Lytic polysaccharide monooxygenases(LPMOs)are important enzymes that boost the hydrolysis of recalcitrant polysaccharides,such as chitin.They are found extensively in different insect species and are classified as auxiliary activities family 15(AA15)LPMOs(LPMO15).Some of them were identified from the insect midgut and proven to act on chitin.However,knowledge about their physiological roles during insect growth and development remains limited.Here,we found that midgut-specific LPMO15s are widely distributed in different insect orders,such as the orthopteran Locusta migratoria and the lepidopteran Bombyx mori.Using L.migratoria as a model insect,the function of midgut-specific LmLPMO15-3 during development was investigated.Double-stranded RNA-mediated downregulation of LmLPMO15-3 expression at the 4th or 5th instar nymph stage severely decreased the survival rate and resulted in lethal phenotypes.Hematoxylin and eosin staining results indicated that the deficient individuals exhibited incompletely digested peritrophic matrix(PM),which suggested that LmLPMO15-3 is essential for the deconstruction of the PM during molting.This study provides direct evidence of the physiological importance of a midgut-specific LPMO15 during insect development.As L.migratoria is one of the most destructive agricultural pests,LmLPMO15-3 is a potential target for pest management.

Human carboxylesterases:a comprehensive review

Mammalian carboxylesterases(CEs) are key enzymes from the serine hydrolase superfamily.In the human body, two predominant carboxylesterases(CES1 and CES2) have been identified and extensively studied over the past decade. These two enzymes play crucial roles in the metabolism of a wide variety of endogenous esters, ester-containing drugs and environmental toxicants. The key roles of CES in both human health and xenobiotic metabolism arouse great interest in the discovery of potent CES modulators to regulate endobiotic metabolism or to improve the efficacy of ester drugs. This review covers the structural and catalytic features of CES, tissue distributions, biological functions, genetic polymorphisms, substrate specificities and inhibitor properties of CES1 and CES2, as well as the significance and recent progress on the discovery of CES modulators. The information presented here will help pharmacologists explore the relevance of CES to human diseases or to assign the contribution of certain CES in xenobiotic metabolism. It will also facilitate medicinal chemistry efforts to design prodrugs activated by a given CES isoform, or to develop potent and selective modulators of CES for potential biomedical applications.

Fullerene bisadduct stabilizers:The effect of different addition positions on inhibiting the autocatalytic decomposition of nitrocellulose absorbed nitroglycerin

To explore the effect of different positions and number of pyrrolidine bound to the carbon cage on the stabilization effect of fulleropyrrolidine derivatives to nitrocellulose(NC)/nitroglycerine(NG),we synthesized N-(4-methoxy)phenylpyrrolidine-C_(60) and four different of bis(N-(4-methoxy)phenylpyrrolidine)-C_(60) compounds through Prato reaction.Their structures were characterized by UVevis,^(1)H NMR,^(13)C NMR,high-resolution mass spectroscopy,and single-crystal X-ray diffraction.Their stabilization effect to NC/NG were investigated using differential scanning calorimetry,methyl violet,vacuum stabilization effect,weight loss,and accelerating rate calorimeter tests.The results indicated these compounds had excellent stabilization effect to NC/NG.The stabilization effect of the fulleropyrrolidine bisadducts to NC/NG is significantly better than that of fulleropyrrolidine monoadduct and C_(60).Moreover,the position where pyrrolidine binds to fullerene in fulleropyrrolidine bisadducts is different,and its stabilization effect to NC is also different.The stabilization effect order of different bisadduct isomers to nitrocellulose is as follows:e-edge>trans-2>cis-2>trans-3.Electron paramagnetic resonance(EPR)and FT-IR were used to study the stabilization mechanism of fulleropyrrolidine derivatives to NC/NG.The EPR results also show that fulleropyrrolidine bisadducts with different addition sites have different abilities to absorb nitroxide,and their ability is better than that of the monoadduct and C_(60),which is consistent with the results of stabilization effect performance test.

Evaluating the impact of sampling schemes on leaf area index measurements from digital hemispherical photography in Larix principis-rupprechtii forest plots

