Hereditary spherocytosis （HS）, the most common cause of congenital hemolytic anemia, is caused by deficiency of the ery- throcyte membrane proteins. Five causative genes （ANK1, SPTB, SPTA1, SLC4AI, and EPB42） have been identified. To date, molecular genetic studies have been performed in different populations, including the American, European, Brazilian, Japanese and Korean populations, whereas only a few studies have been described in the Chinese population. Here, by reanalysis of the exome data, we revealed causative mutations and established a definitive diagnosis of HS in all 38 Chinese families. We found 34 novel mutations and four reported mutations in three known HS-causing genes--17 in ANK1, 17 in SPTB and four in SLC4A1, suggesting that ANK1 and SPTB are the major genes in Chinese patients with HS. All of the ANK1 or SPTB mutations, scattered throughout the entire genes, are non-recurrent; and most of them are null mutations, which might cause HS via a hap-loinsufficiency mechanism. De novo mutations in ANK1 or SPTB often occur with an unexpected high frequency （87.5% and 64.2%, respectively）. Our study updates our knowledge about the genetic profile of HS in Chinese and shows that family-based, especially parent-offspring trio, sequencing analysis can help to increase the diagnostic power and improve diagnostic efficiency.

Oleic acid-capped lanthanum borate(abbreviated as OA/La BO_3·H_2O) nanosheets were prepared by the hydrothermal method. The microstructures of as-prepared OA/La BO_3·H_2O were characterized by means of SEM, TEM, EDS, FTIR and XRD, respectively. Moreover, the friction and wear properties of OA/La BO_3·H_2O as a lubricant additive in rapeseed oil were evaluated on a four-ball tribotester. The tribochemical characteristics of worn surfaces were investigated by SEM and XPS. The results showed that the hydrophobic OA/La BO_3·H_2O nanosheets exhibited their morphology with a diameter in the range of 100 nm to 300 nm and a thickness of about 25 nm, and displayed excellent dispersing stability in rapeseed oil. In the meantime, the rapeseed oil doped with OA/La BO_3·H_2O nanosheets markedly decreased the friction and wear of steel balls, and the optimal friction-reducing and antiwear ability of rapeseed oil was obtained at an OA/La BO_3·H_2O content of 1.0%. The outstanding tribological performance of OA/La BO_3·H_2O in rapeseed oil was attributed to the formation of a composite boundary lubrication film mainly composed of lubricious tribochemical species of B_2O_3, La_2O_3 and Fe_2O_3, and deposits of OA/La BO_3·H_2O nanosheets as well as the adsorbates of rapeseed oil on rubbed surfaces.

利用镀自组装薄膜的双峰谐振长周期光纤光栅（LPFG）实现了高灵敏的折射率传感,给出了提高镀膜双峰谐振光纤光栅对环境折射率（SRI）灵敏度的途径。确定最佳镀膜厚度,使光纤包层模位于模式转换区附近;调整光纤半径,使镀膜后光栅的双峰位于相位匹配转折点（PMTP）附近,给出了确定最优膜厚和光纤半径的方法。将不同半径的双峰光栅镀自组装聚丙烯胺盐酸盐（PAH）/聚丙烯酸（PAA）薄膜后,放入不同浓度的葡萄糖溶液中进行折射率传感实验。结果表明：通过优化光纤半径和薄膜厚度,镀膜后工作于模式转换区附近和PMTP附近的双峰光栅对1.333-1.372范围内折射率的灵敏度高达3985nm/RIU（RIU为单位折射率）,明显高于不在模式转换区或PMTP附近的光栅,亦高于已报道的非双峰镀膜长周期光纤光栅。

Effects of Nitrogen-Containing Biodegradation Enhancers on Sorption of n-Hexadecane in Soil-Water System

Effects of nitrogen-containing biodegradation enhancers(methyl diethanolamine oleate(MDEAO) and oleic diethanolamide(ODEA)) on the adsorption of n-hexadecane in soil were studied by laboratory batch experiments. The partition coefficients(K_d values) of n-hexadecane sorption in soil-water system and those partition coefficients normalized to organic matter(K_(oc) values) were both determined. The adsorption isotherm curve of n-hexadecane in soil-water system was plotted. The measured results demonstrated that K_(oc) values changed in the soil-water system with different enhancers but are relatively invariant for the soil at the same site. The K_d values rose with the increase of the organic matter content in different soil. The average values of K_(oc) in the soil-water system with MDEAO, ODEA,and blank soil were 0.412, 0.252,and 0.309, respectively. The critical micelle concentration of ODEA and MDEAO was 0.7 mg/L and 1.9 mg/L, respectively,denoting that the solubilization capacity of ODEA was much stronger than MDEAO in the soil-water system. Consequently,the adsorption of MDEAO onto the solid surface increased the organic matter content in soil,which could make it more effective in enhancing the n-hexadecane adsorption. On the contrary, ODEA could inhibit the adsorption of n-hexadecane because of its less adsorption rate onto the soil particle surface and higher concentration in the soil-water system. According to the correlation coefficients, it was found that both the Henry linear and the Freundlich nonlinear isotherm sorption models were fitted to the data very well, however the Freundlich model was better than the Henry model.

