+)γδT cells in the spinal ***,we observed that Treg cells migrated from the gut to the spinal cord region after *** above findings confirm that SCFAs can regulate Treg cells in the gut and affect the balance of Treg and IL-17^(+)γδT cells in the spinal cord,which inhibits the inflammatory response and promotes the motor function in SCI *** findings suggest that there is a relationship among gut,spinal cord,and immune cells,and the“gut-spinal cord-immune”axis may be one of the mechanisms regulating neural repair after SCI.
