Plantations and production of yellowhorn,one of the most important woody oil and urban greening trees widely cultivated in northern China,have gradually become limited by drought *** epicuticular wax layer plays a key role in the protection of yellowhorn trees from drought and other ***,there is no research on the mechanism of wax loading in yellowhorn *** this study,we investigated the anatomical and physiological characteristics of leaves from different germplasm resources and different parts of the same tree and compared their cuticle *** addition,the different expression patterns of genes involved in wax accumulation were analyzed,and a coexpression network was built based on transcriptome sequencing *** and physiological comparisons found that the sun leaves from the outer part of the crown had thicker epicuticular wax,which altered the permeability and improved the drought resistance of leaves,than did shade *** on transcriptome data,a total of 3008 and 1324 differentially expressed genes(DEGs)were identified between the sun leaves and shade leaves in glossy-and non-glossy-type germplasm resources,*** identified 138 DEGs involved in wax biosynthesis and transport,including structural genes(such as LACS8,ECH1,and ns-LTP)and transcription factors(such as MYB,WRKY,and bHLH transcription factor family proteins).The coexpression network showed a strong correlation between these *** differences in gene expression patterns between G-and NG-type germplasm resources under different light conditions were very *** results not only provide a theoretical basis for screening and developing drought-resistant yellowhorn germplasm resources but also provide a data platform to reveal the wax accumulation process of yellowhorn leaves.

The Sulige tight gas reservoir is characterized by low-pressure, low-permeability and lowabundance. During production, gas flow rate and reservoir pressure decrease sharply; and in the shut- in period, reservoir pressure builds up slowly. Many conventional methods, such as the indicative curve method, systematic analysis method and numerical simulation, are not applicable to determining an appropriate gas flow rate. Static data and dynamic performance show permeability capacity, kh is the most sensitive factor influencing well productivity, so criteria based on kh were proposed to classify vertical wells. All gas wells were classified into 4 groups. A multi-objective fuzzy optimization method, in which dimensionless gas flow rate, period of stable production, and recovery at the end of the stable production period were selected as optimizing objectives, was established to determine the reasonable range of gas flow rate. In this method, membership functions of above-mentioned optimizing factors and their weights were given. Moreover, to simplify calculation and facilitate field use, a simplified graphical illustration （or correlation） was given for the four classes of wells. Case study illustrates the applicability of the proposed method and graphical correlation, and an increase in cumulative gas production up to 37% is achieved and the well can produce at a constant flow rate for a long time.

Protein Disulfide Isomerase and Its Potential Function on Endoplasmic Reticulum Quality Control in Diatom Phaeodactylum tricornutum

PDI is a molecular chaperone and plays an important role in Endoplasmic Reticulum quality control (ERQC).PDI participates in the refolding of the misfolded/unfolded proteins to maintain cellular homeostasis under differentstresses. However, bioinformatic characteristics and potential functions of PDIs in diatom Phaeodactylumtricornutum (Pt) are still unknown so far. Hence, the genome-wide characteristics of PtPDI proteins in P. tricornutumwere first studied via bioinformatic and transcriptomic methods. 42 PtPDI genes were identified from thegenome of P. tricornutum. The motif, protein structure, classification, number of introns, phylogenetic relationship,and the expression level of 42 PtPDI genes under the tunicamycin stress were analyzed. A pair of tandemduplicated genes (PtPDI15 and PtPDI18) was observed in P. tricornutum. The 42 PtPDIs with different genecharacteristics were divided into three independent clades, indicating different evolutional relationships and functionsof these PtPDIs. The 14 up-regulated PtPDI genes under the tunicamycin treatment might have a positiveeffect on the ER quality control of the unfolded/misfolded proteins, while the 7 down-regulated PtPDIs mightnegatively affect the ERQC. The characteristics of all 42 PtPDIs and their proposed working model here providea comprehensive understanding of the PtPDIs gene family. The differential expression of 21 PtPDIs will be usefulfor further functional study in the ERQC.

Genome-Wide Identification, Characterization, and Expression Analysis of Calmodulin-Like Proteins(CMLs) in Apple

Calcium(Ca^(2+)), a crucial second messenger in plants, is involved in diverse signaling pathways including biotic and abiotic stress responses. However, the functions of most calcium sensors including calcium-dependent protein kinases(CDPKs), calmodulins(Ca Ms), calmodulinlike proteins(CMLs), and calcineurin B-like proteins(CBLs) remain limited in plants, particularly in woody plants. Currently, a total of 83 CMLs and seven Ca Ms were discovered in the apple genome in this study. Functional domain analysis showed that these CMLs and Ca Ms contain a highly conserved EF-hand motif. q RT-PCR assays indicated that these CMLs were expressed ubiquitously in apple, including in the roots, stems,leaves, flowers, and fruits, and also possessed spatial specificity. Finally, most of these CMLs were induced by hormonal and abiotic stress, suggesting their potential roles in the regulation of growth, development, and the stress response in apple. In summary, our findings provide potential evidence that apple CMLs may be involved in abiotic stress and the regulation of plant growth and development.

T cells engineered with chimeric antigen receptor （CAR） have been successfully applied to treat advanced refractory B cell malignancy. However, many challenges remain in extending its application toward the treatment of solid tumors. The immunosuppressive nature of tumor microenvironment is considered one of the key factors limiting CART efficacy. One negative regulator of T cell activity is lymphocyte activation gene-3 （LAG-3）. We successfully generated LAG-3 knockout T and CAR-T cells with high efficiency using CRISPR-Cas9 mediated gene editing and found that the viability and immune phenotype were not dramatically changed during in vitro culture. LAG-3 knockout CART cells displayed robust antigen-specific antitumor activity in cell culture and in murine xenograft model, which is comparable to standard CAR-T cells. Our study demonstrates an efficient approach to silence immune checkpoint in CAR-T cells via gene editing.
