Inspired by our previous studies to discover novel human immunodeficiency virus-1(HIV-1)nonnucleoside reverse transcriptase inhibitors(NNRTIs)by targeting the tolerant region II of the NNRTIs binding pocket(NNIBP),a series of novel benzo[4,5]thieno[2,3-d]pyrimidine derivatives were designed through structure-based drug design as novel potent HIV-1 *** results showed that compound16b was the most active inhibitor,exhibiting 50% effective concentration(EC50)values from 0.021μmol/L to 0.298μmol/L against wild-type(WT)and a panel of NNRTIs-resistant HIV-1 ***,16b was demonstrated with a significantly low 50% cytotoxicity concentration(CC_(50))value(>200μmol/L)and high selectivity index(SI)*** addition,16b yielded moderate reverse transcriptase(RT)enzyme inhibition with a 50% inhibition concentration(IC_(50))value of 0.183μmol/L,which demonstrated that it acted as HIV-1 *** binding mode of 16b with RT was also illustrated via molecular ***,this work provided a novel lead compound for developing potent HIV-1 NNRTIs.

AIM：To determine the epidemiological characteristics,clinical signs,laboratory findings,and outcomes in patients with corneal infection in shandong peninsula of china.·METHODS：The medical records of 578 inpatients（578eyes）with corneal infection were reviewed retrospectively for demographic characteristics,risk factors,seasonal variation,clinical signs,laboratory findings,and treatment *** history,ocular examination findings using slit-lamp biomicroscopy,laboratory findings resulted from microbiological cultures,and treatment.·RESULTS：Fungal keratitis constituted 58.48%of cases of infectious keratitis among the inpatients,followed by herpes simplex keratitis（20.76%）,bacterial keratitis（19.03%）and acanthamoeba keratitis（1.73%）.The most common risk factor was corneal trauma（71.80%）.The direct microscopic examination（338 cases）using potassium hydroxide（KOH）wet mounts was positive in296 cases（87.57%）.Among the 298 fungal culturepositive cases,Fusarium species were the most common isolates（70.47%）.A total of 517 cases（89.45%）received surgical intervention,including 255（44.12%）cases of penetrating keratoplasty,74（12.80%）cases of lamellar keratoplasty which has become increasingly popular,and77 cases（13.32%）of evisceration or enucleation.·CONCluSION：At present,infectious keratitis is a primary corneal disease causing blindness in *** Fusarium species being the most commonly identified pathogens,fungal keratitis is the leading cause of severe infectious corneal ulcers in shandong peninsula of china.

Drug resistance remains to be a serious problem with type Ⅰ human immunodeficiency virus(HIV-1) nonnucleoside reverse transcriptase inhibitors(NNRTIs). A series of novel boronic acid-containing diarylpyrimidine(DAPY) derivatives were designed via bioisosterism and scaffold-hopping strategies,taking advantage of the ability of a boronic acid group to form multiple hydrogen bonds. The target compounds were synthesized and evaluated for their anti-HIV activities and cytotoxicity in MT-4 *** 10 j yielded the most potent activity and turned out to be a single-digit nanomolar inhibitor towards the HIV-1 ⅢB [wild-type(WT) strain], L100 I and K103 N strains, with 50% effective concentration(EC_(50)) values of 7.19–9.85 nmol/L. Moreover, 10 j inhibited the double-mutant strain RES056 with an EC_(50) value of 77.9 nmol/L, which was 3.3-more potent than that of EFV(EC_(50)= 260 nmol/L) and comparable to that of ETR(EC_(50)= 32.2 nmol/L). 10j acted like classical NNRTIs with high affinity for WT HIV-1 reverse transcriptase(RT) with 50% inhibition concentration(IC_(50)) value of 0.1837 μmol/L. Furthermore,molecular dynamics simulation indicated that 10 j was proposed as a promising molecule for fighting against HIV-1 infection through inhibiting RT activity. Overall, the results demonstrated that 10 j could serve as a lead molecule for further modification to address virus-drug resistance.

