EVI2A has emerged as a significant biomarker in various diseases;however,its biological role and mechanism in kidney renal clear cell carcinoma(KIRC)remains *** used TCGA and GEO databases to analyze EVI2A gene expression comprehensively and performed pan-cancer *** relevance was evaluated through Kaplan-Meier analysis and ROC *** gene’s immune relevance was explored through analyses of the tumor microenvironment(TME),Tumor Immune Single-cell Hub(TISCH),immune checkpoints,and immunotherapy *** results indicate that EVI2A expression is upregulated in KIRC,showing correlations with tumor grade and T/N/M ***2A demonstrates high diagnostic accuracy(AUC=0.906)and predicts poor overall and progression-free survival in KIRC ***,EVI2A expression exhibits significant associations with immunity,including TME scores and specific immune cell types such as Tfh cells,CD4 memory T cells,and CD8+T *** EVI2A expression suggests increased sensitivity to PD-1/CTLA-4 and tyrosine kinase *** vitro assays confirmed the impact of EVI2A on KIRC behavior,with its knockdown resulting in reduced cell proliferation and *** conclusion,our comprehensive analysis identifies EVI2A as a promising biomarker and a novel therapeutic target for intervening in *** findings hold significant implications for further research and potential clinical applications.

Effects of dietary astaxanthin supplementation on growth performance, antioxidant status, immune response, and intestinal health of rainbow trout(Oncorhynchus mykiss)

A feeding trial was conducted to assess the impacts of dietary astaxanthin from wall-broken Haematococcus pluvialis(WBHPA) on the growth performance, antioxidant status, immune response, and intestinal health of rainbow trout(Oncorhynchus mykiss). Six experimental diets were formulated with various concentrations of WBHPA, ranging from 0 to 8.4 g/kg(containing 0 to 125 mg/kg astaxanthin). Each diet was fed to triplicate groups of rainbow trout(mean initial weight of 561 g) twice daily for 9 consecutive weeks. The survival rate and feed intake of fish exhibited no significant differences among the dietary groups(P > 0.05). Similarly, dietary inclusion of 25 to 100 mg/kg astaxanthin did not significantly affect the weight gain and daily growth coefficient(P > 0.05), but excessive inclusion of astaxanthin(125 mg/kg) slightly depressed these parameters(P < 0.05). Dietary inclusion of 25 to 50 mg/kg astaxanthin increased the activities of intestinal digestion and absorption enzymes(lipase, creatine kinase, and alkaline phosphatase), while the inclusion of 25 to 75 mg/kg astaxanthin improved the immune response of fish. Furthermore, regardless of inclusion level(25 to 125 mg/kg), dietary astaxanthin supplementation strengthened the intestinal mucosal barrier function and improved antioxidant activity,thereby promoting intestinal development. Conclusively, 25 to 75 mg/kg astaxanthin from WBHPA was recommended to be included in diets for rainbow trout.

Sirtuin 3(SIRT3)is well known as a conserved nicotinamide adenine dinucleotide^(+)(NAD^(+))-dependent deacetylase located in the mitochondria that may regulate oxidative stress,catabolism and ATP *** evidence has recently revealed that SIRT3 plays its critical roles in cardiac fibrosis,myocardial fibrosis and even heart failure(HF),through its deacetylation ***,discovery of SIRT3 activators and elucidating their underlying mechanisms of HF should be urgently ***,we identified a new small-molecule activator of SIRT3(named 2-APQC)by the structure-based drug designing strategy.2-APQC was shown to alleviate isoproterenol(ISO)-induced cardiac hypertrophy and myocardial fibrosis in vitro and in vivo rat ***,in SIRT3 knockout mice,2-APQC could not relieve HF,suggesting that 2-APQC is dependent on SIRT3 for its protective ***,2-APQC was found to inhibit the mammalian target of rapamycin(mTOR)-p70 ribosomal protein S6 kinase(p70S6K),c-jun N-terminal kinase(JNK)and transforming growth factor-β(TGF-β)/small mother against decapentaplegic 3(Smad3)pathways to improve ISO-induced cardiac hypertrophy and myocardial *** upon RNA-seq analyses,we demonstrated that SIRT3-pyrroline-5-carboxylate reductase 1(PYCR1)axis was closely assoiated with *** activating PYCR1,2-APQC was shown to enhance mitochondrial proline metabolism,inhibited reactive oxygen species(ROS)-p38 mitogen activated protein kinase(p38MAPK)pathway and thereby protecting against ISO-induced mitochondrialoxidative ***,activation of SIRT3 by 2-APQC could facilitate AMP-activated protein kinase(AMPK)-Parkin axis to inhibit ISO-induced ***,our results demonstrate that 2-APQC is a targeted SIRT3 activator that alleviates myocardial hypertrophy and fibrosis by regulating mitochondrial homeostasis,which may provide a new clue on exploiting a promising drug candidate for the future HF therapeutics.

