Synergistic defensive mechanism of phytochelatins and antioxidative enzymes in Brassica chinensis L. against Cd stress

Brassica chinensis L. was chosen and exposed to different concentrations of Cd exposure to evaluate its Cd-accumulating capacity and its potential cellular defensive mechanisms. Cd accumulation in the shoots and roots of B. chinensis was up to 1348.3±461.8 and 3761.0±795.0 mg per killogram of dry weight, respectively, under 200 μmol/L of Cd exposure. Increasing Cd accumulation in the plant was accompanied by rapid accumulation of phytochelatins (PCs), and the sequestration of Cd by PCs provided a primary cellular mechanism for Cd detoxification and tolerance of B. chinensis. Furthermore, malondialdehyde formation, hydrogen peroxide content and antioxidative enzyme activities such as superoxide dismutase, catalase, guaiacol peroxidase and ascorbate peroxidase were observed in the shoots of Cd-stressed B. chinensis. Increasing enzyme activities in response to concentrations of 5 to 50 μmol/L Cd showed an efficient defense against oxidative stress, suggesting that the antioxidative system was a secondary defensive mechanism. These resulted in reduced free Cd damage and enhanced Cd accumulation and tolerance. Glutathione plays a pivotal role in these two detoxification pathways. In general, these results suggested that PCs and the antioxidative system are synergistic in combatting Cd-induced oxidative stress and that they play important roles in Cd detoxification of B. chinensis, and also give a deep understanding of the natural defensive mechanisms in plants under heavy metal stress.

目的观察吡非尼酮对大鼠心肌纤维化的作用,并探讨其机制。方法选取8周龄SPF级雄性SD大鼠24只,构建大鼠心房纤维化模型,随机分为假手术组、模型组、吡非尼酮低剂量组和吡非尼酮高剂量组,每组6只。假手术组和模型组分别经尾静脉注射生理盐水和异丙肾上腺素,吡非尼酮低剂量组和吡非尼酮高剂量组在注射异丙肾上腺素的基础上分别灌胃不同剂量的吡非尼酮。Masson染色检测心肌纤维化程度,免疫组织化学染色检测胶原组织Ⅰ型(Collogen-1)、核苷酸结合寡聚化结构域样受体蛋白3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)炎性小体、半胱氨酸天冬氨酸蛋白酶1(cysteinyl aspartate-specific proteinase-1,Caspase-1)的表达,Western blot检测Collogen-1和心房消皮素D(gasdermin D,GSDMD)的表达。结果与假手术组比较,模型组心肌纤维化改变显著,Collogen-1、NLRP3、Caspase-1和GSDMD表达显著升高,差异有统计学意义(P<0.05);与模型组比较,吡非尼酮低剂量组和吡非尼酮高剂量组心肌纤维化程度显著减轻,Collogen-1、NLRP3、Caspase-1和GSDMD表达显著降低,差异有统计学意义[(8.14±1.40)%,(6.56±0.75)%vs(22.15±2.57)%,P<0.05;0.14±0.03 vs 0.33±0.05,0.42±0.13,P<0.05;(10.34±1.40)%,(10.33±3.40)%vs(23.22±1.99)%,P<0.05;(15.67±0.56)%,(17.33±0.78)%vs(22.87±1.92)%,P<0.05;0.43±0.06,0.46±0.11 vs 0.65±0.03,P<0.05]。结论吡非尼酮通过NLRP3/Caspase-1/GSDMD信号轴抑制心肌细胞焦亡,减轻心肌纤维化。

随着信号传输速率的提升,对多层印制电路板(PCB)背钻的要求越来越高。对背钻对位方式、背钻盖板和背钻专用钻头3方面展开研究,建立背钻对位方式的误差计算模型,分析不同盖板对背钻堵孔的影响,并设计了一款渐变式芯厚和渐变式沟幅比的背钻专用钻头。通过上述措施,改善背钻流程,简化图形电镀和碱性蚀刻工艺流程,得到厚径比为19∶1,最小通孔孔径为0.20mm,背钻孔径0.35mm,背钻深度3.60mm的背钻孔。确保达到经电镀处理,直接背钻不堵孔的品质标准。

