Optogenetic inhibition of medial prefrontal cortex projections to the nucleus accumbens core and methyl supplementation via l-Methionine attenuates cocaine-primed reinstatement

DNA methylation has been identified as a powerful and activity-dependent regulator of changes in the brain that may underlie neuroadaptations in response to various types of stimuli,including exposure to drugs of ***,the medial prefrontal cortex(mPFC)projections to the nucleus accumbens(NAc)are critically important for reinstated cocaine-seeking in a rodent model of cocaine *** circuitry undergoes several epigenetic modifications following cocaine exposure,including changes in DNA methylation that are associated with drug-seeking *** have previously shown that methyl supplementation via l-Methionine(MET)administration attenuates cocaine-seeking behavior and reverses expression and methylation patterns of the immediate early gene c-fos,suggesting that MET may act by altering the excitability of this circuitry during cocaine *** the current study,male rats were microinjected with an adeno-associated virus overexpressing halorhodopsin in the mPFC,optical fibers were surgically implanted into the NAc,and the rats were given injections of MET *** underwent acquisition of cocaine self-administration(0.75 mg/kg/infusion,2-h sessions)followed by extinction training in the absence of drug-paired *** reinstatement tests were conducted:cue-induced reinstatement without optogenetic manipulations and cocaine-primed reinstatement with optogenetic inhibition of mPFC-to-NAc *** were no group differences before the cocaine-primed reinstatement session,and all groups showed robust cue-induced *** rats treated with MET and rats that received mPFC-to-NAc inhibition showed an abolishment of cocaine-primed reinstatement,suggesting that systemic methyl supplementation may act through this critical circuity.

Australian sugarcane soldier fly’s salivary gland transcriptome in response to starvation and feeding on sugarcane crops

The soldier fly is an endemic pest of sugarcane in *** numbers of larvae can cause significant damage to roots and reduce the crop *** is known about the composition and function of the soldier fly salivary gland,its secretions,and their roles in insect—plant *** this study,we performed transcriptome analysis of the salivary glands of starved and sugarcane root-fed soldier fly larvae.A total of 31119 highly expressed assembled contigs were identified in the salivary glands and almost 50%of them showed high levels of similarity to known proteins in Nr *** all the obtained contigs,only 9727 sequences contain an open reading frame of over 100 amino *** 31%of contigs were predicted to encode secretory proteins,including some digestive and detoxifying enzymes and potential *** known salivary secreted peptides such as serine protease,cysteine proteinase inhibitors,antimicrobial peptides and venom proteins were among the top 100 highly expressed *** gene expression analysis revealed significant modulation of 850 transcripts in salivary glands upon exposure to plant roots or starvation ***,we identified some venom proteins which were significantly upregulated in the salivary glands of soldier fly larvae exposed to sugarcane *** other insects and nematodes some of these proteins have been used to manipulate host plant defense systems and facilitate the invasion of the host *** findings provide a further insight into the identification of potential effector proteins involved in soldier fly-sugarcane interactions.

Repeatability of decision-making behaviour in male threespine stickleback Gasterosteus aculeatus： Effects of dummy vs. live stimuli

Individuals select from a number of behaviours when responding to various situations and the decisions they make may affect their fitness. The costs and benefits of these responses vary among individuals causing them to differ even in identical situations. One example of this type of situation is when territorial males encounter both a male and female simultaneously, gene- rating a trade-off that likely leads to individual differences due to differing costs of various actions among males. This situation commonly occurs in threespine stickleback, Gasterosteus aculeatus. However, for selection to act effectively, individuals must behave in a consistent manner and measuring repeatability can aid in understanding how selection may shape such trade-offs. Males of this species exhibit consistent individual differences in their response to dummy males and females but it is unknown ff patterns are similar when feedback from the stimuli is present. To assess this, male threespine stickleback were tested with dummy and live male and female conspecifics, presented separately and simultaneously. While the same trends were found re- gardless of stimulus type, males were more aggressive towards the live conspecifics than to the dummies. Repeatability values were similar within a treatment regardless of whether live or dummy conspecifics were used, suggesting that individuals show the same level of consistency. This study adds to our understanding of consistent individual differences by demonstrating that feed- back may not affect responses to conflicting stimuli and that male threespine stickleback respond in a consistent manner to both dummy and live stimuli

Purpose To measure changes in strength,power,and agility over consecutive seasons based on competition level in figure *** A total of 197 competitive singles figure skaters(age range=9-25 years old)completed combines with United States Figure Skating on two consecutive seasons between 2011 and *** either remained in the same competitive on-ice level or increased by one level on consecutive *** athletes completed the hexagon agility test,maximal verti-cal jump,timed tuck jumps,push-ups,and bent knee ***-measures analyses of variance separated by sex with Tukey’s post hoc were performed for each dependent variable and Cohen’s d effect sizes were calculated for all significant *** Female skaters who remained in the same level demonstrated improved performance on vertical jump(3.66 cm higher),timed tuck jumps(3.77 more jumps),v-ups(1.59 more v-ups),and hexagon jump(1.17 s faster).There was an interaction for female skaters who remained in the same level(P=0.004),with senior and junior levels outperforming inter-mediate level skaters on the hexagon jump *** skaters who increased level demonstrated improved performance on the vertical jump(2.24 cm higher),hexagon jump(1.16 cm faster),and tuck jump(3.03 more jumps).Male skaters did not demonstrate any changes in performance except v-ups(2.86 more v-ups)for those who increased *** The greatest changes were in lower level female skaters who remained in the same level,supporting the impor-tance of participating in strength and conditioning programs earlier in skating careers to maximize athleticism before reaching the senior level,when changes in off-ice performance plateau.

