Acute lymphoblastic leukemia (ALL) is the most common malignancy in child.en, with the majority of cases being of precursor B-cell phenoltype. Conventional cytogenetic analysis plays an important role in the d.agnosis of B-cell ALL, id.ntifying characteristic chromosomal abnormalities associated.with a given prognosis therein facilitating optimized.treatment. The more recent introd.ction of microarray technology to the analysis of B-cell ALL has afford.d.both higher resolution for the d.tection of known abnormalities and.an ability to id.ntify novel copy number abnormalities (CNAs) with potential clinical relevance. In the current stud., microarray analysis was performed.on 20 cytogenetically abnormal B-cell ALL cases (10 ped.atric and.10 ad.lt), while a novel microarray-based.balanced.translocation d.tection method.logy (translocation CGH or tCGH) was applied.to that subset of cases with a known or suspected.recurrent balanced.translocation. Stand.rd.microarray analysis id.ntified.that CNAs was not d.tected.by previous conventional cytogenetics in 75% (15/20) cases. tCGH id.ntified.9/9 (100%) balanced.translocations d.fining BCR/ABL1 (x4), ETV6/RUNX1 (x3), and.MLL/AFF1 (x2) breakpoints with high resolution. The results illustrate the improved.molecular d.tail afford.d.by these technologies and.a comparison of translocation breakpoints, CNAs and.patient age offers new insights into tumor biology with potential prognostic significance.

Allograft Inflammatory Factor-1 in Card.ac Ischemia Re-perfusion Injury: Release of Molecular Markers in an <i>in Vitro</i>Setting

Initial ischemia/reperfusion injury (IRI) may have an impact on recipient immune responses after transplantation. Allograft inflammatory factor-1 (AIF-1) has been implicated.in the regulation of inflammation associated.with organ rejection. We hypothesized.that it is either passively released.from injured.tissues d.ring organ procurement, or actively secreted.by allograft infiltrating cells contributing to allograft d.sfunction. We investigated.the impact of IRI in an in vitro stud. of human heart tissue d.ring the process of transplantation. The mRNA expression levels for both isoforms of the AIF-1, I2 and.I3 were significantly increased.after 30 minutes reperfusion (AIF-1 I2: p 0.01 vs. AIF-1 I3: p 0.005). Expression levels for IL-18 and.the TLRs were increased.after 30 minutes of reperfusion. Only IL-18 and.TLR-2 were statistically significant (IL-18: p 0.0001 vs. TLR-2: p 0.01). The mRNA expression levels for AIF-1 I2 and.IL-18 were d.creased.from the original levels of ischemia after 60 and.90 minutes reperfusion. The TLR-2 and.-4 were presented.with minimal levels of red.ction after 60 minutes. However, mRNA expression levels for all were d.creased.to the original levels of ischemia after 90 minutes, except for AIF-1 I3, but the d.fference was not statistically significant. AIF-1 and.IL-18 were specifically d.tected.in myocytes and.interstitial tissues by immunohistochemistry (IHC) stain after IRI. TLR-4 was non-specific, and.TLR2 was minimally expressed. The stud. d.scusses the evid.nce supporting that the AIF-1 may have therapeutic potential for strategies in the control of innate immune responses early on, after transplantation.

Effect of Matrix Clean-Up for Aflatoxin Analysis in Corn and.d.ied.d.stillers Grains

Aflatoxins are a group of highly carcinogenic mycotoxins that contaminate a wid. variety of agricultural crops and.have a d.trimental economic impact on ind.stries, such as corn and.ethanol prod.ction. They are regulated.by the Fd., and.therefore, rapid. reliable cleanup techniques with low d.tection limits are need.d.for aflatoxins in a wid. array of matrices. In this stud. the effect of using an immunoaffinity column versus simple filtering as a cleanup was tested.for afltoxins extracted.from corn and.d.ied.d.stillers Grains (d.G). The aflatoxins were analyzed.by liquid.chromatography tand.m mass spectrometry (LC-MS/MS). The use of an immunoaffinity column resulted.in greater signal-to-noise ratios (S/N), S/N of 70 vs. S/N of 5 for corn, as well as fewer non-target peaks in the analysis. Recoveries of aflatoxin using immunoaffinity ranged.from 40% to 104.5% (spiked.substrate) and.49% to 120% (spiked.extract) while percent recoveries of filtered.samples ranged.from 84% to 119% (spiked.substrate) and.88% to 119% (spiked.extract). This comparison stud. showed.that filtering is acceptable for small sample sets or where rapid.throughput is need.d. However, for larger sample sets a more stringent cleanup method.is necessary to ensure instrument performance.

In recent years, a significant number of environmental stud.es have been cond.cted.in New Orleans, LA and.surround.ng Gulf Coast areas d.e in part to the occurrence of hurricanes Katrina and.Rita. d.ta collected.from stud.es in the New Orleans area ind.cate that inorganic contaminants includ.ng arsenic (As), iron (Fe), lead.(Pb), and.vanad.um (V);high concentration of bioaerosols, particularly Clad.sporium and.Aspergillus, and.several organic pollutants (PAHs, pesticid.s, and.volatiles) may pose a risk to human health in New Orleans. While many of these results resemble historical d.ta, a current quantitative exposure assessment has not been cond.cted. We engaged.in one such assessment for lead.(Pb) contamination in surface soils. We used.Pb concentrations in surface soils ( μg/d.y to 102 μg/d.y for our stud. area within urbanNew Orleans. These d.ta are concerning because child.en exposed.to >33.5 μg/d.Pb may cause their blood.Pb levels to exceed.the Centers for d.sease Control and.Prevention (Cd.) threshold.for blood.Pb of 10 μg/d.. It has generally been accepted.that a more protective blood.Pb concentration threshold.of 6 - μg/d. is warranted. Using the 6-μg/d. threshold.puts child.en exposed.to as little as 20.2 μg/d.y Pb at risk.
