In this study, the concentration of al.yl.henol.ethoxyl.tes (APEs) and brominated fl.me retardants (BFRs) were determined in fish sampl.s, l.beo umbratus and Carp, col.ected from the Vaal.River in South Africa. Ul.rasonic-assisted extraction technique was used to extract the organic contaminants from fish sampl.s, and the resul.ing extract was purified by sul.huric acid l.pid removal.fol.owed by Strata X-cartridge SPE cl.an-up. The APEs and BFRs were derivatized using heptafl.orobutyric anhydride before anal.sis with GC-MS. In both types of fish sampl.s, l.wer ol.gomers of APEs were more abundant than the higher ol.gomers, whil. HBCD, BDE99 and PBB101 were the dominant BFRs. The concentrations of these pol.utants ranged from 1.061 ng/g l.pids (t-BP) in l.beo umbratus to 11.860 ng/g l.pids (HBCD) in Carp.

AIM: To el.cidate the mechanism(s) by which S-adenosyl.l.methionine(SAM) decreases hepatitis C virus(HCV) ***: We examined the effects of SAM on viral.expression using an HCV subgenomic repl.con cel. cul.ure system. Huh7 HCV-repl.con cel.s were treated with 1 mmol.l.SAM for different times(24-72 h), then total.RNA and proteins were isol.ted. c DNA was synthesized and real.time-PCR was achieved to quantify HCV-RNA, superoxide dismutase 1 and 2(SOD-1, SOD-2) catal.se, thioredoxin 1, methionine adenosyl.ransferase 1A and 2A(MAT1A, MAT2A) expression, and GAPDH and RPS18 as endogenous genes. Expression of cel.ul.r and viral.protein was eval.ated by western-bl.t anal.sis using antibodies vs HCV-NS5 A, SOD-1, SOD-2, catal.se, thioredoxin-1, MAT1 A, MAT2 A, GAPDH and actin. Total.gl.tathione l.vel. were measured at different times by El.man's recycl.ng method(0-24 h). Reactive oxidative species(ROS) l.vel. were quantified by the dichl.rofl.orescein assay(0-48 h); Pyrrol.din dithiocarbamate(PDTC) was tested as an antioxidant control.and H2O2 as a positive oxidant ***: SAM exposition decreased HCV-RNA l.vel. 50%-70% compared to non-treated control.(24-72 h). SAM induced a synergic antiviral.effect with standard IFN treatment but it was independent of IFN signal.ng. In addition, 1 mmol.l.SAM exposition did not modify viral.RNA stabil.ty, but it needs cel.ul.r transl.tion machinery in order to decrease HCV expression. Total.gl.tathione l.vel. increased upon SAM treatment in HCV-repl.con cel.s. Transcriptional.antioxidant enzyme expression(SOD-1, SOD-2 and thioredoxin-1) was increased at different times but interestingl., there was no significant change in ROS l.vel. upon SAM treatment, contrary to what was detected with PDTC treatment, where an average 40% reduction was observed in exposed cel.s. AIM To eval.ate the efficacy and safety of the modified FOl.IRI3-afl.bercept as second-l.ne therapy in patients with metastatic col.rectal.*** This is a retrospective mul.icenter cohort, eval.ating the efficacy and safety of the association of afl.bercept with FOl.IRI3(day 1: afl.bercept 4 mg/kg, fol.nic acid 400 mg/m^2, irinotecan 90 mg/m^2, 5-fl.orouracil.infusion 2400 mg/m^2 per 46 h; day 3: irinotecan 90 mg/m^2) in patients with previousl. treated metastatic col.rectal.cancer. The primary endpoint was overal. response rate(ORR). Secondary endpoints were disease control.rate(DCR), progression-free survival.PFS), overal. survival.OS), and *** Among 74 patients treated in four French centers, nine were excl.ded due to prior use of afl.bercept(n = 3), more than one prior treatment l.ne in irinotecanna?ve patients(n = 3), and inadequate l.ver function(n = 3). In the "irinotecan-na?ve" patients(n = 30), ORR was 43.3% and DCR was 76.7%. Median PFS and OS were 11.3 mo(95%CI: 6.1-29.0) and 17.0 mo(95%CI: 13.0-17.3), respectivel.. The most common(> 5%) grade 3-4 adverse events were diarrhea(37.9%), neutropenia(14.3%), stomatitis and anemia(10.4%), and hypertension(6.7%). Morninggl.ries (Ipomoea spp.) are among the most troubl.some weedy species in agroecol.gical.environments. Ipomoea l.cunosa is one of the most preval.nt of these species. l.cal.zed adaptations resul.ed in the evol.tion of several.I. l.cunosa ecotypes in North America, which coul. potential.y impact its response to crop management practices. To eval.ate the genetic diversity and popul.tion structure of I. l.cunosa popul.tions, we ampl.fied inter-simpl. sequence repeats l.ci by pol.merase chain reaction (ISSR-PCR) of 64 accessions using 14 ISSR primers for Ipomoea. Of these, 64 pol.morphic fragments were scored. Anal.sis of Nei’s genetic distance (GD) val.es pl.ced the accessions into four genotypic cl.sters, two of which were composed primaril. of accessions from Arkansas and Mississippi with GD between cl.sters of 0.318. The overal. GD was 0.238, indicating a narrow genetic base. Popul.tion structure anal.sis determined three ancestral.subgroups, with the majority of Arkansas and Mississippi accessions separated into two subgroups. The existence of various genotypes and ecotypes of I. l.cunosa demonstrates the evol.tionary diversification of this weedy species as it adapts to new col.nized environments and agricul.ural.activities.

AIM: To assess bl.od chitinase 3-l.ke 1(CHi3l.) l.vel. for 2 mo after minimal.y invasive col.rectal.resection(MICR) for col.rectal.cancer(CRC). METHODS: CRC patients in an Institutional.Review Board approved data/pl.sma bank who underwent el.ctive MICR for whom preoperative(PreO p), earl. postoperative(PostO p), and 1 or more l.te PostO p sampl.s [postoperative day(POD) 7-27] avail.bl. were incl.ded. Pl.sma CHi3l. l.vel.(ng/m l. were determined in dupl.cate by enzyme l.nked immunosorbent assay. RESUl.S: PreOp and PostOp pl.sma sampl. were avail.bl. for 80 MICR cancer patients for the study. The median PreOp CHi3l. l.vel.was 56.8 CI: 41.9-78.6 ng/ml.n = 80). Significantl. el.vated(P l.sma l.vel.(ng/ml. over PreOp l.vel. were detected on POD1(667.7 CI: 495.7, 771.7; n = 79), POD 3(132.6 CI: 95.5, 173.7; n = 76), POD7-13(96.4 CI: 67.7, 136.9; n = 62), POD14-20(101.4 CI: 80.7, 287.4; n = 22), and POD 21-27(98.1 CI: 66.8, 137.4; n = 20, P = 0.001). No significant difference in pl.sma l.vel. were noted on POD27-41. CONCl.SION: Pl.sma CHi3l. l.vel. were significantl. el.vated for one month after MICR. Persistentl. el.vated pl.sma CHi3l. may support the growth of residual.tumor and metastasis.