Unveiling T cell evasion mechanisms to immune checkpoint inhibitors in acute myeloid leukemia

Acute myeloid leukemia (AMl) is a heterogeneous and aggressive hematologic malignancy that is associated with a high relapse rate and poor prognosis. Despite advances in immunotherapies in solid tumors and other hematologic malignancies, AMl has been particularly difficult to treat with immunotherapies, as their efficacy is limited by the ability of leukemic cells to evade T cell recognition. In this review, we discuss the common mechanisms of T cell evasion in AMl: (1) increased expression of immune checkpoint molecules;(2) downregulation of antigen presentation molecules;(3) induction of T cell exhaustion;and (4) creation of an immunosuppressive environment through the increased frequency of regulatory T cells. We also review the clinical investigation of immune checkpoint inhibitors (ICIs) in AMl. We discuss the limitations of ICIs, particularly in the context of T cell evasion mechanisms in AMl, and we describe emerging strategies to overcome T cell evasion, including combination therapies. Finally, we provide an outlook on the future directions of immunotherapy research in AMl, highlighting the need for a more comprehensive understanding of the complex interplay between AMl cells and the immune system.

Surface roughness plays an important role in microwave remote *** the agricultural domain,surface roughness is crucial for soil moisture retrieval methods that use electromagnetic surface scattering or microwave radiative transfer ***,improved characterization of Soil Surface Roughness(SSR)is of considerable *** this study,three approaches,including a standard pin profiler,a liDAR point cloud generated from an iPhone 12 Pro,and a Structure from Motion(SfM)photogrammetric point cloud,were applied over 24 surface profiles with different roughness variations to measure surface *** objective of this study was to evaluate the capability of smartphone-based liDAR technology to measure surface roughness parameters and compare the results of this technique with the more common *** showed that the iPhone liDAR technology,when point cloud data is captured in a fine-resolution mode,has a significant correlation with SfM photogrammetry(R2=0.70)and a relatively close agreement with pin profiler(R2=0.60).However,this accuracy tends to be greater for random surfaces and rough profiles with row structure *** results of this study confirm that smartphone-based liDAR can be used as a cost-effective,fast,and time-efficient alternative tool for measuring surface roughness,especially for rough,wide,and inaccessible areas.

This study develops a method to use deep learning models to predict the mineral assemblages and their relative abundances in paleolake cores using high-resolution XRF core scan elemental data and X-ray diffraction(XRD)mineralogical results from the same core taken at coarser *** uses the XRF core scan data along with published mineralogical information from the Olduvai Gorge Coring Project(OGCP)2014 sediment cores 1A,2A,and 3A from Paleolake Olduvai,*** regression and classification models were developed using a Keras deep learning framework to assess the predictability of mineral assemblages with their relative abundances(in regression models)or at least the mineral assemblages(in classification models)using XRF core scan *** were created using the Sequential class and Functional API with different model *** correlation matrix of element ratios calculated from XRF element intensity records from the cores and XRD-derived mineralogical information was used to select the most useful features to train the models.1057 training data records were used for the *** classes were also used for some models using Wide&Deep neural networks since those combine the benefits of memorization and generalization for mineral *** results were validated using 265 validation data records unseen by the model and discuss the accuracy of models using six test *** optimized Deep Neural Network(DNN)classification model achieved over 86%binary accuracy while the regression models were also able to predict the relative mineral abundances of samples with high ***,the study shows the efficacy of a carefully crafted Deep learning(Dl)model for predicting mineral assemblages and abundances using high-resolution XRF core scan data.

Hematopoietic stem cell-derived adipocytes and fibroblasts in the tumor microenvironment

The tumor microenvironment(TME) is complex and constantly evolving. This is due, in part, to the crosstalk between tumor cells and the multiple cell types that comprise the TME, which results in a heterogeneous population of tumor cells and TME cells. This review will focus on two stromal cell types, the cancerassociated adipocyte(CAA) and the cancer-associated fibroblast(CAF). In the clinic, the presence of CAAs and CAFs in the TME translates to poor prognosis in multiple tumor types. CAAs and CAFs have an activated phenotype and produce growth factors, inflammatory factors, cytokines, chemokines, extracellular matrix components, and proteases in an accelerated and aberrant fashion. Through this activated state, CAAs and CAFs remodel the TME, thereby driving all aspects of tumor progression, including tumor growth and survival, chemoresistance, tumor vascularization, tumor invasion, and tumor cell metastasis. Similarities in the tumorpromoting functions of CAAs and CAFs suggest that a multipronged therapeutic approach may be necessary to achieve maximal impact on disease. While CAAs and CAFs are thought to arise from tissues adjacent to the tumor, multiple alternative origins for CAAs and CAFs have recently been identified. Recent studies from our lab and others suggest that the hematopoietic stem cell, through the myeloid lineage, may serve as a progenitor for CAAs and CAFs. We hypothesize that the multiple origins of CAAs and CAFs may contribute to the heterogeneity seen in the TME. Thus, a better understanding of the origin of CAAs and CAFs, how this origin impacts their functions in the TME, and thetemporal participation of uniquely originating TME cells may lead to novel or improved anti-tumor therapeutics.
