Diabetes and high-glucose could upregulate the expression of receptor for activated C kinase 1 in retina

BACKGROUND Diabetic retinopathy(DR)is a major ocular complication of diabetes mellitus,leading to visual impairment.Retinal pigment epithelium(RPE)injury is a key component of the outer blood retinal barrier,and its damage is an important indicator of DR.Receptor for activated C kinase 1(RACK1)activates protein kinase C-ε(PKC-ε)to promote the generation of reactive oxygen species(ROS)in RPE cells,leading to apoptosis.Therefore,we hypothesize that the activation of RACK1 under hypoxic/high-glucose conditions may promote RPE cell apoptosis by modulating PKC-ε/ROS,thereby disrupting the barrier effect of the outer blood retinal barrier and contributing to the progression of DR.AIM To investigate the role and associated underlying mechanisms of RACK1 in the development of early DR.METHODS In this study,Sprague-Dawley rats and adult RPE cell line-19(ARPE-19)cells were used as in vivo and in vitro models,respectively,to explore the role of RACK1 in mediating PKC-εin early DR.Furthermore,the impact of RACK1 on apoptosis and barrier function of RPE cells was also investigated in the former model.RESULTS Streptozotocin-induced diabetic rats showed increased apoptosis and upregulated expression of RACK1 and PKC-εproteins in RPE cells following a prolonged modeling.Similarly,ARPE-19 cells exposed to high glucose and hypoxia displayed elevated mRNA and protein levels of RACK1 and PKC-ε,accompanied by an increases in ROS production,apoptosis rate,and monolayer permeability.However,silencing RACK1 significantly downregulated the expression of PKC-εand ROS,reduced cell apoptosis and permeability,and protected barrier function.CONCLUSION RACK1 plays a significant role in the development of early DR and might serve as a potential therapeutic target for DR by regulating RPE apoptosis and barrier function.

Preparation of biomass-derived carbon loaded with MnO_(2) as lithium-ion battery anode for improving its reversible capacity and cycling performance

Biomass-derived carbon materials for lithiumion batteries emerge as one of the most promising anodes from sustainable perspective.However,improving the reversible capacity and cycling performance remains a long-standing challenge.By combining the benefits of K2CO_(3) activation and KMnO_(4) hydrothermal treatment,this work proposes a two-step activation method to load MnO_(2) charge transfer onto biomass-derived carbon(KAC@MnO_(2)).Comprehensive analysis reveals that KAC@MnO_(2) has a micro-mesoporous coexistence structure and uniform surface distribution of MnO_(2),thus providing an improved electrochemical performance.Specifically,KAC@MnO_(2) exhibits an initial chargedischarge capacity of 847.3/1813.2 mAh·g^(-1) at 0.2 A·g^(-1),which is significantly higher than that of direct pyrolysis carbon and K2CO_(3) activated carbon,respectively.Furthermore,the KAC@MnO_(2) maintains a reversible capacity of 652.6 mAh·g^(-1) after 100 cycles.Even at a high current density of 1.0 A·g^(-1),KAC@MnO_(2) still exhibits excellent long-term cycling stability and maintains a stable reversible capacity of 306.7 mAh·g^(-1) after 500 cycles.Compared with reported biochar anode materials,the KAC@MnO_(2) prepared in this work shows superior reversible capacity and cycling performance.Additionally,the Li+insertion and de-insertion mechanisms are verified by ex situ X-ray diffraction analysis during the chargedischarge process,helping us better understand the energy storage mechanism of KAC@MnO_(2).

A reweighted damped singular spectrum analysis method for robust seismic noise suppression

(Multichannel) Singular spectrum analysis is considered as one of the most effective methods for seismic incoherent noise suppression. It utilizes the low-rank feature of seismic signal and regards the noise suppression as a low-rank reconstruction problem. However, in some cases the seismic geophones receive some erratic disturbances and the amplitudes are dramatically larger than other receivers. The presence of this kind of noise, called erratic noise, makes singular spectrum analysis(SSA) reconstruction unstable and has undesirable effects on the final results. We robustify the low-rank reconstruction of seismic data by a reweighted damped SSA(RD-SSA) method. It incorporates the damped SSA, an improved version of SSA, into a reweighted framework. The damping operator is used to weaken the artificial disturbance introduced by the low-rank projection of both erratic and random noise. The central idea of the RD-SSA method is to iteratively approximate the observed data with the quadratic norm for the first iteration and the Tukeys bisquare norm for the rest iterations. The RD-SSA method can suppress seismic incoherent noise and keep the reconstruction process robust to the erratic disturbance. The feasibility of RD-SSA is validated via both synthetic and field data examples.