In the boreal summer and autumn of 2023,the globe experienced an extremely hot period across both oceans and *** consecutive record-breaking mean surface temperature has caused many to speculate upon how the global temperature will evolve in the coming 2023/24 boreal *** this report,as shown in the multi-model ensemble mean(MME)prediction released by the Institute of Atmospheric Physics at the Chinese Academy of Sciences,a medium-to-strong eastern Pacific El Niño event will reach its mature phase in the following 2−3 months,which tends to excite an anomalous anticyclone over the western North Pacific and the Pacific-North American teleconnection,thus serving to modulate the winter climate in East Asia and North *** some uncertainty due to unpredictable internal atmospheric variability,the global mean surface temperature(GMST)in the 2023/24 winter will likely be the warmest in recorded history as a consequence of both the El Niño event and the long-term global warming ***,the middle and low latitudes of Eurasia are expected to experience an anomalously warm winter,and the surface air temperature anomaly in China will likely exceed 2.4 standard deviations above climatology and subsequently be recorded as the warmest winter since ***,the necessary early warnings are still reliable in the timely updated mediumterm numerical weather forecasts and sub-seasonal-to-seasonal prediction.

Bananas(Musa spp.)are monocotyledonous plants with high genetic diversity in the Musaceae family that are cultivated mainly in tropical and subtropical *** fruits are a popular food,and the plants themselves have diverse *** genetic groups(genomes)are thought to have contributed to current banana cultivars:Musa acuminata(A genome),Musa balbisiana(B genome),Musa schizocarpa(S genome),and species of the Australimusa section(T genome).However,the T genome has not been effectively ***,we present the high-quality TT genomes of two representative accessions,Abaca(Musa textilis),with high-quality naturalfiber,and Utafun(Musa troglodytarum,Fe’i group),with abundant *** the Abaca and Utafun assemblies comprise 10 pseudochromosomes,and their total genome sizes are 613 Mb and 619 Mb,*** genome analysis revealed that the larger size of the T genome is likely attributable to rapid expansion and slow removal of *** with those of Musa AA or BB accessions or sisal(Agava sisalana),Abacafibers exhibit superior mechanical properties,mainly because of their thicker cell walls with a higher content of cellulose,lignin,and *** of MusaCesA cellulose synthesis genes peaks earlier in Abaca than in AA or BB accessions during plant development,potentially leading to earlier cellulose accumulation during secondary cell wall *** Abaca-specific expressed gene MusaMYB26,which is directly regulated by MusaMYB61,may be an important regulator that promotes precocious expression of secondary cell wall ***,MusaWRKY2 and MusaNAC68,which appear to be involved in regulating expression of MusaLAC and MusaCAD,may at least partially explain the high accumulation of lignin in *** work contributes to a better understanding of banana domestica-tion and the diverse genetic resources in the Musaceae family,thus providing resources for Musa genetic improvement.

Wide-bandgap mixed-halide perovskite solar cells(WBG-PSCs)are promising top cells for efficient tandem photovoltaics to achieve high power conversion efficiency(PCE)at low ***,the open-circuit voltage(V_(OC))of WBG-PSCs is still unsatisfactory as the V_(OC)-deficit is generally larger than 0.45 ***,we report a buried interface engineering strategy that substantially improves the V_(OC)of WBG-PSCs by inserting amphiphilic molecular hole-selective materials featuring with a cyanovinyl phosphonic acid(CPA)anchoring group between the perovskite and *** assembly and redistribution of CPA-based amphiphilic molecules at the perovskite-substrate buried interface not only promotes the growth of a low-defect crystalline perovskite thin film,but also suppresses the photo-induced halide phase *** energy level alignment between wide-bandgap perovskite and the hole-selective layer is further improved by modulating the substituents on the triphenylamine donor moiety(methoxyls for MPA-CPA,methyls for Me PA-CPA,and bare TPA-CPA).Using a 1.68 e V bandgap perovskite,the Me PA-CPA-based devices achieved an unprecedentedly high V_(OC)of 1.29 V and PCE of 22.3%under standard AM 1.5 *** V_(OC)-deficit(yers in PSCs.

An analog of phthalascidin （Pt-650） was synthesized with an improved synthetic route. With precursor 2 as the starting material, compound 1 was prepared through 4 steps in a total yield of 47%. In vitro antitumor test of this Pt-650 analog showed that it possessed strong toxicity against a number of tumor cell lines including A2780, A549, Bel-7402, BGC-823, HELA, KB, KeTr3 and HCT-8.
