The pervasive effects of lighting environments on sensory drive in bluefin killifish： an investigation into male/male competition, female choice, and predation

Sensory drive predicts that the conditions under which signaling takes place have large effects on signals, sensory systems, and behavior. The coupling of an ecological genetics approach with sen sory drive has been fruitful. An ecological genetics approach compares populations that experi ence different environments and asks whether population differences are adaptive and are the result of genetic and/or environmental variation. The multifaceted effects of signaling environ ments are wellexemplified by the bluefin killifish. In this system, males with blue anal fins are abundant in tanninstained swamps that lack UV/blue light but are absent in clear springs where UV/blue light is abundant. Past work indicates that lighting environments shape genetic and envir onmental variation in color patterns, visual systems, and behavior. Less is known about the select ive forces creating the across population correlations between UV/blue light and the abundance of blue males. Here, we present three new experiments that investigate the roles of lighting environ ments on male competition, female mate choice, and predation. We found strong effects of lighting environments on male competition where blue males were more likely to emerge as dominant in teastained water than in clear water. Our preliminary study on predation indicated that blue males may be less susceptible to predation in teastained water than in clear water. However, there was little evidence for female preferences favoring blue males. The resulting pattern is one where the effects of lighting environments on genetic variation and phenotypic plasticity match the direction of selection and favor the expression of blue males in swamps.

Capacities in Facing Natural Hazards: A Small Island Perspective

Isolated communities on small islands are often characterized as vulnerable and marginalized. We studied the recent history of Laingpatehi, a village on Ruang Island off the north coast of Sulawesi, Indonesia to show that the marginalization-vulnerability nexus can be offset by capacity and social cohesion to enable sustainable livelihoods. The island has been impacted by volcanic eruptions,earthquakes, and competition for marine resources from mainland-based fishermen. The community has shown a remarkable ability to cope and prosper in the face of a series of external hazards. We used a sustainable livelihoods approach to identify the assets that enabled the villagers to cope. Strong social cohesion was central to the ability to organize the community and confront hazards. A diversified livelihood strategy drawing on the small island environment and its coastal and marine resources, income generating activities in a distant satellite village, and significant remittances from employment in other parts of Indonesia underpinned people’s capacities to face *** assistance played a supporting role. The case of Laingpatehi demonstrates how remoteness, rather than being a source of vulnerability, can provide access to existing resources and facilitate innovation. Disaster risk reduction strategies should focus more on reinforcing these existing capacities to deal with hazards and less on physical protection and postdisaster responses.

Damage to peripheral nerve tissue may cause loss of function in both the nerve and the targeted muscles it innervates. This study compared the repair capability of engineered nerve conduit （ENC）, engineered fibroblast conduit （EFC）, and autograft in a 10-mm tibial nerve gap. ENCs were fabricated utilizing primary fibroblasts and the nerve cells of rats on embryonic day 15 （E 15）. EFCs were fabricated utilizing primary fi- broblasts only. Following a 12-week recovery, nerve repair was assessed by measuring contractile properties in the medial gastrocnemius muscle, distal motor nerve conduction velocity in the lateral gastrocnemius, and histology of muscle and nerve. The autografts, ENCs and EFCs reestablished 96%, 87% and 84% of native distal motor nerve conduction velocity in the lateral gastrocnemius, 100%, 44% and 44% of native specific force of medical gastrocnemius, and 63%, 61% and 67% of native medial gastrocnemius mass, re- spectively. Histology of the repaired nerve revealed large axons in the autograft, larger but fewer axons in the ENC repair, and many smaller axons in the EFC repair. muscle histology revealed similar muscle fiber cross-sectional areas among autograft, ENC and EFC repairs. In conclusion, both ENCs and EFCs promot- ed nerve regeneration in a 10-mm tibial nerve gap repair, suggesting that the El5 rat nerve cells may not be necessary for nerve regeneration, and EFC alone can suffice for peripheral nerve injury repair.

Background: Active video games(AVGs) encourage whole body movements to interact or control the gaming system, allowing the opportunity for skill development. Children with autism spectrum disorder(ASD) show decreased fundamental movement skills in comparison with their typically developing(TD) peers and might benefit from this approach. This pilot study investigates whether playing sports AVGs can increase the actual and perceived object control(OC) skills of 11 children with ASD aged 6–10 years in comparison to 19 TD children of a similar *** was a secondary ***: Actual(Test of Gross motor Development) and perceived OC skills(Pictorial Scale of Perceived movement Skill Competence for Young Children) were assessed before and after the intervention(6 × 45 min).Results: Actual skill scores were not improved in either group. The ASD group improved in perceived skill. All children completed the required dose and parents reported the intervention was ***: The use of AVGs as a play-based intervention may not provide enough opportunity for children to perform the correct movement patterns to influence skill. However, play of such games may influence perceptions of skill ability in children with ASD, which could improve motivation to participate in physical activities.