High-dimensional uncertainty quantification of projectile motion in the barrel of a truck-mounted howitzer based on probability density evolution method

This paper proposed an efficient research method for high-dimensional uncertainty quantification of projectile motion in the barrel of a truck-mounted howitzer.Firstly,the dynamic model of projectile motion is established considering the flexible deformation of the barrel and the interaction between the projectile and the barrel.Subsequently,the accuracy of the dynamic model is verified based on the external ballistic projectile attitude test platform.Furthermore,the probability density evolution method(PDEM)is developed to high-dimensional uncertainty quantification of projectile motion.The engineering example highlights the results of the proposed method are consistent with the results obtained by the Monte Carlo Simulation(MCS).Finally,the influence of parameter uncertainty on the projectile disturbance at muzzle under different working conditions is analyzed.The results show that the disturbance of the pitch angular,pitch angular velocity and pitch angular of velocity decreases with the increase of launching angle,and the random parameter ranges of both the projectile and coupling model have similar influence on the disturbance of projectile angular motion at muzzle.

Polysaccharide of Alocasia cucullata Exerts Antitumor Effect by Regulating Bcl-2, Caspase-3 and ERK1/2 Expressions during Long-Time Administration

Objective: To study the in vitro and in vivo antitumor effects of the polysaccharide of Alocasia cucullata(PAC) and the underlying mechanism. Methods: B16F10 and 4T1 cells were cultured with PAC of 40 μg/m L, and PAC was withdrawn after 40 days of administration. The cell viability was detected by cell counting kit-8. The expression of Bcl-2 and Caspase-3 proteins were detected by Western blot and the expressions of ERK1/2 mRNA were detected by quantitative real-time polymerase chain reaction(qRT-PCR). A mouse melanoma model was established to study the effect of PAC during long-time administration. Mice were divided into 3 treatment groups: control group treated with normal saline, positive control group(LNT group) treated with lentinan at 100 mg/(kg·d), and PAC group treated with PAC at 120 mg/(kg·d). The pathological changes of tumor tissues were observed by hematoxylin-eosin staining. The cell apoptosis of tumor tissues was detected by TUNEL staining. Bcl-2 and Caspase-3 protein expressions were detected by immunohistochemistry, and the expressions of ERK1/2, JNK1 and p38 mRNA were detected by qRT-PCR. Results: In vitro, no strong inhibitory effects of PAC were found in various tumor cells after 48 or 72 h of administration. Interestingly however, after 40 days of cultivation under PAC, an inhibitory effect on B16F10 cells was found. Correspondingly, the long-time administration of PAC led to downregulation of Bcl-2 protein(P <0.05), up-regulation of Caspase-3 protein(P<0.05) and ERK1 mRNA(P<0.05) in B16F10 cells. The above results were verified by in vivo experiments. In addition, viability of B16F10 cells under long-time administration culture in vitro decreased after drug withdrawal, and similar results were also observed in 4T1 cells. Conclusions: Long-time administration of PAC can significantly inhibit viability and promote apoptosis of tumor cells, and had obvious antitumor effect in tumor-bearing mice.

Engineering singlet and triplet excitons of TADF emitters by different host-guest interactions

Understanding the host-guest interactions for thermally activated delayed fluorescence(TADF)emitters is critical because the interactions between the host matrices and TADF emitters enable precise control on the optoelectronic performance,whereas technologically manipulating the singlet and triplet excitons by using different kinds of host-guest interactions remains elusive.Here,we report a comprehensive picture that rationalizes host-guest interaction-modulated exciton recombination by using time-resolved spectroscopy.We found that the early-time relaxation is accelerated in polar polymer because dipole-dipole interaction facilitates the stabilization of the 1CT state.However,an opposite trend is observed in longer delay time,and faster decay in the less polar polymer is ascribed to theπ-πinteraction that plays the dominant role in the later stage of the excited state.Our findings highlight the technological engineering singlet and triplet excitons using different kinds of host-guest interactions based on their electronic characteristics.

Production logistics(PL)is considered as a critical factor that affects the efficiency and cost of production operations in discrete manufacturing systems.To effectively utilize manufacturing big data to improve PL efficiency and promote job shop floor economic benefits,this study proposes a PL trajectory analysis and optimization decision making method driven by a manufacturing task data chain(MTDC).First,the manufacturing task chain(MTC)is defined to characterize the discrete production process of a product.To handle manufacturing big data,the MTC data paradigm is designed,and the MTDC is established.Then,the logistics trajectory model is presented,where the various types of logistics trajectories are extracted using the MTC as the search engine for the MTDC.Based on this,a logistics efficiency evaluation indicator system is proposed to support the optimization decision making for the PL.Finally,a case study is applied to verify the proposed method,and the method determines the PL optimization decisions for PL efficiency without changing the layout and workshop equipment,which can assist managers in implementing the optimization decisions.