AIM： To establish a new, reliable vomit model of minks METHODS： Adult male minks were randomly divided into 8 groups （n=6）： cisplatin （7.5 mg/kg） intraperitoneal injection （ip） group, copper sulfate （40 mg/kg） intragastric injection （ig） group, apomorphine （1.6 mg/kg） subcutaneous injection （sc） group, and 18 Gy whole-body X-irradiation group, ondansetron injection group （2 mg/kg ip） 30 min later followed by cisplatin （7.5 mg/kg） ip, normal saline （NS） ip injection control group, metoclopramide injection group （4 mg/kg ip） 30 rain later followed by apomorphine （1.6 mg/kg） sc, NS ig control group. The frequency of retching and vomiting was calculated. After behavioral experiment, distribution of 5-HT in the ileum was detected by immunohistologic method. RESULTS： Cisplatin, apomorphine, copper sulfate and X-irradiation administered to minks evoked a profound emetic response in the animals. However, retching and vomiting were significantly inhibited by pretreatment with ondansetron and metoclopramide in cisplatin and copper sulfate groups （P=0.018）. Immunohistologic result showed that 5-HT released from enterochromaffin cells （EC cells） was involved in vomiting mechanism. CONCluSION： Mink vomit model has a great value in studying the vomiting mechanism and screening new antiemetic drugs.

HIV-1 capsid protein(CA) has emerged as a promising target for antiviral treatment considering its structural and regulatory roles in HIV-1 replication. Here, we disclose the design, synthesis, biological assessment, and mechanism investigation of a novel series of phenylalanine derivatives gained by further structural modification of PF74. The newly synthesized compounds demonstrated potent anti-HIV activity, represented by 7n displayed anti-HIV-1 activity 6.25-fold better than PF74, and 7h showed anti-HIV-2activity with nearly 139 times improved efficacy over PF74. Surface plasmon resonance(SPR) studies of representative compounds proved that HIV-1 CA was the binding target. Competitive SPR studies using CPSF6 and NUP153 peptides identified that 7n binds to a vital CA assembly interface between the Nterminal and C-terminal domain(NTD-CTD interface). Action stage determination assay revealed that the newly synthesized compounds were antiviral with a dual-stage inhibitory profile. Molecular dynamics(MD) simulations offered the crucial foundation for the hopeful antiviral potency of 7n. Besides, 7m and7n modestly increased metabolic stabilities in human liver microsome(HLM) and human plasma compared to PF74. Overall, these studies offer valuable insights and can regard as the beginning for succedent medicinal chemistry endeavors to discover promising HIV capsid inhibitors with improved efficacy and better drug-like characteristics.

Effect of gingerol on substance P and NK1 receptor expression in a vomiting model of mink

Background Gingerol is the generic term for pungent constituents in ginger, which has been reported to be effective for inhibiting vomiting. We attempted to investigate the antiemetic effect of gingerol and its effective mechanism on substance P and NK1 receptors in minks. Methods The antiemetic effect of gingerol was investigated during a 6-hour observation on a vomiting model in minks induced by cisplatin, （7.5 mg/kg, intraperitoneal）. The distribution of substance P and NK1 receptors in the area postrema and ileum were measured by immunohistochemistry, and the expression of NK1 receptor in the area postrema and ileum were measured by Western blotting. Results The frequency of cisplatin induced retching and vomiting was significantly reduced by pretreatment with gingerol in a dose-dependent manner （P 〈0.05）. Substance P-immunoreactive was mainly situated in the mucosa and submucosa of the ileum as well as in the neurons of the area postrema. The immunoreactive production of NK1 receptor was mainly situated in the muscular and submucosa of ileum and the neurons of area postrema, gingerol markedly suppressed the increased immunoreactivity of substance P and NK1 receptor induced by cisplatin in a dose-dependent manner （P 〈0.05）, and exhibited effective inhibition on the increased expression levels of NK1 receptor in both the ileum and area postrema dose-dependently （P 〈0.05）. Conclusions Gingerol has good activity against cisplatin-induced emesis in minks possibly by inhibiting central or peripheral increase of substance P and NK1 receptors.

Using e^(+)e^(−)annihilation data corresponding to an integrated luminosity of 2.93 fb^(−1)taken at the center-of-mass energy√s=3.773 GeV with the BESIII detector,a joint amplitude analysis is performed on the decays D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η).The fit fractions of individual components are obtained,and large interferences among the dominant components of the decays D^(0)→a_(1)(1260)π,D^(0)→π(1300)π,D^(0)→ρ(770)ρ(770),and D^(0)→2(ππ)_(S)are observed in both *** the obtained amplitude model,the CP-even fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are determined to be(75.2±1.1_(stat).±1.5_(syst.))%and(68.9±1.5_(stat).±2.4_(syst.))%,*** branching fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are measured to be(0.688±0.010_(stat.)±0.010_(syst.))%and(0.951±0.025_(stat.)±0.021_(syst.))%,*** amplitude analysis provides an important model for the binning strategy in measuring the strong phase parameters of D^(0)→4πwhen used to determine the CKM angleγ(ϕ_(3))via the B^(−)→DK^(−)decay.