In about half the cases of involuntary childlessness, a male infertility factor is involved. The PIWI-LIKE genes, a subclade of the Argonaute protein family, are involved in RNA silencing and transposon control in the germline. Knockout of murine Piwi-like 1 and 2 homologs results in complete infertility in males. The aim of this study was to analyze whether the mRNA expression of human PIWI-LIKE I-4 genes is altered in ejaculated spermatozoa of men with impaired sperm characteristics. Ninety male participants were included in the study, among which 47 were with normozoospermia, 36 with impaired semen characteristics according to the World Health Organization （WHO） manual, 5th edition, and 7 with azoospermia serving as negative control for the PIWI-LIKE 1-4 mRNA expression in somatic cells in the ejaculate. PIWI-LIKE 1-4 mRNA expression in the ejaculated spermatozoa of the participants was measured by quantitative real-time PCR. In nonazoospermic men, PIWIoLIKE 1-4 mRNA was measurable in ejaculated spermatozoa in different proportions. PIWI-LIKE 1 （100.0%） and PIWI-LIKE 2 （49.4%） were more frequently expressed than PIWI-LIKE3 （9.6%） and PIWI-LIKE 4 （15.7%）. Furthermore, a decreased PIWI-LIKE2 mRNA expression showed a significant correlation with a decreased sperm count （P = 0.022） and an increased PIWI-LIKE 1 mRNA expression with a decreased progressive motility （P = 0.048）. PIWI-LIKE 1 and PIWI-LIKE 2 mRNA expression exhibited a significant association with impaired sperm characteristics and may be a useful candidate for the evaluation of the impact of PIWI-LIKE 1-4 mRNA expression on male infertility.

Purpose:The study aimed to investigate the role of training load characteristics and injury and illness risk in youth ski ***:The training load characteristics as well as traumatic injuries,overuse injuries,and illnesses of 91 elite youth ski racers(age=12.1±1.3 years,mean±SD)were prospectively recorded over a period of 1 season by using a sport-specific online *** linear regression analyses were performed to monitor the influence of training load on injuries and *** in mean training load characteristics between preseason,in-season,and post-season were calculated using multivariate analyses of ***:Differences were discovered in the number of weekly training sessions(p=0.005)between pre-season(4.97±1.57)and post-season(3.24±0.71),in the mean training volume(p=0.022)between in-season(865.8±197.8 min)and post-season(497.0±225.5 min)and in the mean weekly training intensity(Index)(p=0.012)between in-season(11.7±1.8)and post-season(8.9±1.7).A total of 185 medical problems were reported(41 traumatic injuries,12 overuse injuries,and 132 illnesses).The weekly training volume and training intensity was not a significant risk factor for injuries(p>0.05).Training intensity was found to be a significant risk factor for illnesses in the same week(β=0.348;p=0.044;R^(2)=0.121)and training volume represents a risk factor for illnesses in the following week(β=0.397;p=0.027;R^(2)=0.157).Conclusion:A higher training intensity and volume were associated with increased illnesses,but not with a higher risk of *** training and ensuring appropriate progression of training load between weeks may decrease incidents of illness in-season.

Spontaneous Ca2+ oscillations in vascular smooth muscle cells have been modeled using a single Ca2+ pool. This report describes spontaneous Ca2+ oscillations dependent on two separate Ca2+ sources for the nuclear versus cytoplas- mic compartments. Changes in free intracellular Ca2+ were monitored with ratiometric Ca2+- fluorophores using confo- cal microscopy. On average, spontaneous oscillations developed in 79% of rat aortic smooth muscle cells that were synchronous between the cytoplasm and nucleus. Reduction of extracellular Ca2+ (m) decreased the frequency and amplitude of the cytoplasmic oscillations with 48% of the oscillations asynchronous between the nuclear and cytoplasmic compartments. Similar results were obtained with the Ca2+ channel blockers, nimodipine and diltiazem. Arg-vasopressin (AVP) induced a rapid release of intracellular Ca2+ stores that was greater in the nuclear compartment (4.20 ± 0.23 ratio units, n = 56) than cytoplasm (2.54 ± 0.28) in cells that had spontaneously developed prior oscillations. Conversely, cells in the same conditions lacking oscillations had a greater AVP-induced Ca2+ transient in the cytoplasm (4.99 ± 0.66, n = 17) than in the nucleus (2.67 ± 0.29). Pre-treatment with Ca2+ channel blockers depressed the AVP responses in both compartments with the cytoplasmic Ca2+ most diminished. Depletion of internal Ca2+ stores prior to AVP exposure blunted the nuclear response, mimicking the response of cells that lacked prior oscillations. Spontaneous oscillating cells had a greater sarcoplasmic reticulum network than cells that did not oscillate. We propose that sponta- neous nuclear oscillations rely on perinuclear sarcoplasmic reticulum stores, while the cytoplasmic oscillations rely on Ca2+ influx.