The Paleogene is a crucial period when terrestrial and marine ecosystems recovered from major disruptions and gradually approached their modern states.In the Qinghai-Tibetan Plateau and its surrounding regions,the Paleogene also represents a significant phase of tectonic evolution in the Qinghai-Tibetan Plateau-Himalaya orogeny,reorganization of Asian climates,and evolution of biodiversity.Due to limitations in research conditions and understanding,there are still many controversies regarding stratigraphic divisions in the Qinghai-Tibetan Plateau and its surrounding regions.In recent years,extensive studies on sedimentary petrology,magnetostratigraphy,and isotope dating have been conducted in the region.Numerous fossils have been discovered and reported,contributing to a more systematic understanding of biostratigraphy.These studies have laid a solid foundation for the comprehensive investigation of the stratigraphy,biotas and paleogeographic evolution of the Qinghai-Tibetan Plateau and its surrounding regions during the Paleogene.In this paper,we integrate recent research on fossils,isotopic dating,magnetostratigraphy,and geochemistry to refine the stratigraphic divisions and correlation framework of different tectonic units in the region,building upon previous studies.Since the Second Tibetan Plateau Scientific Expedition and Research,the knowledge of Paleogene floras has gradually expanded.This paper discusses the biostratigraphic significance of extinct and newly appeared taxa based on the latest dating results of these plant species.The new understanding of fossil species such as the“Eucalyptus”and Arecaceae establishes connections between the Paleogene flora of the Qinghai-Tibetan region and the biotas of Gondwana,specifically Oceania and South America.The evolutionary history of key taxa near the Yarlung Zangbo suture zone indicates that the collision between the Indian and Eurasian plates occurred approximately 65-54 Ma.Paleoelevation reconstructions,based on plant fossils,suggest that the Hengduan Mounta

Evaluation of suitability,adaptability,and reserve potential of construction land on the Qinghai-Tibet Plateau

Construction land is the leading carrier of human activities such as production and living.Evaluating the construction land suitability(CLS)on the Qinghai-Tibet Plateau(QTP)holds significant implications for harmonizing the relationship between ecological protection and human activity and promoting population and industry layout optimization.However,no relevant studies provide a complete CLS assessment of the QTP.In this study,we developed a model-based CLS evaluation framework coupling of pattern and process to calculate the global CLS on the QTP based on a previously developed CLS evaluation model.Then,using the land-use data of 1990,2000,2010,and 2020,we examined the adaptability of existing construction land(ECL)to the CLS assessment result through the adaptability index and vertical gradient index and further analyzed the limitations of maladaptive construction land.Finally,we calculated the potential area of reserve suitable construction land.This article includes four conclusions:(1)The highly suitable,suitable,moderately suitable,marginally suitable,and unsuitable CLS classes cover areas of 0.33×10^(4)km^(2),10.42×10^(4)km^(2),18.06×10^(4)km^(2),24.12×10^(4)km^(2),and 205.29×10^(4)km^(2),respectively.Only approximately 11%of the study area on the QTP is suitable for large-scale permanent construction land,and approximately 79.50%of the area is unsuitable under current economic and technological conditions.(2)The ECL adaptability index is 85.16%,85.93%,85.18%,and 78.01%during 1990–2020,respectively,with an average adaptability index exceeding 80%on the QTP.The ECL distribution generally conforms to construction land suitable space characteristics but with a significant spatial difference.(3)From 1990 to 2020,the maladaptive ECL was dominated by rural settlement land,transport land,and special land,with a rapidly increasing proportion of urban and other construction land.The maladaptive ECL is constrained by both elevation and slope in the southern Qinghai Plateau,the Hengduan Mountains,and the Qilian Mount

The Effect of Lateral Offset Distance on the Aerodynamics and Fuel Economy of Vehicle Queues

The vehicle industry is always in search of breakthrough energy-saving and emission-reduction technologies.In recent years,vehicle intelligence has progressed considerably,and researchers are currently trying to take advantage of these developments.Here we consider the case of many vehicles forming a queue,i.e.,vehicles traveling at a predetermined speed and distance apart.While the majority of existing studies on this subject have focused on the influence of the longitudinal vehicle spacing,vehicle speed,and the number of vehicles on aerodynamic drag and fuel economy,this study considers the lateral offset distance of the vehicle queue.The group fuel consumption savings rate is calculated and analyzed.As also demonstrated by experimental results,some aerodynamic benefits exist.Moreover,the fuel consumption saving rate of the vehicle queue decreases as the lateral offset distance increases.