Influence of nozzle enabling strategy on spray deposition of crop protection unmanned aerial system

Proper nozzles arrangement is significant to improve spray deposition of crop protection unmanned aerial system(UAS).Besides fuselage structure,the control strategy is another nozzle location changing method when there are multiple sets of nozzles.A four-rotor crop protection UAS equipped with four centrifugal nozzles was used to conduct a field experiment in the rupturing stage of *** sets of nozzles in the front and rear of the fuselage were enabled independently to investigate spray deposition,including spray coverage and droplet density on the sampling *** nozzle rotating speeds and flight speeds were employed in the experiment to study the influence of nozzle location on the *** different nozzle rotating speeds,the droplet spectrum could be *** results show that the average coverage and average cumulative droplet density are negatively correlated with flight *** droplet density is also negatively correlated with the droplet *** deposition of front nozzles is significantly reduced compared with that of the rear nozzles under the same spray parameters,especially when the droplet size is too large or the flight speed is too *** reduction is mainly concentrated in the center area of the spray *** a result,the average cumulative droplet density of the front nozzles decreases by 25.96%in *** average droplet density decreases by 18.54%when the droplet size is smaller than 100μm,decreases by 25.02%when between 100μm and 200μm,and decreases by more than half when larger than 200μ*** research can provide guidance for the installation of UAS nozzles and spray control strategy design.

Development and characterization of interspecific hybrids between Oryza sativa and O. latifolia by in situ hybridization

Oryza sativa and O. latifolia belong to the AA and CCDD genomes of Oryza, respectively. In this study, interspecific hybrids of these species were obtained using the embryo rescue technique. Hybrid panicle traits, such as long awns, small grain, exoteric large purple stigma, grain shattering and dispersed panicles, resemble that of the paternal parent, O. latifolia, whereas there is obvious heterosis in such respects as plant height, tillering ability and vegetative vigor. Chromosome pairing and the genomic components of the hybrid were subsequently investigated using genomic in situ hybridization (GISH) and fluorescent in situ hybridization (FISH) analysis. Based on the mitotic metaphase chromosome numbers of the root tips investigated, the hybrid is a triploid with 36 chromosomes. The genomic constitution of the hybrid is ACD. In the meiotic metaphase I of the hybrid pollen mother cell, poor chro- mosome pairing was identified and most of the chromosomes were univalent, which resulted in com- plete male sterility in the hybrid.

Mounting evidence indicates that amyloid β protein（Aβ） exerts neurotoxicity by disrupting the blood-brain barrier（BBB） in Alzheimer＇s disease. Hyperoside has neuroprotective effects both in vitro and in vivo against Aβ. Our previous study found that hyperoside suppressed Aβ1-42-induced leakage of the BBB, however, the mechanism remains unclear. In this study, bEnd.3 cells were pretreated with 50, 200, or 500 μM hyperoside for 2 hours, and then exposed to Aβ1-42 for 24 hours. Cell viability was determined using 3-（4,5-dimethyl-2-thiazolyl）-2,5-diphenyl-2-H-tetrazolium bromide assay. Flow cytometry and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay were used to analyze cell apoptosis. Western blot assay was carried out to analyze expression levels of Bax, Bcl-2, cytochrome c, caspase-3, caspse-8, caspase-9, caspase-12, occludin, claudin-5, zonula occludens-1, matrix metalloproteinase-2（MMP-2）, and MMP-9. Exposure to Aβ1-42 alone remarkably induced bEnd.3 cell apoptosis; increased ratios of cleaved caspase-9/caspase-9, Bax/Bcl-2, cleav ed caspase-8/caspase-8, and cleaved caspase-12/caspase-12; increased expression of cytochrome c and activity of caspase-3; diminished levels of zonula occludens-1, claudin-5, and occludin; and increased levels of MMP-2 and MMP-9. However, hyperoside pretreatment reversed these changes in a dose-dependent manner. Our findings confirm that hyperoside alleviates fibrillar Aβ1-42-induced BBB disruption, thus offering a feasible therapeutic application in Alzheimer＇s disease.

Cloning and expression of a cDNA encoding ribosomal protein S4 from Rice (Oryza sativa)

A cDNA clone, pS4, has been isolated from a cDNA library prepared from rice anthers of about 1.0 mm in length. DNA sequence analysis and database search show that the cDNA encodes a protein which is highly homologous to eukaryotic 80S ribosomal protein subunit 4 (S4). Northern hybridization indicates that this gene expresses in all tissues analyzed although the expression level varies and it cannot be induced by mechanical wounding in leaves.