Background:Digital hemispherical photography(DHP)is widely used to estimate the leaf area index(LAI)of forest plots due to its advantages of high efficiency and low cost.A crucial step in the LAI estimation of forest plots via DHP is choosing a sampling scheme.However,various sampling schemes involving DHP have been used for the LAI estimation of forest plots.To date,the impact of sampling schemes on LAI estimation from DHP has not been comprehensively investigated.Methods:In this study,13 commonly used sampling schemes which belong to five sampling types(i.***.dispersed,square,cross,transect and circle)were adopted in the LAI estimation of five Larix principis-rupprechtii plots(25m×25 m).An additional sampling scheme(with a sample size of 89)was generated on the basis of all the sample points of the 13 sampling schemes.Three typical inversion models and four canopy element clumping index(Ωe)algorithms were involved in the LAI estimation.The impacts of the sampling schemes on four variables,including gap fraction,Ωe,effective plant area index(PAIe)and LAI estimation from DHP were analysed.The LAI estimates obtained with different sampling schemes were then compared with those obtained from litter collection measurements.Results:Large differences were observed for all four variable estimates(i.***.gap fraction,Ωe,PAIe and LAI)under different sampling schemes.The differences in impact of sampling schemes on LAI estimation were not obvious for the three inversion models,if the fourΩe algorithms,except for the traditional gap-size analysis algorithm were adopted in the estimation.The accuracy of LAI estimation was not always improved with an increase in sample size.Moreover,results indicated that with the appropriate inversion model,Ωe algorithm and sampling scheme,the maximum estimation error of DHP-estimated LAI at elementary sampling unit can be less than 20%,which is required by the global climate observing system,except in forest plots with extremely large LAI values(~>6.0).However,obtaining an LAI from DHP with an

An Expedient Method for Regioselective Methylation of Catechol Coumarins

An expedient and efficient method for selective methylation of catechol coumarins by working with different alkalis under appropriate reaction conditions was reported. Esculetins were selectively methylated at position 6 and position 7 in good yields using CH3I in the presence of Nail and Na2CO3, respectively. However, daphnetins showed less regioselectivity under the same reaction conditions. Furthermore, the site preference for the methylation reaction was interpreted by the density functional theory at B3LYP/6-31 ＋G（d） level.

Objective:This study aimed to investigate whether perceived stress mediated the relationship between hope and anxiety/depression symptoms among patients with COVID-19 during the epidemic.In addition,the potential moderating effect of coping styles was examined.Methods:From February 26 to March 10,2020,patients with COVID-19 were asked to complete a questionnaire online,which included demographic characteristics,as well as the SCL-90-Anxiety,SCL-90-Depression,Chinese Perceived Stress Scale(CPSS),Herth Hope Index(HHI),and Trait Coping Style Questionnaire(TCSQ).Hierarchical linear regression was performed to explore independent factors of anxiety/depression.A multi-group structural equation modeling with the collected data from patients in the Negative Coping style(NC)group and Positive Coping style(PC)group was used to test the hypothesized mechanism.Results:In total,382 valid questionnaires of patients were obtained,including 96 from NC patients and 286 from PC patients.In the hierarchical linear regression,hope and perceived stress were independent risk factors for both anxiety and depression in the total sample and PC group.However,hope was not independently related to anxiety/depression in the NC group.As hypothesized,the hope of patients had significant and negative indirect effects on both anxiety and depression that were mediated by perceived stress,However,the direct effect from stress on anxiety and depression was stronger for NC patients than for PC patients.Besides,hope had significant direct effects on anxiety/depression in PC patients,but not in NC patients.Conclusion:During the COVID-19 epidemic,perceived stress could mediate the relationship between hope and anxiety/depression symptoms among COVID-19 patients,with coping style moderating this cultivation process.

目的:研究RBM47对下游基因HMGA2的调控作用,以及对人慢性髓系白血病细胞株K562增殖的调控作用。方法:使用过表达、敲低RBM47的慢病毒载体感染K562细胞,CCK-8法检测RBM47对K562细胞增殖的影响,流式细胞术检测RBM47对K562细胞周期的影响;通过RNA免疫共沉淀结合方法检测RBM47与HMGA2mRNA的结合情况,逆转录实时定量PCR方法检测RBM47对HMGA2 mRNA稳定性的影响,Western blot检测RBM47对HMGA2蛋白表达的影响。结果:在K562细胞内过表达RBM47可抑制K562细胞的增殖和周期运行,抑制RBM47则可显著促进K562细胞增殖和细胞周期运行;RBM47可通过与HMGA2 mRNA结合促进其降解;过表达RBM47可显著降低K562细胞内HMGA2蛋白的表达。结论:RNA结合蛋白RBM47通过调控HMGA2抑制K562细胞的增殖。

目的以办公空间形态的转变及职场人更高层面的健康体验追求为背景,在办公家具“健康化”的发展趋势下,探究办公“第三空间”中健康家具的设计路径。方法分析办公“第三空间”特性与职场人的健康需求,以生理与精神两类需求为引线,基于体验设计视角与案例研究,根据感官、情感、行为、认知、社交五个体验模块构建办公“第三空间”健康家具设计模型,并指出当前家具在满足职场人精神健康层面中心理疗愈、认知提升与个人实现需求的不足。结果提出在办公“第三空间”家具设计中融入与“家”相关的情感、创造多元化的互动方式、结合兴趣引导社群活动的办公“第三空间”家具设计优化路径,通过不同体验模块的叠加作用增加家具在情感关怀、认知促进与自我实现方面的效用。结论满足办公者更深层次的健康需求,赋予家具更深刻的健康内涵,提升办公“第三空间”家具体验价值。
