DNA methylation has been identified as a powerful and activity-dependent regulator of changes in the brain that may underlie neuroadaptations in response to various types of stimuli,including exposure to drugs of ***,the medial prefrontal cortex(mPFC)projections to the nucleus accumbens(NAc)are critically important for reinstated cocaine-seeking in a rodent model of cocaine *** circuitry undergoes several epigenetic modifications following cocaine exposure,including changes in DNA methylation that are associated with drug-seeking *** have previously shown that methyl supplementation via l-Methionine(MET)administration attenuates cocaine-seeking behavior and reverses expression and methylation patterns of the immediate early gene c-fos,suggesting that MET may act by altering the excitability of this circuitry during cocaine *** the current study,male rats were microinjected with an adeno-associated virus overexpressing halorhodopsin in the mPFC,optical fibers were surgically implanted into the NAc,and the rats were given injections of MET *** underwent acquisition of cocaine self-administration(0.75 mg/kg/infusion,2-h sessions)followed by extinction training in the absence of drug-paired *** reinstatement tests were conducted:cue-induced reinstatement without optogenetic manipulations and cocaine-primed reinstatement with optogenetic inhibition of mPFC-to-NAc *** were no group differences before the cocaine-primed reinstatement session,and all groups showed robust cue-induced *** rats treated with MET and rats that received mPFC-to-NAc inhibition showed an abolishment of cocaine-primed reinstatement,suggesting that systemic methyl supplementation may act through this critical circuity.

Repeatability of decision-making behaviour in male threespine stickleback Gasterosteus aculeatus： Effects of dummy vs. live stimuli

Individuals select from a number of behaviours when responding to various situations and the decisions they make may affect their fitness. The costs and benefits of these responses vary among individuals causing them to differ even in identical situations. One example of this type of situation is when territorial males encounter both a male and female simultaneously, gene- rating a trade-off that likely leads to individual differences due to differing costs of various actions among males. This situation commonly occurs in threespine stickleback, Gasterosteus aculeatus. However, for selection to act effectively, individuals must behave in a consistent manner and measuring repeatability can aid in understanding how selection may shape such trade-offs. Males of this species exhibit consistent individual differences in their response to dummy males and females but it is unknown ff patterns are similar when feedback from the stimuli is present. To assess this, male threespine stickleback were tested with dummy and live male and female conspecifics, presented separately and simultaneously. While the same trends were found re- gardless of stimulus type, males were more aggressive towards the live conspecifics than to the dummies. Repeatability values were similar within a treatment regardless of whether live or dummy conspecifics were used, suggesting that individuals show the same level of consistency. This study adds to our understanding of consistent individual differences by demonstrating that feed- back may not affect responses to conflicting stimuli and that male threespine stickleback respond in a consistent manner to both dummy and live stimuli

Australian sugarcane soldier fly’s salivary gland transcriptome in response to starvation and feeding on sugarcane crops

The soldier fly is an endemic pest of sugarcane in *** numbers of larvae can cause significant damage to roots and reduce the crop *** is known about the composition and function of the soldier fly salivary gland,its secretions,and their roles in insect—plant *** this study,we performed transcriptome analysis of the salivary glands of starved and sugarcane root-fed soldier fly larvae.A total of 31119 highly expressed assembled contigs were identified in the salivary glands and almost 50%of them showed high levels of similarity to known proteins in Nr *** all the obtained contigs,only 9727 sequences contain an open reading frame of over 100 amino *** 31%of contigs were predicted to encode secretory proteins,including some digestive and detoxifying enzymes and potential *** known salivary secreted peptides such as serine protease,cysteine proteinase inhibitors,antimicrobial peptides and venom proteins were among the top 100 highly expressed *** gene expression analysis revealed significant modulation of 850 transcripts in salivary glands upon exposure to plant roots or starvation ***,we identified some venom proteins which were significantly upregulated in the salivary glands of soldier fly larvae exposed to sugarcane *** other insects and nematodes some of these proteins have been used to manipulate host plant defense systems and facilitate the invasion of the host *** findings provide a further insight into the identification of potential effector proteins involved in soldier fly-sugarcane interactions.