Pressure ulcers(PUs)are common skin injuries known for their high morbidity,rapid onset,susceptibility to infection,and challenging healing *** potential therapy for PUs is cell-based therapy using mesenchymal stem cells(MSCs).However,poor survival and low cell retention of MSCs on skin lesions limit their therapeutic effects and *** this study,we prepared an extracellular matrix(dECM)hydrogel decellularized from the human umbilical cord(UC).A patch composed of UC-dECM and UC-MSCs was employed in the treatment of PUs in C57BL/6 *** results indicate that the UC-dECM hydrogel effectively sustains cell viability,enhances the stemness-related gene expression in UC-MSCs,and promotes human umbilical vein endothelial cells(HUVECs)migration and *** to the groups treated with the patch containing only UC-dECM,injection of UC-MSCs or gauze dressing,the patch combining UC-dECM hydrogel with UC-MSCs significantly accelerated PU *** positive outcome can be attributed to the promotion of tissue re-epithelialization,collagen deposition,angiogenesis,and inflammation *** results suggest that the composite patch,comprised of UC-dECM hydrogel and UC-MSCs,may be a promising therapeutic approach for PU treatment.

A forensic validation study of the Early Access HuaxiaTM Platinum Polymerase Chain Reaction (PCR) kit was completed to document the performance capabilities and *** genotyping of DNA samples was consistent across a large range of template DNA concentrations,with complete profiles obtained at 0.125 ng;however,no more than 2 mm× 1.2 mm punches of samples would be recommended for direct *** size precision and accuracy test revealed the genotyping ability;while consistent results were obtained when comparing the kit with other commercially available *** addition,the whole PCR amplification can finish within approximately 45 min,making the system suitable for ***,only partial profiles may be obtained with challenging samples,including DNA stored on Foam-Tipped Applicators (FTA) cards or some case *** the forensic application in ethnic groups,a total of 282 and 229 alleles were obtained in Han and Mongolia,*** the 23 short tandem repeats were independent from each other,the cumulative power of exclusion in duos was 0.999999157188 and the cumulative power of exclusion in trios was 0.999999999859 in the Han group while the cumulative power of exclusion in duos (CPEduo) was 0.999 998 848 26 and cumulative power of exclusion in trios (CPEtrio) was 0.999 999 999 79 in the Mongolia *** good internal consistency was found between the two investigated groups and the Sichuan Han,Hui,Tibetan and Uygur according to available reference data.

Short tandem repeat(STR)profiling is one of the mostly used systems for forensic *** certain circumstances,STR profiling is time-consuming and costly,which potentially leads to delays in criminal ***(Laboratory of the Government Chemist,UK)Forensics has developed a robust STR profiling platform called the ParaDNAVR Intelligence Test System which can provide early tactical intelligence and aid investigators in making informed decisions on sample prioritization for ***,we validated the ParaDNA^(■) intelligence test for its application in forensic cases using a range of mock evidence items following guidelines set by the Scientific Working Group on DNA Analysis Methods(SWGDAM).Specifically,we tested the sensitivity and accuracy of the ParaDNA intelligence test,as well as the success rates for detecting mock samples and for use in case *** findings demonstrate that the ParaDNA intelligence test generates useful DNA profiles,especially for samples such as blood,saliva,and semen that contain ample DNA,indicating the benefits of including ParaDNA as a prior step in forensic STR profiling pipelines.

Women’s Awareness and Attitudes towards Labor Analgesia Influencing Practice between Developed and Developing Countries

Childbirth experience is one of the most intense pain that majority of women will endure during their lifetime. Concerns about pain in labor remain a hot topic, and its popularity gets more common day by day as more women become aware of their rights to achieve a better quality of care during labor. There are various non-pharmacologic (transcutaneous electrical nerve stimulation, hydrotherapy, intradermal water injections and acupuncture) and pharmacologic treatments (nitrous oxide, opioids and regional analgesia techniques: spinal, epidural and combined epidural analgesia) available today. Among these, epidural analgesia offers the most effective form of pain relief and is considered to be the gold standard of labor analgesia. Despite having labor analgesic services, most women still go through painful labor due to lack of knowledge regarding it, particularly in developing countries. The main source of information regarding pain reliefs is from friends and relatives, revealing the lack of information from caregiver’s side. So this study reflects that there is a wide gap in the communication between pregnant women and obstetricians. It supports the fact that obstetricians through the practice of routinely offering labor analgesia can significantly improve the maternal and perinatal outcomes of pregnancy. Provision of standardized epidural analgesia information at an appropriate time in their pregnancy may benefit them by the practice of mutual decision-making. Thus, it may prevent women from making a difficult choice of cesarean section to avoid the fear of painful labor.