Nanoscience is not physics, chemistry, engineering or biology. It is all of them, and it is time for a text that integrates the disciplines. This is such a text, aimed at advanced undergraduates and beginning graduate students in the sciences. The consequences of smallness and quantum behaviour are well known and described Richard Feynman''s visionary essay''There''s Plenty of Room at the Bottom''(which is reproduced in this book). Another, critical, but thus far neglected, aspect of nanoscience is the complexity of nanostructures. Hundreds, thousands or hundreds of thousands of atoms make up systems that are complex enough to show what is fashionably called''emergent behaviour''. Quite new phenomena arise from rare configurations of the system. Examples are the Kramer''s theory of reactions (Chapter 3), the Marcus theory of electron transfer (Chapter 8), and enzyme catalysis, molecular motors, and fluctuations in gene expression and splicing, all covered in the final Chapter on Nanobiology. The book is divided into three parts. Part I (The Basics) is a self-contained introduction to quantum mechanics, statistical mechanics and chemical kinetics, calling on no more than basic college calculus. A conceptual approach and an array of examples and conceptual problems will allow even those without the mathematical tools to grasp much of what is important. Part II (The Tools) covers microscopy, single molecule manipulation and measurement, nanofabrication and self-assembly. Part III (Applications) covers electrons in nanostructures, molecular electronics, nano-materials and nanobiology. Each chapter starts with a survey of the required basics, but ends by making contact with current research literature.

Unveiling T cell evasion mechanisms to immune checkpoint inhibitors in acute myeloid leukemia

Acute myeloid leukemia (AMl) is a heterogeneous and aggressive hematologic malignancy that is associated with a high relapse rate and poor prognosis. Despite advances in immunotherapies in solid tumors and other hematologic malignancies, AMl has been particularly difficult to treat with immunotherapies, as their efficacy is limited by the ability of leukemic cells to evade T cell recognition. In this review, we discuss the common mechanisms of T cell evasion in AMl: (1) increased expression of immune checkpoint molecules;(2) downregulation of antigen presentation molecules;(3) induction of T cell exhaustion;and (4) creation of an immunosuppressive environment through the increased frequency of regulatory T cells. We also review the clinical investigation of immune checkpoint inhibitors (ICIs) in AMl. We discuss the limitations of ICIs, particularly in the context of T cell evasion mechanisms in AMl, and we describe emerging strategies to overcome T cell evasion, including combination therapies. Finally, we provide an outlook on the future directions of immunotherapy research in AMl, highlighting the need for a more comprehensive understanding of the complex interplay between AMl cells and the immune system.

Surface roughness plays an important role in microwave remote *** the agricultural domain,surface roughness is crucial for soil moisture retrieval methods that use electromagnetic surface scattering or microwave radiative transfer ***,improved characterization of Soil Surface Roughness(SSR)is of considerable *** this study,three approaches,including a standard pin profiler,a liDAR point cloud generated from an iPhone 12 Pro,and a Structure from Motion(SfM)photogrammetric point cloud,were applied over 24 surface profiles with different roughness variations to measure surface *** objective of this study was to evaluate the capability of smartphone-based liDAR technology to measure surface roughness parameters and compare the results of this technique with the more common *** showed that the iPhone liDAR technology,when point cloud data is captured in a fine-resolution mode,has a significant correlation with SfM photogrammetry(R2=0.70)and a relatively close agreement with pin profiler(R2=0.60).However,this accuracy tends to be greater for random surfaces and rough profiles with row structure *** results of this study confirm that smartphone-based liDAR can be used as a cost-effective,fast,and time-efficient alternative tool for measuring surface roughness,especially for rough,wide,and inaccessible areas.

Next Generation lubricants and Their Potential Use in Fertility Treatment

While generally recognized as a potential source of contamination during the collection process, lubricants are often used at the preference of the male partner to prevent irritation. While older lubricants have been studied, there is currently no conscience within programs as to what constitutes a “safe” lubricant. The object of the current study was designed to evaluate chemically unique “next generation” lubricants in comparison to lubricants currently in use in fertility treatment and/or recognized as fertility safe;the first was a silicone-based lubricant, the second is a water-based, plant-based organic compound, in comparison to two established medical lubricants and a control. Twelve deidentified semen samples from the clinical andrology laboratory were used to test the lubricants following semen analysis. In order to enter the study, the sample had to have a minimum of 30 × 106 motile cells. Samples were then processed using a simple sperm wash modified to reconstitute the pellet into a final volume of 7 ml. Half-milliliter aliquots were then transferred into 13 wells of a standard 24-well culture plate. One well was used as a control. The remaining wells received one of the four lubricants at one of three volumes (10, 50, or 100 ul), producing 12 treatment combinations (four lubricants + 3 concentration levels) and the control. The samples were then cultured at room temperature for 24 hours. At times 0, 1, 3, 12, and 24 hrs, the plate was agitated to remix the sample, and a 4 ul aliquot of each well was analyzed for standard semen parameters using a computer-assisted semen analyzer. Results indicated the expected decrease in semen parameters over time in all treatments (P lso a dose-dependent drop in most of the lubricants. However, samples contaminated with the newer lubricants appeared to maintain semen parameters similar to the controls at all but the 100 ul level of contamination, while the older lubricants caused decrea
